Aberrant Mannosylated and Highly Fucosylated Glycoepitopes of Prostatic Acid Phosphatase as Potential Ligands for Dendritic-Cell Specific ICAM-Grabbing Nonintegrin (DC-SIGN) in Human Seminal Plasma-A Step towards Explaining Idiopathic Infertility.

Semen prostatic acid phosphatase (PAP) has been proposed as an endogenous ligand for dendritic cell-specific ICAM-3-grabbing nonintegrin (DC-SIGN), which plays a critical immuno-modulating role in maintaining homeostasis in the female reproductive tracts. In the current study, we assumed that semen PAP bears a set of fucosylated and mannosylated glycans, which may mediate the efficient binding of PAP to DC-SIGN. To investigate this hypothesis, we developed ELISA assays using Galanthus nivalis and Lotus tetragonolobus lectins capable of binding mannose-containing glycans or LewisX and LewisY mo

Low Interfacial Resistance and Superior Suppression to Li-Dendrite Penetration Facilitated by Air-Stable and Mechanically Robust Al/Mg-Co-Doped Li-La-Zirconate as Electrolyte for Li-Based Solid-State Cells.

In the context of usage as a solid electrolyte (SE) for Li-based solid-state cells, the interfacial characteristics of Li-La-zirconate (LLZO) with the electrodes and the mechanical properties of LLZO influence the overall impedance and stability. In this regard, the newly developed air-stable Al/Mg-co-doped LLZO has been found to possess greater resistance to crack propagation (by ∼31%) and fracture stress (by ∼52%), along with elevated hardness and stiffness, as compared to simply Al-doped LLZO. Furthermore, as directly visualized via cross-section electron microscopy at the Li/LLZO interface

Statin Drugs Plus Th1 Cytokines Potentiate Apoptosis and Ras Delocalization in Human Breast Cancer Lines and Combine with Dendritic Cell-Based Immunotherapy to Suppress Tumor Growth in a Mouse Model of HER-2pos Disease.

A dendritic cell-based, Type 1 Helper T cell (Th1)-polarizing anti-Human Epidermal Growth Factor Receptor-2 (HER-2) vaccine supplied in the neoadjuvant setting eliminates disease in up to 30% of recipients with HER-2-positive (HER-2pos) ductal carcinoma in situ (DCIS). We hypothesized that drugs with low toxicity profiles that target signaling pathways critical for oncogenesis may work in conjunction with vaccine-induced immune effector mechanisms to improve efficacy while minimizing side effects. In this study, a panel of four phenotypically diverse human breast cancer lines were exposed in v

Rapamycin-conditioned, alloantigen-pulsed myeloid dendritic cells present donor MHC class I/peptide via the semi-direct pathway and inhibit survival of antigen-specific CD8(+) T cells in vitro and in vivo.

Dendritic cells (DC) are "professional" bone marrow-derived antigen (Ag)-presenting cells of interest both as therapeutic targets and potential cellular vaccines due to their ability to regulate innate and adaptive immunity. Harnessing the inherent tolerogenicity of DC is a promising and incompletely explored approach to the prevention of allograft rejection. Previously, we and others have reported the ability of pharmacologically-modified DC, that resist maturation, to inhibit CD4(+) T cell responses and prolong allograft survival. Here we evaluated the ability of murine myeloid DC conditione

Thymoquinone from nutraceutical black cumin oil activates Neu4 sialidase in live macrophage, dendritic, and normal and type I sialidosis human fibroblast cells via GPCR Galphai proteins and matrix metalloproteinase-9.

Anti-inflammatory activities of thymoquinone (TQ) have been demonstrated in in vitro and in vivo studies. However, the precise mechanism(s) of TQ in these anti-inflammatory activities is not well understood. Using a newly developed assay to detect sialidase activity in live macrophage cells (Glycoconj J doi: 10.1007/s10719-009-9239-8 ), here we show that TQ has no inhibitory effect on endotoxin lipopolysaccharide (LPS) induced sialidase activity in live BMC-2 macrophage cells. In contrast, the parent black seed oil (BSO) and another constituent of BSO para-cymene (p-CY) completely block LPS in

Immunohistologic analysis of ineffective CD40-CD40 ligand interaction in lymphoid tissues from patients with X-linked immunodeficiency with hyper-IgM. Abortive germinal center cell reaction and severe depletion of follicular dendritic cells.

The CD40 ligand (CD40L) is an activation-induced surface membrane protein expressed by CD4+ T helper cells in lymphoid follicles, and is involved in the contact-dependent signaling-mediated activation, proliferation, and differentiation of CD40+ B cells. Using immunohistochemistry, the present study analyzes the cell microenvironment of lymphoid tissues in two cases of X-linked hyper-IgM syndrome, a congenital immunodeficiency caused by mutations of the CD40L gene, and which represents a unique model to dissect the functional and morphologic consequences of disrupted CD40/CD40L interactions. Persistent ashen-gray maculae and freckles induced by long-term PUVA treatment.

Persistent ashen-gray maculae 5-7 mm in diameter and freckles developed after prolonged PUVA therapy of psoriasis. Histopathology of maculae showed extensive collections of melanophages in the upper dermis. The melanocytes were strongly stimulated at the site of the freckles. The number of melanosomes, mitochondria, endoplasmic reticulum and dendrites were increased. Pathological features such as large amounts of lipid droplets and lysosome-melanosome complexes were also found within the melanocytes.

Changes in the subcortical white matter and the pyramidal neurons in the sensorimotor cortex of juvenile hydrocephalic rats.

Hydrocephalus, the aberrant enlargement of the brain's ventricles, is caused by a build-up of cerebrospinal fluid that stretches the periventricular white matter and may disrupt the connections between the pyramidal neurons in the cerebral cortex. Using an intra-cisternal kaolin injection hydrocephalic rat model, we aimed to determine if the effects of hydrocephalus on the white matter will subsequently impact the dendrites of cortical pyramidal neurons, which are the synaptic sites for the white matter fibres, particularly the afferent fibres. Three-week-old hydrocephalic rats were compared w…

Studies of consciousness are hindered by the complexity of the brain, but it is possible to study the consciousness of a sensation, namely p … quency input results in the development of a long-term potentiation (LTP) at synapses on the apical dendrites of pyramidal neurons. The LTP acts via α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) and N-methyl-D-aspartate (NMDA) receptors, and an anterior cingulate cortex-specific type-1 adenylate cyclase is necessary for both the LTP and the pa …

Disruptive lysosomal-metabolic signaling and neurodevelopmental deficits that precede Purkinje cell loss in a mouse model of Niemann-Pick Type-C disease.

Purkinje cell (PC) loss occurs at an early age in patients and animal models of Niemann-Pick Type C (NPC), a lysosomal storage disease caused by mutations in the Npc1 or Npc2 genes. Although degeneration of PCs occurs early in NPC, little is known about how NPC1 deficiency affects the postnatal development of PCs. Using the Npc1nmf164 mouse model, we found that NPC1 deficiency significantly affected the postnatal development of PC dendrites and synapses. The developing dendrites of Npc1nmf164 PCs were significantly deficient in mitochondria and lysosomes. Furthermore, anabolic (mTORC1) and cat…

Regulation of the Interfaces Between Argyrodite Solid Electrolytes and Lithium Metal Anode.

Lithium-ion batteries (LIBs) are widely used in portable electronic devices, electric vehicles and large scale energy storage, due to their considerable energy density, low cost and long cycle life. However, traditional liquid batteries suffer from safety problems such as leakage, thermal runaway and even explosion. Part of the issues are caused by lithium dendrites puncturing the liquid electrolyte during cycling. In order to achieve the objective of higher safety and energy density, a rigid solid-state electrolyte (SSE) is proposed instead of liquid electrolyte (LE). Thereinto, sulfide SSEs …

HIV influences microtubule associated protein-2: potential marker of HIV-associated neurocognitive disorders.

OBJECTIVE: Postmortem brains of patients diagnosed with HIV-1-associated neurocognitive disorders (HAND) exhibit loss of dendrites. However, the mechanisms by which synapses are damaged are not fully understood. DESIGN: Dendrite length and remodeling occurs via microtubules, the dynamics of which are regulated by microtubule-binding proteins, including microtubule-associated protein 2 (MAP2). The HIV protein gp120 is neurotoxic and interferes with neuronal microtubules. We measured MAP2 concentrations in human cerebrospinal fluid (CSF) and MAP2 immunoreactivity in rat cortical neurons exposed…

Melanogenic effect of purified mushroom tyrosinase on B16F10 Melanocytes: A Phase Contrast and Immunofluorescence Microscopic Study.

Cutaneous melanin pigment plays a critical role in camouflage, mimicry, social communication, and protection against harmful effects of solar radiation. It has been proven that tyrosinase plays a pivotal role in melanocytes dendrite formation; however, the molecular mechanism underlying this process has not been fully elucidated. The morphological changes were observed under a phase contrast microscope. These changes were evident, with globular cell bodies and increased numbers of tree branch-like dendrites. The present work aimed to study the morphoanatomic effects of purified tyrosinase to d…

Prickle1 is expressed in distinct cell populations of the central nervous system and contributes to neuronal morphogenesis.

Development of axons and dendrites constitutes a critical event in neuronal maturation and seems to require signaling through the planar cell polarity (PCP) pathway. Mutations in components of the PCP pathway lead to a spectrum of neurological phenotypes and disorders. For example, a missense mutation in Prickle 1 (Pk1) is associated with progressive myoclonus epilepsy (PME) in humans, and its reduced gene dosage increases sensitivity to induced seizure in mice. In an effort to unravel the role of the PCP pathway in mammalian neuronal development, we examined the expression of Pk1 in the centr…

Synaptologic and fine structural features distinguishing a subset of basal forebrain cholinergic neurons embedded in the dense intrinsic fiber network of the caudal extended amygdala.

Cholinergic basal forebrain neurons confined within the intrinsic connections of the extended amygdala in the caudal sublenticular region and anterior amygdaloid area (cSLR/AAA) differ from other basal forebrain cholinergic neurons in several morphological and neurochemical respects. These cSLR/AAA cholinergic neurons have been subjected to additional investigations described in this report. First, fibers traced anterogradely following injections of Phaseolus vulgaris-leucoagglutinin in the central amygdaloid nucleus were shown to contact cSLR/AAA cholinergic neurons and dendrites. Second, the…

Analyses of smooth endoplasmic reticulum of cerebellar parallel fibers in aging, ethanol-fed rats.

The smooth endoplasmic reticulum (SER), a calcium storage organelle, is essential for normal neuronal function. Dilation of the SER is pathologic and a threat to neuronal calcium homeostasis. Dilation of the SER has been reported within the dendrites of cerebellar Purkinje neurons of aging rats after lengthy ethanol treatment. Ethanol-related alterations of parallel fiber SER have not been investigated despite the fact that such dilation may precede and contribute transsynaptically to SER dilation and degeneration in Purkinje neuron dendrites. Male Fischer 344 rats (n = 120; age = 12 months ol…

Abnormalities in neuronal process extension, hippocampal development, and the ventricular system of L1 knockout mice.

In humans, mutations in the L1 cell adhesion molecule are associated with a neurological syndrome termed CRASH, which includes corpus callosum agenesis, mental retardation, adducted thumbs, spasticity, and hydrocephalus. A mouse model with a null mutation in the L1 gene (Cohen et al., 1997) was analyzed for brain abnormalities by Nissl and Golgi staining and immunocytochemistry. In the motor, somatosensory, and visual cortex, many pyramidal neurons in layer V exhibited undulating apical dendrites that did not reach layer I. The hippocampus of L1 mutant mice was smaller than normal, with fewer …
