Bongkrekic acid reprograms hepatic lipid metabolism to promote systemic organ injury.

Bongkrekic acid (BK), a highly lethal mitochondrial toxin produced by Burkholderia gladioli pathovar cocovenenans, poses a significant threat to food safety. Although clinical reports have documented its harmful effects, the underlying metabolic mechanisms driving BK toxicity remain fullly understood. In this study, we utilized multi-omics techniques to explore the impact of BK on metabolic processes across multiple organs using a mouse model. Our results demonstrated that BK treatment significantly altered the liver's cellular composition, with hepatocytes becoming the dominant cell type. Not

Modulation of inflammatory pathways by omega-3 fatty acids in knee joint health for the management of post-traumatic osteoarthritis: a review.

The knee joint facilitates movement essential for daily activities. Its intricate anatomy makes it particularly prone to injury, specifically to the anterior cruciate ligament (ACL). ACL injury is a leading cause of post-traumatic osteoarthritis (PTOA), a degenerative joint disease that affects millions and results in pain, decreased mobility, and reduced quality of life. The cascade of initial inflammatory activity triggered by ACL injury plays a critical role in the pathogenesis of PTOA, as elevated levels of pro-inflammatory cytokines and catabolic enzymes lead to ongoing joint damage and d

Mitochondrial Metabolism in Neutrophils: Emerging Roles in Inflammatory Skin Diseases.

Neutrophils, the most abundant of the circulating leucocytes, play crucial roles in antimicrobial defence, tissue remodelling, and immune regulation. Traditionally regarded as predominantly glycolytic, relying on aerobic glycolysis (the Warburg effect) for rapid ATP generation, neutrophils are now recognised to possess broader metabolic flexibility. Emerging evidence has revealed their capacity for oxidative phosphorylation (OXPHOS), fatty acid oxidation (FAO), and glutaminolysis. As a result, mitochondrial metabolism is dynamically reprogrammed during differentiation and activation. These met

Drug-induced hyperuricemia: multi-pathway regulation, causative drugs, and individualized management strategies.

Hyperuricemia, a prevalent metabolic disorder, is not only the primary cause of gout but also an independent risk factor for various chronic conditions, including hypertension, cardiovascular and cerebrovascular diseases, and diabetes mellitus, thereby posing a significant threat to multi-organ health. Iatrogenic factors represent a key pathogenic trigger for hyperuricemia. With the expanding spectrum of clinical medications and the widespread adoption of polypharmacy, drug-induced hyperuricemia (DIH) now affects up to 25% of hospitalized patients and over 80% of transplant recipients on cyclo

Targeting metabolic reprogramming in multiple myeloma: Molecular mechanisms and potential therapeutic targets.

Multiple myeloma (MM) is a hematologic malignancy characterized by clonal proliferation of malignant plasma cells. While proteasome inhibitors and novel immunotherapies have markedly shifted the clinical trajectory of multiple myeloma, the persistence of therapeutic resistance and disease recurrence remains a formidable challenge. Metabolic reprogramming has emerged as a critical functional layer driving MM progression and drug resistance. It extends beyond mere energetic anomalies, integrating genetic context, bone marrow microenvironmental stress, and therapeutic selection pressure into a dy

HDAC3 Regulates Transcriptional Networks Governing Decidualization.

Decidualization is essential for embryo implantation and maintenance of pregnancy, during which quiescent endometrial stromal fibroblasts proliferate and differentiate into decidual stromal cells. Emerging evidence indicates that epigenetic regulators, including histone modifications, play critical roles in uterine receptivity, implantation, and stromal cell decidualization. Our previous study demonstrated that loss of histone deacetylase 3 (HDAC3) impairs endometrial receptivity and decidualization, resulting in female infertility. However, the genome-wide transcriptomic alterations responsib

GPAT4 regulates antitumor immune response through activating the mtDNA-cGAS axis in cancer cells.

Activating tumor-intrinsic innate immune pathways can promote tumor antigen presentation and immune cell recruitment, thereby improving the immunosuppressive microenvironment and overcoming resistance to immune checkpoint blockade (ICB). However, the specific molecular targets remain insufficiently defined. Bioinformatic analysis revealed that Glycerol-3-phosphate acyltransferase 4 (GPAT4) is highly expressed in various tumors, including pancreatic cancer and breast cancer, and is significantly associated with low immunogenicity, poor immune-cell infiltration, and unfavorable prognosis. The global epidemiology of alcohol-associated liver disease.

Alcohol-associated harm remains a major, largely preventable driver of global morbidity, mortality, and societal costs, and alcohol-associated liver disease (ALD) is a major consequence of long-term exposure. According to the World Health Organization, average consumption was 5.0 L of pure alcohol per person aged ≥15 years in 2022, with 21% unrecorded consumption and substantial regional variation. Per-capita intake declined from 5.7 L in 2010 to 5.0 L in 2022, yet current models forecast an increase to 5.7 L by 2030. In 2023, an estimated 3.15 million people were living with alcohol-associate

Human retinal organoid model of disease-relevant photoreceptor cell death amenable to drug screening.

Dry age-related macular degeneration (AMD) is characterized by the progressive loss of retinal pigment epithelium cells in the macula, leading to photoreceptor degeneration and loss of central vision. Current treatments only modestly delay disease progression, but once photoreceptors are damaged, vision loss becomes irreversible. Therefore, there is an urgent need to develop therapies that prevent photoreceptor cell death and that may complement current and emerging treatment strategies. A critical step toward this goal is establishing pathophysiologically relevant human disease models for the

A novel tRNA-derived small RNA tsRNA-Ile-5-0044 is a candidate biomarker for laryngeal squamous cell carcinoma.

tRNA-derived small RNAs (tsRNAs) have emerged as pivotal regulators and promising biomarkers in multiple malignancies, however their functional roles and clinical significance in HNSCC remain incompletely elucidated. Here, we found tsRNA-Ile-5-0044 was significantly upregulated in HNSCC tissues, notably, survival analysis demonstrated that its prognostic significance was confined to HNSCC and mesothelioma, underscoring its specificity for HNSCC diagnosis and prognosis. qRT-PCR validation confirmed that tsRNA-Ile-5-0044 expression was markedly elevated in both LSCC tissues and plasma, and highe

TAK1 Regulates Skeletal Muscle Mass, Hypertrophic Signaling, and Metabolic Homeostasis in Male and Female Mice.

Skeletal muscle is the most abundant tissue in the human body and is essential for locomotion and the regulation of whole-body metabolism. The maintenance of skeletal muscle mass is essential for health, yet the molecular and signaling mechanisms that control skeletal muscle mass remain poorly understood. Transforming growth factor-β-activated kinase 1 (TAK1) is a key signaling protein that regulates multiple intracellular pathways. Recent studies have demonstrated that TAK1 is a critical regulator of skeletal muscle mass. However, the mechanisms by which TAK1 regulates muscle mass and whether

MicroRNA-Ferroptosis-Spinal Cord Injury: A Complex Interplay in Neurodegeneration and Repair.

Spinal cord injury (SCI) is a devastating neurological condition characterized by irreversible primary damage followed by a complex secondary injury cascade involving oxidative stress, neuroinflammation, iron dysregulation, and regulated cell death. Among these mechanisms, ferroptosis, a distinct, iron-dependent form of regulated cell death driven by lipid peroxidation and redox imbalance, is increasingly recognized as a critical mediator of neurodegeneration after SCI. The miRNAs play essential roles in neural injury responses by modulating inflammation, oxidative stress, and cell-death pathw

Effects of adverse childhood experiences on behavior in adolescence: Insights from brain imaging and genetic risk.

Adolescence is a critical developmental stage, and adverse childhood experiences (ACEs) are important environmental factors influencing behavior. However, the relationship between ACEs, individual behavior, brain mechanisms, and the role of genetic risk remains underexplored. In this study, data from 9796 youths aged 9-13 years in the Adolescent Brain Cognitive Development (ABCD) study were analyzed, including 67 ACE-related variables and 30 behavioral outcomes. Using sparse canonical correlation analysis, we identified two distinct patterns: a parental mental health-child psychopathology, lin

Long-Term Consequences of Perinatal Asphyxia in the Development of Alzheimer's Disease.

Despite remarkable progress in medicine, perinatal asphyxia (PA) remains a significant clinical problem, and concerns regarding its long-term complications are increasingly being raised. Emerging evidence indicates that similar pathological pathways are activated following PA and during neurodegeneration in Alzheimer's disease (AD). These similarities involve various mechanisms, including alterations in AD-related proteins and their gene expression. Disturbances in neurotransmitter function can lead to excitotoxicity and cell death via necrosis and delayed apoptosis. Additionally, as observed
