Charcot-Marie-Tooth disease and intracellular traffic.

Mutations of genes whose primary function is the regulation of membrane traffic are increasingly being identified as the underlying causes of various important human disorders. Intriguingly, mutations in ubiquitously expressed membrane traffic genes often lead to cell type- or organ-specific disorders. This is particularly true for neuronal diseases, identifying the nervous system as the most sensitive tissue to alterations of membrane traffic. Charcot-Marie-Tooth (CMT) disease is one of the most common inherited peripheral neuropathies. It is also known as hereditary motor and sensory neuropa

Tripod pinch strength and thumb opposition are the major determinants of manual dexterity in Charcot-Marie-Tooth disease type 1A.

BACKGROUND: Clinical features of Charcot-Marie-Tooth disease type 1A (CMT1A) include slowly progressive distal muscle weakness, atrophy and sensory loss. Upper-limb involvement results in reduced manual dexterity interfering with the execution of daily activities. OBJECTIVE: To identify which hand function impairments are determinants of manual dexterity in CMT1A. METHODS: In a cross-sectional, observational study, hand function (strength, mobility and sensory function) and manual dexterity (Sollerman hand function test) were evaluated in adults with CMT1A. Multiple linear regression analysi

Overeruption of periodontally affected unopposed molars in adult rats.

BACKGROUND AND OBJECTIVE: In clinical practice, anterior teeth with periodontal disease exhibiting signs of overeruption are occasionally encountered. However, the influence of periodontitis on unopposed teeth needs to be further elucidated. This study investigated, in rats, the overeruption pattern of unopposed mandibular molars with experimentally induced periodontitis. MATERIAL AND METHODS: Sixty adult male rats were divided equally into four groups. In two groups, periodontitis was induced by a silk thread placed around the cervix of the right mandibular molar. In two groups with and with

A hyperactive neutrophil phenotype in patients with refractory periodontitis.

BACKGROUND: Neutrophils (PMNs) are critical components of the innate immune system and help to maintain oral health in the face of a constant bacterial challenge. However, along with protecting the periodontium from microbial invasion, these cells release potent lysosomal enzymes and oxygen radicals that can be destructive to periodontal tissues and lead to tooth loss. We examined neutrophil function in a unique population of patients diagnosed with refractory aggressive periodontitis (RAP). METHODS: Venous blood was obtained from 12 non-smoking patients who had been diagnosed with RAP, 10 pa…

The prevalence of inflammatory periodontitis is negatively associated with serum antioxidant concentrations.

Chronic periodontitis is an inflammatory disease that affects the supporting tissues of the teeth. It is initiated by specific bacteria within the plaque biofilm and progresses due to an abnormal inflammatory-immune response to those bacteria. Periodontitis is the major cause of tooth loss and is also significantly associated with an increased risk of stroke, type-2 diabetes and atheromatous heart disease. Oxidative stress is reported in periodontitis both locally and peripherally (serum), providing potential mechanistic links between periodontitis and systemic inflammatory diseases. It is the…

Intragenic SNP haplotypes associated with 84dup18 mutation in TNFRSF11A in four FEO pedigrees suggest three independent origins for this mutation.

Familial expansile osteolysis (FEO) is a rare disorder causing bone dysplasia. The clinical features of FEO include early-onset hearing loss, tooth destruction, and progressive lytic expansion within limb bones causing pain, fracture, and deformity. An 18-bp duplication in the first exon of the TNFRSF11A gene encoding RANK has been previously identified in four FEO pedigrees. Despite having the identical mutation, phenotypic variations among affected individuals of the same and different pedigrees were noted. Another 18-bp duplication, one base proximal to the duplication previously reported,

A retrospective study of periodontal disease severity in smokers and non-smokers.

BACKGROUND: Smoking has been associated with increased risk of periodontitis. The aim of the present study was to compare the periodontal disease severity of adult heavy smokers and never-smokers referred for assessment and treatment of chronic periodontitis. METHODS: A random sample of patients with at least 20 teeth, stratified for smoking and age (5-year blocks, 35 to 55 years), was selected from an original referral population of 1,221 subjects with chronic adult periodontitis. Adequate records for 59 never-smokers and 44 subjects who smoked at least 20 cigarettes per day were retrieved.

Quantitative measures of aggressive periodontitis show substantial heritability and consistency with traditional diagnoses.

BACKGROUND: Aggressive periodontitis (AgP) research nearly always classifies subjects into traditional discrete categories of localized or generalized, based upon degree of attachment loss (AL) and types of affected teeth. Since AL is continuous and quantitative, however, useful information is lost. We developed quantitative measures of AgP, compared these to traditional methods, and estimated heritabilities in families. METHODS: We examined 237 healthy, 169 localized AgP, and 204 generalized AgP subjects. We used the site of maximum AL of each tooth to calculate means for each subject for di

Retropharyngeal abscess: a clinical experience at the University College Hospital Ibadan.

BACKGROUND: Infection within the retropharyngeal space could progress on to an abscess formation resulting into retropharyngeal abscess (RPA), which can either be acute or chronic. RPA can be a life-threatening emergency, with potential for airway compromise and other catastrophic complications. This review is aimed at highlighting our experience with patients with a diagnosis of retropharyngeal abscess. METHODS: Retrospective review of thirty patients with a confirmed diagnosis of retropharyngeal abscess, who were admitted and managed within an eleven-year period (1993 to 2003) in the Otolar

Pathological findings in the x-linked form of Charcot-Marie-Tooth disease: a morphometric and ultrastructural analysis.

Mutations in the connexin 32 gene (Cx 32) are associated with the x-linked form of Charcot-Marie-Tooth disease (CMTX) and segregate with a CMT 1 phenotype. The gap junction protein Cx 32 is expressed in myelinating Schwann cells and has been localized to regions of non-compacted cytoplasm in paranodes and in Schmidt-Lanterman incisures. Mutant Cx 32 myelin proteins are predicted to impair Schwann cell functions. We have studied the resulting pathology in motor and sensory nerves from the probands of 13 CMTX kindreds with precisely defined genotype. This report provides a detailed descriptive a

Ocular and oculomotor signs in Joubert syndrome.

A number of oculomotor defects have been described in Joubert syndrome. This study systematically examined the oculomotor systems of 13 individuals previously diagnosed with Joubert syndrome. Twelve had the characteristic "molar tooth sign" seen on magnetic resonance imaging scan. In all individuals, smooth pursuit gain and vestibulo-ocular reflex cancellation were decreased in the horizontal and vertical directions and volitional saccades, when generated, were hypometric. We believe that these defects arise from a disorder in the posterior cerebellar vermis. All individuals also had partial t

Oral Health in Early and Advanced Stages (1-4) of Chronic Kidney Disease: A Systematic Review and Meta-Analysis.

It is essential to identify oral health changes in the early stages of chronic kidney disease (CKD) to prevent oral degeneration. The effects of early-stage CKD on the periodontium and the manifestation of associated oral symptoms remain poorly understood. Periodontitis is well documented in late-stage CKD. These changes represent a considerable burden for patients, leading to consequences like tooth loss and stomatognathic disorders. This study evaluates oral health parameters in patients with early- and advanced-stage CKD (stages 1-4), to identify potential changes in oral health and provide…
