[Genetic background of rheumatoid arthritis in connection with a family tree].

Authors present a rare history of a family with accumulation of autoimmune diseases. From eight siblings four had rheumatoid arthritis, one systemic lupus erythematodes, one primary Sjögren syndrome, and one Reiter disease. Interestingly, neither the parents nor the offsprings were affected. Because of the possible genetic background, the authors performed HLA serologic and DNA investigations in nine members of the family. The results showed, in agreement with data from the literature, the accumulation of HLA-DQ7 (DQB1*0301), DR4, B27 and DR6 (DR13) risk factors. This observation confirms, tha

Reconstruction of the block matching profiles.

Block matching is a valuable tool for selecting donors for bone marrow transplantation. Identical, electrophoretic profiles of unrelated bone marrow donor-recipient pairs have been shown to be associated with long-term survival and a reduction of graft versus host disease (GVHD). This study was undertaken to determine the sequences of the PCR products which are generated. PCR products obtained with beta-block primers following the amplification of DNA extracted from cell lines homozygous for 7.1 and 8.1 ancestral haplotypes were cloned and sequenced. The PCR products were characterised and the

Determination of alpha thalassaemia phenotypes by messenger RNA analysis.

The possibility of using alpha/beta globin messenger RNA (mRNA) ratios to distinguish between the carrier states for different forms of alpha thalassaemia has been explored. Alpha/beta globin mRNA ratios were determined in the red cells of a series of normal individuals and in members of four Cypriot families, one Thai and one Chinese family in which at least one person has haemoglobin H disease. It was found that there was a clear distinction in the ratios between normals, alpha thalassaemia 1 carriers, alpha thalassaemia 2 carriers, and those with haemoglobin H disease. This method should be

Influence of chemotherapy and serum factors on the mitogenic response of peripheral leucocytes of leprosy patients to phytohaemagglutinin.

Mitogenic response to phytohaemagglutinin (PHA) of peripheral leucocytes from patients with polar forms of leprosy namely lepromatous leprosy (LL) and tuberculoid leprosy (TT) has been evaluated. No defect in blast transformation as assessed by the incorporation of 3H-thymidine into DNA was observed in tuberculoid leprosy patients. In lepromatous leprosy patients, the response varied with the status of the patient. Leucocytes from untreated cases of lepromatous leprosy showed a depressed response to PHA, whereas the leucocytes from patients treated with diaminodiphenylsulphone (DDS) gave an en

A Colorimetric Approach to Identifying Single-Nucleotide Mutations Using Gold-Based Magnetic Nanoparticles.

Detection of single-nucleotide mutations (SNPs) in the genome of infectious agents is one of the ways to identify their drug-resistant strains. Antibiotic-resistant strains of Mycobacterium tuberculosis (MTB) impede advances in the fight against the spread of tuberculosis. Diagnostic tools exist for both MTB itself and its drug-resistant strains, most of which are technically complex, resource-intensive, and time-consuming. This paper presents the development of a colorimetric test system, requiring no expensive equipment, for the rapid and accurate determination of the SNPs associated with dr

Redox-responsive dual-drug nanomedicine integrating cisplatin and trypsin for synergistic reversal of tumor chemoresistance.

To overcome cisplatin resistance without increasing systemic toxicity, we rationally elaborated a glutathione-activatable prodrug nanomedicine that chemically co-encapsulates cisplatin and a masked protease. First, a redox-labile succinimide linker (NC-ss-COOH) was covalently grafted onto the ε-amino groups of trypsin to create a "pro-protease" (ssTrypsin) whose catalytic activity is completely silenced in circulation but instantly restored (≥96%) upon cleavage by intratumoral GSH. Simultaneously, cisplatin was stably coordinated to the carboxyl-rich backbone of cRGD-PEG-polyglutamic acid and

Genetic Markers Remain Detectable in Genetically Engineered Microbes Biocontained with a CRISPR Kill Switch.

Biocontainment strategies, such as kill switches, have been developed to avoid the unintended proliferation of genetically engineered microbes (GEMs) intended for open-release environmental applications. However, the presence of GEM DNA after successful biocontainment presents new environmental risks and challenges for monitoring. In this study, we investigated whether biocontainment using a CRISPR-Cas9 kill switch, which causes double-strand breaks in target genes essential for GEM growth, could resolve this challenge in a model Escherichia coli GEM. Surprisingly, the escape rates of the GEM

An Engineering-Editable Hybrid Membrane‑Camouflaged DNA Origami Drug Enables Checkpoint Blockade and Immune Activation for Triple-Negative Breast Cancer Immunotherapy.

The clinical response to immunotherapy in triple-negative breast cancer (TNBC) is often limited by the immunosuppressive microenvironment, immune cell exhaustion, and multidrug resistance. Through a high-throughput screen of 2,500 FDA-approved drugs using a three-dimensional tumor organoid platform, we identified thonzonium bromide (THO) as an inducer of gasdermin D (GSDMD)-mediated pyroptosis, which results in the release of damage-associated molecular patterns (DAMPs) to activate antitumor immunity. To achieve targeted delivery, we designed a core-shell-structured biomimetic nanoparticle (TH

Epigenetic regulation and chromatin organization in cystic fibrosis airways.

Cystic fibrosis (CF) is caused by pathogenic variants of the Cystic Fibrosis Transmembrane Conductance Regulator (CFTR) gene. CF in the airways is associated with dehydrated and hyperconcentrated mucus, impaired mucociliary transport, and inflammation. Although CF is a genetic disorder, CFTR sequence variations alone may not fully explain its phenotypic variation. Additionally, while pharmacological CFTR modulators that correct CFTR protein function greatly improve clinical outcomes, people with CF treated with modulators can still exhibit abnormal mucus and persistent inflammation compared to

Pan-cancer analysis of GINS1 identifies its prognostic and immunotherapy predictive value with in vitro experimental validation in nasopharyngeal carcinoma.

BACKGROUND: GINS complex subunit 1 (GINS1) is part of the tetrameric GINS complex involved in DNA replication, playing a vital role in controlling the start and continuation of DNA replication. Its role in cancer, however, is not well understood. Still, its relevance to cancer is uncertain. This study focused on exploring the role of GINS1 in different forms of cancer. METHODS: Using a range of publicly available databases, we conducted a comprehensive study on the oncogenic function of GINS1 in pan-cancer through analyses of differential expression, survival, enrichment, gene mutations and t

Emerging Role of Natural Topoisomerase Inhibitors as Anticancer agents.

Topoisomerases I and II are the functionally two forms of DNA topoisomerase. In anticancer research, novel anticancer chemotherapeutical capable of blocking topoisomerase enzymes have been discovered. Most commonly, topoisomerase causes replication fork arrest and doublestrand breaks, and this is how a clinically successful topoisomerase-targeting anticancer medicines work. Unfortunately, this novel mechanism of action has been linked to the development of secondary malignancies as well as cardiotoxicity. The specific binding locations and mechanisms of topoisomerase poisons have been identifi

Effects of the histone-like protein on biofilm formation and pathogenicity of Listeria monocytogenes.

Histone-like protein HU is essential for DNA recombination, repair, and transcriptional regulation in bacteria. However, the physiological roles of HU proteins in Listeria monocytogenes (LmHU) remain unexplored. Given the significant biofilm-forming ability of this foodborne pathogen and its associated cross-contamination risks, identifying novel control targets is critical. LmHU, as the sole double-stranded DNA-binding protein in L. monocytogenes, is a promising candidate. This study systematically explored its contributions to biofilm formation, motility, and the regulation of virulence fact

Mannheimia indoligenes sp. nov., proposed for clade V organisms of Mannheimia.

A set of 25 strains belonging to clade V of Mannheimia mainly isolated from cattle was investigated and is proposed to represent Mannheimia indoligenes sp. nov. The species can be separated from the other validly published species of the genus by pheno- and genotype. Only indole separates M. indoligenes and Mannheimia varigena while two to seven characters separate M. indoligenes from other species of Mannheimia. Thirteen strains belonging to biogroups 6, 7, 8C, 9, 10, 12 and UG5 formed a monophyletic group based on 16S rRNA gene sequence comparisons with 98-100 % similarity. Eight of these st

Genomic and Phenotypic Characterization of Streptomyces sirii sp. nov., Amicetin-Producing Actinobacteria Isolated from Bamboo Rhizospheric Soil.

In our large-scale search for antimicrobial-producing bacteria, we isolated an actinomycete strain from rhizospheric soil of Bambusa vulgaris. The strain designated BP-8 showed noticeable antibacterial activity. BP-8 was subjected to a whole-genome analysis via a polyphasic taxonomy approach, and its antibacterial metabolite was identified by HRLS-MS. The results of the physiological and morphological analyses indicated that BP-8 is an aerobic, neutrophilic, mesophilic organism that is tolerant to 8% NaCl and can use a wide range of carbohydrates. It forms curly sporophores with a warty surfac

Comparison on binding interactions of quercetin and its metal complexes with calf thymus DNA by spectroscopic techniques and viscosity measurement.

Quercetin (Qu) and its metal complexes have received great attention during the last years, due to their good antioxidant, antibacterial, and anticancer activities. In this contribution, binding interactions of Qu and Qu-metal complexes with calf thymus DNA (ctDNA) were investigated and compared systematically by using spectroscopic techniques and viscosity measurement. UV-vis absorption spectra of ctDNA-compound systems showed obvious hypochromic effect. Relative viscosity and melting temperature of ctDNA increased after the addition of Qu and Qu-metal complexes, and the change tendency is Qu

Rapid, multiplexed detection of the let-7 miRNA family using γPNA amphiphiles in micelle-tagging electrophoresis.

miRNA is a promising class of biomarkers whose levels can be assayed to detect various forms of cancer and other serious diseases. These short, noncoding nucleic acids are difficult to detect due to their low abundance and the marginal stability of their duplexes with DNA probes. In addition, miRNAs within the same family have high sequence homology, and often, related miRNA differ in sequence by only a single base. In this report, we demonstrate an independent detection seven members of the let-7 family of miRNA in a single run. Key to success is the use of mini-PEG-substituted PNA amphiphile

Sunscreens Containing Cyclodextrin Inclusion Complexes for Enhanced Efficiency: A Strategy for Skin Cancer Prevention.

Unprotected exposure of skin to solar ultraviolet radiation (UVR) may damage the DNA of skin cells and can lead to skin cancer. Sunscreens are topical formulations used to protect skin against UVR. The active ingredients of sunscreens are UV filters that absorb, scatter, and/or reflect UVR. Preventing the formation of free radicals and repairing DNA damages, natural antioxidants are also added to sunscreens as a second fold of protection against UVR. Antioxidants can help stabilise these formulations during the manufacturing process and upon application on skin. However, UV filters and antioxi

Activity of Antimicrobial Peptides and Ciprofloxacin against Pseudomonas aeruginosa Biofilms.

Pseudomonas aeruginosa is increasingly resistant to conventional antibiotics, which can be compounded by the formation of biofilms on surfaces conferring additional resistance. P. aeruginosa was grown in sub-inhibitory concentrations of the antimicrobial peptides (AMPs) melimine and Mel4 or ciprofloxacin for 30 consecutive days to induce the development of resistance. Antibiofilm effect of AMPs and ciprofloxacin was evaluated using crystal violet and live/dead staining with confocal microscopy. Effect on the cell membrane of biofilm cells was evaluated using DiSC(3)-5 dye and release of intrac

Methylobacterium nonmethylotrophicum sp. nov., isolated from tungsten mine tailing.

A novel pink-pigmented strain, designated 6HR-1T, was isolated from tungsten mine tailings in Jiangxi Province, PR China. Cells were Gram-stain-negative, aerobic, non-spore-forming, rod-shaped and motile with a polar flagellum (monotrichous). It could not utilize methanol, methylamine, formaldehyde or formate as a sole carbon source. The methanol dehydrogenase mxaF gene was absent but the xoxF gene was present. Phylogenomic and 16S rRNA gene phylogenetic analyses clearly showed that strain 6HR-1T was affiliated to the genus Methylobacterium and closely related to 'Methylobacterium terrae' 17Sr
