PCP and Wnt pathway components act in parallel during zebrafish mechanosensory hair cell orientation.

Planar cell polarity (PCP) plays crucial roles in developmental processes such as gastrulation, neural tube closure and hearing. Wnt pathway mutants are often classified as PCP mutants due to similarities between their phenotypes. Here, we show that in the zebrafish lateral line, disruptions of the PCP and Wnt pathways have differential effects on hair cell orientations. While mutations in the PCP genes vangl2 and scrib cause random orientations of hair cells, mutations in wnt11f1, gpc4 and fzd7a/b induce hair cells to adopt a concentric pattern. This concentric pattern is not caused by defect

Neuroplasticity in cerebral visual impairment (CVI): Assessing functional vision and the neurophysiological correlates of dorsal stream dysfunction.

Cerebral visual impairment (CVI) results from perinatal injury to visual processing structures and pathways and is the most common individual cause of pediatric visual impairment and blindness in developed countries. While there is mounting evidence demonstrating extensive neuroplastic reorganization in early onset, profound ocular blindness, how the brain reorganizes in the setting of congenital damage to cerebral (i.e. retro-geniculate) visual pathways remains comparatively poorly understood. Individuals with CVI exhibit a wide range of visual deficits and, in particular, present with impair

The role of alpha7 nicotinic acetylcholine receptors in inflammatory bowel disease: involvement of different cellular pathways.

Autonomic imbalance plays a pivotal role in the pathophysiology of inflammatory bowel diseases (IBD). The central nervous system (CNS) cooperates dynamically with the immune system to regulate inflammation through humoral and neural pathways. In particular, acetylcholine (Ach), the main neurotransmitter in the vagus nerve, decreases the production of pro-inflammatory cytokines through a mechanism dependent on the α7 nicotinic Ach receptors (α7nAChRs). Areas covered: Here, we review the evidence for involvement of the cholinergic anti-inflammatory pathway (CAP) in IBD. We also elaborate the role…

De novo mutations in PLXND1 and REV3L cause Möbius syndrome.

Möbius syndrome (MBS) is a neurological disorder that is characterized by paralysis of the facial nerves and variable other congenital anomalies. The aetiology of this syndrome has been enigmatic since the initial descriptions by von Graefe in 1880 and by Möbius in 1888, and it has been debated for decades whether MBS has a genetic or a non-genetic aetiology. Here, we report de novo mutations affecting two genes, PLXND1 and REV3L in MBS patients. PLXND1 and REV3L represent totally unrelated pathways involved in hindbrain development: neural migration and DNA translesion synthesis, essential fo

Generation of neural progenitor cells by chemical cocktails and hypoxia.

Neural progenitor cells (NPCs) can be induced from somatic cells by defined factors. Here we report that NPCs can be generated from mouse embryonic fibroblasts by a chemical cocktail, namely VCR (V, VPA, an inhibitor of HDACs; C, CHIR99021, an inhibitor of GSK-3 kinases and R, Repsox, an inhibitor of TGF-β pathways), under a physiological hypoxic condition. These chemical-induced NPCs (ciNPCs) resemble mouse brain-derived NPCs regarding their proliferative and self-renewing abilities, gene expression profiles, and multipotency for different neuroectodermal lineages in vitro and in vivo. Regulation of blood pressure by the arterial baroreflex and autonomic nervous system.

Mean arterial pressure (MAP) is a critical hemodynamic factor. The absence of proper regulation of MAP can have important pathophysiological consequences. Low MAP can cause inadequate blood flow to organs, syncope, and shock. On the other hand, elevated MAP contributes to increased oxygen demand by the heart, ventricular remodeling, vascular injury, end organ damage, and stroke. The arterial baroreflex system is a key controller of MAP and is a complex system. It can be considered in its entirety as an integrative physiological system or in terms of its regulated component parts. Those compone

Syndapin--a membrane remodelling and endocytic F-BAR protein.

Syndapin [also called PACSIN (protein kinase C and casein kinase II interacting protein)] is an Fes-CIP4 homology Bin-amphiphysin-Rvs161/167 (F-BAR) and Src-homology 3 domain-containing protein. Three genes give rise to three main isoforms in mammalian cells. They each function in different endocytic and vesicle trafficking pathways and provide critical links between the cytoskeletal network in different cellular processes, such as neuronal morphogenesis and cell migration. The membrane remodelling activity of syndapin via its F-BAR domain and its interaction partners, such as dynamin and neur

Nongenomic actions of adrenal steroids in the central nervous system.

Mineralocorticoids and glucocorticoids are steroid hormones that are released by the adrenal cortex in response to stress and hydromineral imbalance. Historically, adrenocorticosteroid actions are attributed to effects on gene transcription. More recently, however, it has become clear that genome-independent pathways represent an important facet of adrenal steroid actions. These hormones exert nongenomic effects throughout the body, although a significant portion of their actions are specific to the central nervous system. These actions are mediated by a variety of signalling pathways, and lea

Ash1a and Neurogenin1 function downstream of Floating head to regulate epiphysial neurogenesis.

The homeodomain transcription factor Floating head (Flh) is required for the generation of neurones in the zebrafish epiphysis. It regulates expression of two basic helix loop helix (bHLH) transcription factor encoding genes, ash1a (achaete/scute homologue 1a) and neurogenin1 (ngn1), in epiphysial neural progenitors. We show that ash1a and ngn1 function in parallel redundant pathways to regulate neurogenesis downstream of flh. Comparison of the epiphysial phenotypes of flh mutant and of ash1a/ngn1 double morphants reveals that reduced expression of ash1a and ngn1 can account for most of the ne

Enteric nervous system progenitors are coordinately controlled by the G protein-coupled receptor EDNRB and the receptor tyrosine kinase RET.

The enteric nervous system (ENS) in vertebrates is derived mainly from vagal neural crest cells that enter the foregut and colonize the entire wall of the gastrointestinal tract. Failure to completely colonize the gut results in the absence of enteric ganglia (Hirschsprung's disease). Two signaling systems mediated by RET and EDNRB have been identified as critical players in enteric neurogenesis. We demonstrate that interaction between these signaling pathways controls ENS development throughout the intestine. Activation of EDNRB specifically enhances the effect of RET signaling on the prolife

Neural stem cell-derived extracellular vesicles drive early neuroprotective and anti-apoptotic responses in spinal cord injury organotypic slices.

INTRODUCTION: Spinal cord injury (SCI) is a devastating neurological condition with limited regenerative capacity. Stem cell-based approaches have emerged as promising strategies due to their neuroprotective and immunomodulatory properties, largely mediated by small extracellular vesicles (sEVs) and their molecular cargo, including miRNAs. In this study, we aimed to evaluate the neuroprotective and anti-apoptotic potential of sEVs derived from SPC-01 and iMR-90 neural stem cell sources using an in vitro rat model of SCI. METHODS: sEVs were isolated from SPC-01 and iMR-90 culture media and cha

Selective neuroprotection in the ALS-FTD spectrum: Mechanisms of neuronal resilience and translational perspectives.

BACKGROUND: Amyotrophic lateral sclerosis (ALS) is increasingly recognized as part of a broader clinico-pathological continuum encompassing frontotemporal dementia (FTD). Despite progressive degeneration across motor and cognitive neural systems associated with TDP-43 pathology, specific neuronal populations-including the oculomotor and abducens nuclei, Onuf's nucleus, and sensory dorsal column pathways-remain relatively preserved. OBJECTIVE: Selectively preserved neuronal structures in ALS have largely been described as isolated neuropathological observations without sufficient mechanistic i

Structural connectome architecture and biological vulnerability shape cortical atrophy in cocaine use disorder.

BACKGROUND: Cocaine use disorder (CUD) is highly prevalent and characterized by widespread gray matter atrophy across the cerebral cortex. Yet, it remains unclear whether and how connectome-based circuits and biological features shape these structural abnormalities. METHODS: We mapped cortical atrophy patterns in CUD (discovery cohort: N = 53; replication cohort: N = 74; controls: N = 364) onto the brain's structural connectome, functional connectivity, transcriptomic similarity and receptor similarity architecture. Using a multimodal and multiscale connectivity-based framework, we identified

Hybrid Feature Selection-Based Machine Learning and Deep Learning Framework for Biomarker Prediction From RNA-seq Data During Dengue Fever to Severe Dengue Progression.

BACKGROUND: Severe dengue (SD) represents a life-threatening progression of dengue virus infection. Early identification of patients at risk of transitioning from dengue fever (DF) to SD remains a major clinical challenge. Unraveling the transcriptomic changes underlying this progression may aid in developing timely therapeutic interventions. METHODS: RNA-seq datasets comprising 103 samples (62 SD and 41 DF) were retrieved from the GEO repository. Following normalization using DESeq2, differentially expressed genes (DEGs) were identified between the 2 disease stages. Functional enrichment ana

Haemaphysalis longicornis and Carvacrol as Acaricide: Efficacy and Mechanism of Action.

Carvacrol derived from Origanum vulgare oil was evaluated for its acaricidal efficacy and mechanism of action against Haemaphysalis longicornis, a primary vector of severe fever with a thrombocytopenia syndrome. Essential oils extracted from O. vulgare leaves cultivated in Germany, Albania, and Iran were analyzed. Among them, the German oil exhibited the highest acaricidal potency due to its elevated carvacrol content (83.38%). Carvacrol was isolated and its identity was confirmed using GC/MS, NMR, and FT-IR analyses. Carvacrol demonstrated significant toxicity across all developmental stages

Neuropathic pain drives time-dependent reorganization of corticostriatal circuits.

Chronic pain fundamentally alters sensorimotor integration and motivated behaviors, yet the neural mechanisms underlying this transition remain poorly understood. The striatum, composed of dopamine receptor type 1 (D1)- and type 2 (D2)- expressing spiny projection neurons (SPN), integrates cortical sensory and motor inputs to coordinate movement and motivation, making it a critical candidate for mediating pain-induced behavioral adaptations. Although spinal and cortical pain circuits are well-characterized in limited phases of pain, how corticostriatal pathways and distinct striatal cell popul
