The Cu,Zn superoxide dismutases from bovine liver, yeast, Caulobacter crescentus, and Photobacter leiognathi were compared for their susceptibilities to inhibition by cyanide and to inactivation by hydrogen peroxide and phenylglyoxal. All of these enzymes were affected by these reagents, albeit with some differences in sensitivity. The yeast and the bacterial enzymes were thus more sensitive to cyanide than was the bovine enzyme, while the bovine and the yeast enzymes were inactivated more rapidly by hydrogen peroxide and less rapidly by phenylglyoxal than were their bacterial counterparts. Pseudomonas sp. AU10 is an Antarctic psychrotolerant bacterium that produces a dye-decolorizing peroxidase (DyP-AU10). The recombinant enzyme (rDyP-AU10) is a heme-peroxidase that decolors dyes and modifies kraft lignin. In this work, we report the best activity parameters for lignin modification (at 45 °C and pH 4) and show that the enzyme increases the number of aldehydes, ketones, and phenolic compounds. The analyses of the HPLC profile of samples also support that rDyP-AU10 induces the chemical change of kraft lignin. The enzyme also acts as a biobleaching agent on cellulose pulps, as show

The inhibitory effect of pyridoxal phosphate on the Triton X-100 solubilized purified bovine heart succinate-ubiquinone reductase (Choudhry, Z.M., Gavrikova, E.V., Kotlyar, A.B., Tushurashvilli, P.R. and Vinogradov, A.D. (1985) FEBS Lett. 182, 171-175) was studied. The kinetics of the enzyme inactivation by pyridoxal phosphate was found to be strongly dependent both qualitatively and quantitatively on the concentration of the protein-detergent complexes. In the diluted system the inactivation of the ubiquinone-depleted enzyme was completely prevented by the saturating concentrations of Q2, car

Renin is produced from an inactive precursor, prorenin, through endoproteolytic cleavage at paired basic amino-acid residues. Using (35S)methionine-labeled prorenin, that was synthesized with Xenopus oocyte expression system, as a substrate, we have determined the tissue distribution and the nature of prorenin-converting activity in mouse. The highest activity was found in the submandibular gland of male ICR mouse. The activity of the enzyme seemed to be parallel to that of renin. This enzyme activity, with an optimal pH 8.0-8.5, was inhibited by leupeptin, antipain and benzamidine.

Improving enzyme catalytic activity is a key challenge in biocatalysis. To this end, the present study explored a simple and versatile strategy to enhance the catalytic activity of laccase (LAC). LAC was pretreated with ultrasound (US) for 20 min, and the activity of the pretreated LAC (designated as LACUS) was increased by 2.6-fold compared with that of pristine LAC. This increase in activity was attributed to the partial depolymerization of the spatial structure of LAC during ultrasound pretreatment, leading to a shift from the α-helix structure to an irregularly coiled structure. This struc

Activity of alkaline phosphatase in the major salivary glands of male and female mice at various ages of postnatal life, and in females during lactation was studied histochemically. Enzyme activity was not detected on the day of birth, but was found in the terminal tubules of all major salivary glands during the first postnatal week. Alkaline phosphatase activity was increasing gradually with age and a definitive enzymatic pattern was observed by the age of 6 weeks. No difference in enzyme activity was found among the major salivary glands of young adult and old animals. The parenchyma of full

The formation of aqueous humour by the ciliary body is a complex process. Active transport of solutes by the ciliary process epithelium is an energy-dependent mechanism that selectively transports substances against an electrochemical gradient across the cell membranes. Water passively follows the active solute transport. In addition to these active transport processes, ultrafiltration contributes to the formation of aqueous humour. The ciliary epithelium contains enzyme systems that function in the production of aqueous humour. The enzymes sodium-potassium-activated adenosine triphosphatase [

1. An homogenate of liver biopsy taken from a patient with Pompe's disease (type II glycogen-storage disease) was analysed by enzyme microassay and subcellular fractionation by sucrose-density-gradient centrifugation. 2. A strikingly low activity of lysosomal acid alpha-D-glucosidase was noted but there was also increased activity of neutral alpha-D-glucosidase (found in endoplasmic reticulum). 3. Activities of the acid hydrolases, acid phosphatase and beta-N-acetyl-D-glucosaminidase, were elevated compared with those of controls. Measurement of latent beta-N-acetyl-D-glucosaminidase activity

Ornithine carbamyl transferase activity was determined by estimation of the citrulline formed during the reaction. Citrulline is estimated by diacetylmonoxime in the presence of thiosemicarbazide. The conditions of enzyme analysis were then studied in buffer veronal-acetate medium at 37 degrees C. The optimum pH for activity depended on the ornithine concentration, but was independent of carbamyl-phosphate concentration. At pH 7.8, ornithine at concentrations higher than 1.6 mM inhibited enzyme activity, ornithine Km was 0.208 mM and that of carbamyl-phosphate was 1.92 mM. The incubation time

1. Caffeine (CA) is metabolized extensively and at least 17 metabolites arising from primary and secondary biotransformation pathways are found in urine following CA ingestion. The enzymes responsible for the formation of most of the metabolites derived from CA have been identified. 2. Given the near ubiquitous consumption of CA, this compound potentially constitutes a useful substrate probe for assessment of certain xenobiotic metabolizing enzyme activities in vivo. Indeed, various ratios of CA metabolites excreted in urine (urinary metabolic ratios; MRs) are now utilized widely for the popul

Precise localization of xanthine oxidase activity might elucidate physiological functions of the enzyme, which have not been established so far. Because xanthine oxidase is sensitive to chemical (aldehyde) fixation, we have localized its activity in unfixed cryostat sections of rat duodenum, oesophagus and tongue mounted on a semipermeable membrane. Previous studies had shown that this procedure enables the exact localization of activities of peroxisomal oxidases with maintenance of acceptable ultrastructure. Moreover, leakage and/or diffusion of enzyme molecules was prevented with this method

In Gaucher's disease (GD) there is an imbalance in the monocyte-macrophage system between the rate of formation and degradation of glucosylceramide due to low activity of the lysosomal enzyme betaglucorebrosidase. Therapeutic approaches are aimed to reduce the synthesis of glucosylceramide by inhibiting ceramide-glucosyltransferase or increasing the degradation of glucosylceramide using a recombinant beta-glucorebrosidase or recovering the residual activity of mutant enzymes with pharmacological chaperones. Inhibitory molecules of ceramide-glucosyltransferase used mimic glucose or ceramide. Al

Thiamin, or vitamin B1, is crucial for brain function. In its active form, thiamin pyrophosphate (TPP), it is a co-enzyme for several enzymes, including transketolase. Transketolase is an important enzyme in the non-oxidative branch of the pentose phosphate pathway (PPP), a pathway responsible for generating reducing equivalents, which is essential for energy transduction and for generating ribose for nucleic acid synthesis. Transketolase also links the PPP to glycolysis, allowing a cell to adapt to a variety of energy needs, depending on its environment. Abnormal transketolase expression and/

Lysosomal storage disorders are rare, inherited diseases caused by a deficiency of a specific, lysosomal enzyme. In the case of mucopolysaccharidosis type IIIA, a lack of active sulfamidase enzyme results in heparan sulfate accumulation, severe and progressive neurological deficits, and usually premature death. Embryonic stem cells can be genetically modified to overexpress lysosomal enzymes, providing a renewable reservoir of cells that can be readily expanded in culture. Screening clonal lines of embryonic stem cells for desirable properties such as high levels and maintenance of enzyme acti

BACKGROUND: The mucopolysaccharidoses (MPSs) are rare genetic disorders caused by mutations in lysosomal enzymes involved in the degradation of glycosaminoglycans (GAGs). In this study, we analyzed a total of 48 patients including MPSI (n=6), MPSII (n=18), MPSIIIA (n=11), MPSIVA (n=3), and MPSVI (n=10).
METHODS: In MPS patients, urinary GAGs were colorimetrically assayed. Enzyme activity was quantified by colorimetric and fluorimetric assays. To find mutations, all IDUA, IDS, SGSH, GALNS, and ARSB exons and intronic flanks were sequenced. New mutations were functionally assessed by reconstruc
