Successful retreatment with glecaprevir/pibrentasvir after sofosbuvir/velpatasvir failure in a patient with hepatitis C-related decompensated cirrhosis complicated by hepatocellular carcinoma.

Direct-acting antivirals (DAAs) markedly improve sustained virologic response (SVR) rates in chronic hepatitis C virus (HCV) infection, including decompensated cirrhosis. However, retreatment options remain limited in patients with decompensated cirrhosis after DAA failure. We experienced a case of decompensated cirrhosis that was successfully retreated with glecaprevir/pibrentasvir (GLE/PIB) after sofosbuvir/velpatasvir (SOF/VEL) failure. The patient was a woman in her 70 s with HCV genotype 1b and an HCV RNA level of 7.1 log IU/mL. The Child-Pugh classification was class B, with a score of 7

Identification of a novel conserved B-cell epitope in the glycoprotein of rabies virus.

Rabies virus (RABV), a single-stranded RNA virus, invades the central nervous system and causes acute, zoonotic diseases worldwide. The glycoprotein (G) of RABV is the only surface antigen responsible for inducing potent neutralizing antibodies, making it a key target for medical countermeasures. In this study, two anti-G protein monoclonal antibodies (mAbs) were prepared using hybridoma cell technology, with their high specificity and robust reactivity validated by indirect enzyme-linked immunosorbent assay (i-ELISA), indirect immunofluorescence assay (IFA) and Western blot. Through systemati

The CCAT2 enigma: pioneering insights into colorectal cancer pathophysiology and therapeutic innovation.

Colorectal cancer (CRC) remains a major global health burden and a leading cause of cancer-related morbidity and mortality. It ranks as the third most commonly diagnosed malignancy and the second leading cause of cancer-related deaths worldwide. These statistics underscore the urgent need for improved diagnostic, prognostic, and therapeutic strategies to combat the disease. The development of CRC is driven by a combination of genetic predisposition, environmental exposures, and lifestyle-related factors. Major risk factors include a family history of CRC, unhealthy diet, smoking, excessive alc

Red Blood Cell Membrane Vesicles for siRNA Delivery: A Biocompatible Carrier With Passive Tumor Targeting and Prolonged Plasma Residency.

BACKGROUND: Despite many advances in gene therapy, the delivery of small interfering RNAs is still challenging. Erythrocytes are the most abundant cells in the human body, and their membrane possesses unique features. From them, erythrocytes membrane vesicles can be generated, employable as nano drug delivery system with prolonged blood residence and high biocompatibility. METHODS: Human erythrocyte ghosts were extruded in the presence of siRNA, and the objects were termed EMVs (erythrocyte membrane vesicles). An ultracentrifugation-based method was applied to select only the densest EMVs, ie

TREM2 facilitates gastric cancer progression and immune evasion via inhibiting TRIM21-mediated STAT1 degradation in tumor-associated macrophages.

Tumor-associated macrophages (TAMs) play a crucial role in fostering an immunosuppressive tumor microenvironment, promoting cancer progression, contributing to immune evasion and resistance to immunotherapy. However, the mechanisms by which TAMs exert these effects in gastric cancer (GC) remain unclear. Human TAMs were isolated from GC patients through magnetic sorting for RNA sequencing. THP-1 cell line and mice bone marrow-derived macrophages (BMDMs) induced TAMs were applied for functional assays. Two in-house tumor microarrays were utilized for validation: the Zhongshan Cohort, comprising

XPO1/Exportin-1 in Acute Myelogenous Leukemia; Biology and Therapeutic Targeting.

Exportin 1 is responsible for the export of hundreds of proteins, several RNA species and ribosomal components from the nucleus to the cytoplasm. Several transported proteins are important for regulation of cell proliferation and survival both in normal and malignant cells. We review the biological importance and the possibility of therapeutic targeting of Exportin 1 in acute myeloid leukemia (AML). Exportin 1 levels can be increased in human primary AML cells, and even exportin inhibition as monotherapy seems to have an antileukemic effect. The results from Phase I/II studies also suggest tha

Alzheimer Disease: The proposed role of tanycytes in the formation of tau tangles and amyloid beta plaques in human brain.

Currently, the causes for Alzheimer Disease (AD) are thought to lie in the formation of abnormal protein deposits including tau tangles and Amyloid ß (Aβ) plaques in the human cortex. These proteins are believed to accumulate in the brain due to impaired waste removal resulting in neurodegeneration. In an alternative hypothesis we have recently proposed the existence of an aquaporin4 aqua channel (AQP4)-expressing tanycyte-derived canal network that likely internalizes waste for removal from the brain. We propose that both Aβ and tau protein may play important structural roles in this canal sy

Form and function of actin impacts actin health and aging.

The actin cytoskeleton is a fundamental and highly conserved structure that functions in diverse cellular processes, yet its direct contribution to organismal aging remains unclear. Here, we systematically interrogated how genetic and pharmacologic perturbations of actin structure and function influence lifespan and various hallmarks of aging in Caenorhabditis elegans. Whole-animal and tissue-specific knockdown of actin and key actin-binding proteins (ABPs) - arx-2 (Arp2/3), unc-60 (cofilin), and lev-11 (tropomyosin) - led to premature disruption of filament organization, reduced lifespan, and

Epigenetic silencing of MSTN via m6A modification underlies the renoprotective effects of engineered MSC exosomes with RBM15 depletion in diabetic nephropathy.

Diabetic nephropathy (DN) is a leading complication of diabetes mellitus. Engineered exosomes have shown promise in disease treatment by enabling targeted cargo delivery. RNA-binding motif protein 15 (RBM15) accelerates the progression of DN. This study developed siRBM15-loaded exosomes and evaluated their therapeutic efficacy and mechanisms. Exosomes were isolated from human umbilical cord MSCs (hucMSCs) and loaded with siRBM15 (siRBM15-Exo) by electroporation. Human glomerular mesangial cells (HGMCs) were exposed to high glucose (HG). A rat model of DN was generated by streptozotocin inducti

WTAP Silencing protects human aortic smooth muscle cells from angiotensin II-induced senescence, apoptosis, ferroptosis, and inflammation by regulating PCSK9.

Abdominal aortic aneurysm (AAA) is a life-threatening vascular disease characterized by aortic wall degeneration and inflammation. The molecular mechanisms underlying AAA development remain unclear. Wilms tumor 1-associated protein (WTAP) has been implicated in various biological processes, but its role in AAA pathogenesis, particularly in cardiomyocyte regulation, has not been fully explored. Quantitative real-time PCR (qRT-PCR) was performed to detect the mRNA levels of WTAP and proprotein convertase subtilisin/kexin type 9 (PCSK9). Western blotting assay was used to analyze protein expressi

ALKBH5 reverses romidepsin-mediated anti-multiple myeloma activity via regulation of m6A modification of FOXM1.

Multiple myeloma (MM), a prevalent hematological malignancy, is characterized by the abnormal proliferation and accumulation of plasma cells within the bone marrow. ALKBH5, an RNA demethylase, is implicated in various tumor-related processes, yet its role in myeloma remains insufficiently understood. In this study, we revealed that elevated ALKBH5 expression correlates with poor MM prognosis. Furthermore, romidepsin, a recognized histone deacetylase (HDAC) inhibitor, was found to significantly suppress both the protein levels and enzymatic activity of ALKBH5. Interestingly, while ectopic expre

MicroRNA Signatures in Cardiometabolic Disorders as a Next-Generation Diagnostic Approach: Current Insight.

Cardiometabolic diseases, including cardiovascular disorders and type 2 diabetes mellitus, are the leading cause of morbidity and mortality worldwide, placing a significant burden on healthcare systems. Although advances in imaging and risk stratification have improved disease management, conventional diagnostic and prognostic tools often lack the requisite sensitivity and specificity for early and precise risk stratification. This limitation stems from their poor ability to capture the full molecular complexity of these conditions, underscoring an urgent need for innovative biomarkers to brid

HGCJAMH: A Method for circRNA-Drug Sensitivity Prediction Based on Higher-Order Moment-Guided Model and Hypergraph Jumping Learning Mechanism.

Circular RNAs (circRNAs) play a key role in regulating cellular drug sensitivity, and they hold significant promise as potential biomarkers in disease treatment and precision medicine. However, existing circRNA-drug sensitivity association (CDSA) prediction methods generally face the following challenges: high dependence on wet experiments, sparse association data leading to a high percentage of false negatives, insufficient expression of embedded features, and inadequate modeling of higher-order heterogeneous relationships. To solve the above problems, this paper proposes a prediction model,

Cellular responses in acetabular labral reconstruction: effects of strain level on labral, tibialis tendon, and meniscal cells in vitro.

While the clinical outcomes of acetabular labral repair and reconstruction have been reported, comparative cellular responses between common allografts to clinically relevant load are less clear. This study aims to compare acetabular labrum (LAB), anterior tibialis tendon (TEN), and knee meniscus (MEN) cellular responses to biaxial tensile strain during in vitro culture. LAB, TEN, and MEN tissues were recovered from skeletally mature research hounds (n = 5). Primary LAB, TEN, and MEN fibroblast cell cultures were established. Using a bioreactor, cells were loaded at 0%, 4%, or 10% biaxial tens

N-glycosylation stabilized TNFAIP6 promotes ovarian cancer metastasis by activating the PI3K-AKT signaling pathway.

BACKGROUND: Ovarian cancer (OC) represents the most lethal gynecological malignancy, with a dismal 5-year survival rate of ∼30 % in advanced-stage patients, largely due to metastatic dissemination and limited therapeutic options. Elucidating the molecular drivers of OC metastasis remains imperative for developing targeted interventions. METHODS: Integrative multi-omics analyses were performed using bulk RNA-seq and scRNA-seq data from TCGA and GEO repositories. Protein dynamics were validated via Western blot and immunofluorescence. Prognostic significance was assessed through Kaplan-Meier su

Metabolic rewiring during bone development underlies tRNA m7G-associated primordial dwarfism.

Translation of mRNA to protein is tightly regulated by transfer RNAs (tRNAs), which are subject to various chemical modifications that maintain structure, stability, and function. Deficiency of tRNA N7-methylguanosine (m7G) modification in patients causes a type of primordial dwarfism, but the underlying mechanism remains unknown. Here we report that the loss of m7G rewires cellular metabolism, leading to the pathogenesis of primordial dwarfism. Conditional deletion of the catalytic enzyme Mettl1 or missense mutation of the scaffold protein Wdr4 severely impaired endochondral bone formation an

Hypoxia exposure blunts angiogenic signaling and upregulates the antioxidant system in endothelial cells derived from elephant seals.

BACKGROUND: Elephant seals exhibit extreme hypoxemic tolerance derived from repetitive hypoxia/reoxygenation episodes they experience during diving bouts. Real-time assessment of the molecular changes underlying protection against hypoxic injury in seals remains restricted by their at-sea inaccessibility. Hence, we developed a proliferative arterial endothelial cell culture model from elephant seals and used RNA-seq, functional assays, and confocal microscopy to assess the molecular response to prolonged hypoxia. RESULTS: Seal and human endothelial cells exposed to 1% O2 for up to 6 h respond

A New Rat Model of Sacral Cord Injury Producing a Neurogenic Bladder and Its Functional and Mechanistic Studies.

Sacral spinal cord injury (SSCI) can disrupt bladder neuromodulation and impair detrusor function. Current studies provide limited information on the histologic and genetic changes associated with SSCI-related neurogenic lower urinary tract dysfunction (NLUTD), resulting in few treatment options. This study aimed to establish a simple animal model of SSCI to better understand the disease progression. Ninety 8-week-old Sprague-Dawley (SD) rats were randomly separated into sham operation and SSCI groups. The SSCI group underwent sacral spinal cord injury, while the sham group did not. Urodynamic
