Cysteamine prevents inhibition of adenylate kinase caused by cystine in rat brain cortex.

Cystinosis is a systemic genetic disease caused by a lysosomal transport deficiency accumulating cystine in the lysosomes of almost all tissues. Although tissue damage might depend on cystine accumulation, the mechanisms of tissue damage are still obscures. Adenylate kinase, along with creatine kinase, is responsible for the enzymatic phosphotransfer network, crucial for energy homeostasis. Taking into account that cystine is known to inhibit creatine kinase activity, the two enzymes have thiol groups, and the strong interaction between the two activities, our main objective was to investigate

Phenotypic changes and the fate of digestive enzymes during induction of amber disease in larvae of the New Zealand grass grub (Costelytra zealandica).

Amber disease of the New Zealand grass grub Costelytra zealandica (Coleoptera: Scarabaeidae) is caused by ingestion of pADAP plasmid carrying isolates of Serratia entomophila or Serratia proteamaculans (Enterobacteriaceae) and causes infected larvae to cease feeding and clear their midgut to a pale amber colour where midgut serine protease activities are virtually eliminated. Using bacterial strains and mutants expressing combinations of the anti-feeding (afp) and gut clearance (sep) gene clusters from pADAP, we manipulated the disease phenotype and demonstrated directly the relationship betwe

[Preparation of repeatedly and effectively usable L-asparaginase by a chemical modification].

The anti-tumor activity of L-asparaginase (EC. 3.5.1.1.) has been conclusively demonstrated. Its therapeutic application, however, has been hampered by its short clearance time in the circulation and high immunogenicity. In order to solve these problems, polyethylene glycol, a linear synthetic and non-immunogenic polymer, was introduced covalently into the amino groups in the enzyme molecule. Monomethoxypolyethylene glycol with molecular weight of 5000 was activated with cyanuric chloride to obtain activated PEGs [2, 4-bis (O-methoxypolyethyleneglycol)-6-chloro-s-triazine]. L-Asparaginase lost

Modulating thermal folded stability and catalytic activity of a de novo metalloenzyme through chemical backbone modification.

The relationship between conformational heterogeneity and catalytic activity in enzymes is complex and poses important fundamental biological questions. Few experimental tools are available to rationally control conformations within a folded protein ensemble. Modification of the protein backbone through chemical synthesis has previously been used to alter enzyme conformational dynamics at a site essential for catalysis. Here, we explore the application of the approach to manipulate inherent conformational preferences in a dynamic helical region of an enzyme remote from its active site. Focusin

Conserved Asp233-Asp246 hydrogen bond modulates active site dynamics in class A β-lactamases.

Rigid enzymes catalyze chemical reactions by stabilizing the transition state through a specific conformation. Catalytic residues are precisely positioned, and enzyme dynamics are kept to a minimum. It is thought that conserved residues around the active site (second-shell residues) play a crucial role in positioning catalytic residues. Asp233 and Asp246 are two highly conserved second-shell residues in class A β-lactamases, rigid enzymes that inactivate β-lactam antibiotics. The two aspartates share a short hydrogen bond, linking β-strands 3 and 4. The role of this interaction in Mycobacteriu

Insights on intermolecular FMN-heme domain interaction and the role of linker length in cytochrome P450cin fusion proteins.

Cytochrome P450s are an important group of enzymes catalyzing hydroxylation, and epoxidations reactions. In this work we describe the characterization of the CinA-CinC fusion enzyme system of a previously reported P450 using genetically fused heme (CinA) and FMN (CinC) enzyme domains from Citrobacter braaki. We observed that mixing individually inactivated heme (-) with FMN (-) domain in the CinA-10aa linker - CinC fusion constructs results in recovered activity and the formation of (2S)-2β-hydroxy,1,8-cineole (174 µM), a similar amount when compared to the fully functional fusion protein (176

Cyanide hydratases and cyanide dihydratases: emerging tools in the biodegradation and biodetection of cyanide.

The purpose of this study is to summarize the current knowledge of the enzymes which are involved in the hydrolysis of cyanide, i.e., cyanide hydratases (CHTs; EC 4.2.1.66) and cyanide dihydratases (CynD; EC 3.5.5.1). CHTs are probably exclusively produced by filamentous fungi and widely occur in these organisms; in contrast, CynDs were only found in a few bacterial genera. CHTs differ from CynDs in their reaction products (formamide vs. formic acid and ammonia, respectively). Several CHTs were also found to transform nitriles but with lower relative activities compared to HCN. Mutants of CynD

Lipid peroxidation and some antioxidant enzymes in nasal polyp tissue.

Nasal polyp (NP) is considered an inflammatory condition in nasal and paranasal sinus cavities and is frequently encountered in otolaryngology clinics. Although the pathophysiology of nasal polyps is poorly understood, it seems likely that the epithelium may play a critical role in the genesis of inflammatory nasal disease. The aim of this study was to investigate the role of free radicals and antioxidant enzymes in NP and compare these findings with concha bullosa (CB). NP and CB were obtained from 27 and 23 patients, respectively. Glutathione peroxidase (GSH-Px), catalase (CAT), xanthine oxi

Effect of Aerva lanata against hepatotoxicity of carbon tetrachloride in rats.

The partially purified petroleum ether extractable fraction of the whole plant Aerva lanata (PF) was evaluated for the protective effect against liver damage induced by carbon tetra chloride (CCl(4)) in Sprague Dawley rats. Rats were orally administered with PF (50 and 100mg/kg body weight) for 14 days before CCl(4) challenge and 100mg of PF alone for toxicity analysis without CCl(4) administration. The results showed that CCl(4) administration significantly damaged the liver as evident from histopathology and very high activity of serum and liver marker enzymes. It also reduced the antioxidan

Structure and action of bleomycin.

The structures of bleomycins and of other bleomycin-phleomycin group of antibiotics were described. The activity of bleomycins and their derivatives in causing strand scission of SV40 viral DNA suggests that the beta-aminoalanine amide moiety and the carbamoyl group are involved in this reaction. More than one guanido group in the terminal amine of bleomycin-phleomycin group antibiotics caused irreversible renal toxicity in dogs. Pulmonary toxicity varied depending on the terminal amines. A bleomycin-inactivating enzyme which distributes widely in animal cells was shown to be a new aminopeptid

Bidirectional effects of cuminaldehyde-pepsin interaction on enzymatic and antioxidant activity: Potential implications for gastrointestinal health and disorder modulation.

Pepsin is the primary proteolytic enzyme in the stomach and plays a pivotal role in gastric protein digestion, with dysregulation closely associated with gastrointestinal functional disorders. Cuminaldehyde, a dietary phytochemical abundant in Cuminum cyminum (cumin), is widely consumed and recognized for its antioxidant and gastrointestinal bioactivities. In this study, we systematically investigated the interaction between cuminaldehyde and pepsin under physiologically relevant conditions to elucidate its implications for gastric digestion, antioxidant function, and gastrointestinal health.

Osteoblast-Osteoclast Coculture Amplifies Inhibitory Effects of FG-4592 on Human Osteoclastogenesis and Reduces Bone Resorption.

The link between bone and blood vessels is regulated by hypoxia and the hypoxia-inducible transcription factor, HIF, which drives both osteogenesis and angiogenesis. The recent clinical approval of PHD enzyme inhibitors, which stabilize HIF protein, introduces the potential for a new clinical strategy to treat osteolytic conditions such as osteoporosis, osteonecrosis, and skeletal fracture and nonunion. However, bone-resorbing osteoclasts also play a central role in bone remodeling and pathological osteolysis, and HIF promotes osteoclast activation and bone loss in vitro. It is therefore likel

Molecular biology of microbial ureases.

Urease (urea amidohydrolase; EC 3.5.1.5) catalyzes the hydrolysis of urea to yield ammonia and carbamate. The latter compound spontaneously decomposes to yield another molecule of ammonia and carbonic acid. The urease phenotype is widely distributed across the bacterial kingdom, and the gene clusters encoding this enzyme have been cloned from numerous bacterial species. The complete nucleotide sequence, ranging from 5.15 to 6.45 kb, has been determined for five species including Bacillus sp. strain TB-90, Klebsiella aerogenes, Proteus mirabilis, Helicobacter pylori, and Yersinia enterocolitica

Thermal Modulation of Cerium-Based Nanozyme Activity for Colorimetric Detection of Ascorbic Acid.

Metal-organic framework (MOF) nanozyme materials have multiple application prospects, and the calcination process is an effective strategy for regulating and enhancing their enzyme-like catalytic performance. In this work, a series of Ce-ATPA-X materials were prepared by calcining the Ce-based framework precursor at 200-800 °C temperatures, and their structural characteristics and enzyme-like activities were systematically investigated. Among them, Ce-ATPA-400 exhibited the strongest oxidase-like (OXD-like) and peroxidase-like (POD-like) activities, approximately 2.4 and 2.6 times those of Ce-

Beyond model substrates: Reevaluating enzymes claimed to hydrolyze zearalenone.

Zearalenone (ZEN) is a prevalent estrogenic mycotoxin that contaminates cereal-based food and feed and poses a substantial risk to human and animal health. Enzymatic detoxification has emerged as a promising mitigation strategy, and several α/β-hydrolases have been shown to efficiently hydrolyze ZEN (EC 3.1.1.B12). Moreover, indirect screening has identified additional structurally distinct enzymes with potential ZEN-degrading activity. In the present study, we evaluated two such enzymes-a Streptomyces exfoliatus lipase (SeLIP) and a Thermobifida cellulosilytica cutinase (TcCUT)-and compared t

Adenine phosphate-Cu nanozyme with multienzyme mimicking activity for efficient degrading phenolic compounds and detection of hydrogen peroxide, epinephrine and glutathione.

BACKGROUND: With the development of nanotechnology, various nanomaterials with enzyme-like activity (nanozymes) have been reported. Due to their superior properties, nanozymes have shown important application potential in the fields of bioanalysis, disease detection, and environmental remediation. However, only a few nanomaterials with multi-enzyme mimicry activity have been reported. In this study, a novel multienzyme mimic was synthesized through a simple and rapid preparation protocol by coordinating copper ions with N3, N6 (amino), N7, and N9 on adenine phosphate. RESULTS: The prepared ad

Differential expression of enzymes in thymidylate biosynthesis in zebrafish at different developmental stages: implications for dtymk mutation-caused neurodegenerative disorders.

BACKGROUND: Deoxythymidine triphosphate (dTTP) is an essential building block of DNA, and defects in enzymes involved in dTTP synthesis cause neurodegenerative disorders. For instance, mutations in DTYMK, the gene coding for thymidylate kinase (TMPK), cause severe microcephaly in human. However, the mechanism behind this is not well-understood. Here we used the zebrafish model and studied (i) TMPK, an enzyme required for both the de novo and the salvage pathways of dTTP synthesis, and (ii) thymidine kinases (TK) of the salvage pathway in order to understand their role in neuropathology. Peptide-Based Oxidoreductase Mimics: Activity Origins, Design Principles, Construction Strategies, and Future Trends.

Oxidoreductases are basic enzymes that catalyze redox reactions within cells. Naturally occurring oxidoreductases have high catalytic activity and substrate specificity, playing critical roles in bioenergy metabolism, the food industry, and biotechnology. However, limitations, such as high cost, low stability, and poor tolerance to reaction conditions, restrict their practical applications. Developing enzyme mimics with natural oxidoreductase-like activity represents a promising alternative. This review summarizes the activity origins and activity sites of peptide-based oxidoreductase mimics,

Expression and partial biochemical characterization of a recombinant serine protease from Bothrops pauloensis snake venom.

Snake venom serine proteases (SVSPs) are enzymes capable of interfering at several points of hemostasis. Some serine proteases present thrombin-like activity, which makes them targets for the development of therapeutics agents in the treatment of many hemostatic disorders. In this study, a recombinant thrombin-like serine protease, denominated rBpSP-II, was obtained from cDNA of the Bothrops pauloensis venom gland and was characterized enzymatically and biochemically. The enzyme rBpSP-II showed clotting activity on bovine plasma and proteolytic activity on fibrinogen, cleaving exclusively the

Metabolic-epigenetic crosstalk in macrophage activation.

Epigenetic enzymes are emerging as crucial controllers of macrophages, innate immune cells that determine the outcome of many inflammatory diseases. Recent studies demonstrate that the activity of particular chromatin-modifying enzymes is regulated by the availability of specific metabolites like acetyl-coenzyme A, S-adenosylmethionine, α-ketoglutarate, nicotinamide adenine dinucleotide and polyamines. In this way chromatin-modifying enzymes could sense the macrophage's metabolic status and translate this into gene expression and phenotypic changes. Importantly, distinct macrophage activation
