New learning and memory related pathways among the hippocampus, the amygdala and the ventromedial region of the striatum in rats.

BACKGROUND: The hippocampus, central amygdaloid nucleus and the ventromedial region (marginal division) of the striatum have been reported to be involved in the mechanism of learning and memory. This study aimed elucidating anatomical and functional connections among these brain areas during learning and memory. RESULTS: In the first part of this study, the c-Fos protein was used to explore functional connections among these structures. Chemical stimulation of either hippocampus or central amygdaloid nucleus results in dense expression of c-Fos protein in nuclei of neurons in the marginal div

[Folate, vitamin B12 and human health].

During the past decade the role of folate and vitamin B12 in human nutrition have been under constant re-examination. Basic knowledge on the metabolism and interactions between these essential nutrients has expanded and multiple complexities have been unraveled. These micronutrients have shared functions and intertwined metabolic pathways that define the size of the "methyl donor" pool utilized in multiple metabolic pathways; these include DNA methylation and synthesis of nucleic acids. In Chile, folate deficiency is virtually nonexistent, while vitamin B12 deficiency affects approximately 8.5

Axonal regeneration induced by blockade of glial inhibitors coupled with activation of intrinsic neuronal growth pathways.

Several pharmacological approaches to promote neural repair and recovery after CNS injury have been identified. Blockade of either astrocyte-derived chondroitin sulfate proteoglycans (CSPGs) or oligodendrocyte-derived NogoReceptor (NgR1) ligands reduces extrinsic inhibition of axonal growth, though combined blockade of these distinct pathways has not been tested. The intrinsic growth potential of adult mammalian neurons can be promoted by several pathways, including pre-conditioning injury for dorsal root ganglion (DRG) neurons and macrophage activation for retinal ganglion cells (RGCs). Singl

Altered cognitive performance is associated with auditory cortex functional connectivity in patients with unilateral sensorineural hearing loss.

Sensorineural hearing loss (SNHL) has been suggested to be closely associated with cognitive decline, yet the underlying neural mechanisms remain largely unclear. This study sought to explore the potential association between cognitive function and auditory cortex functional connectivity in patients with unilateral sensorineural hearing loss (USNHL) and investigate the possible neural mechanisms underlying subtle cognitive decline. Sixty patients with moderate-to-severe USNHL (disease duration ≥3 years; 30 with right-sided USNHL and 30 with left-sided USNHL) and 30 age-matched normal-hearing c

The role of calcium-dependent gene expression in autism spectrum disorders: lessons from MeCP2, Ube3a and beyond.

During the last decade, autism spectrum disorders (ASD) have become the center of attention where several branches of modern biology unexpectedly meet, such as neural development, molecular biology, epigenetics, neurophysiology and psychiatry. This review will focus on the molecular mechanism by which calcium-dependent gene expression regulates brain development and how ASD may occur if this process is compromised. Specifically, the studies of the calcium-dependent transcriptional repressor MeCP2 gave us much insight about how abnormal development may lead to ASD. Most recently, studies about

Epidermal growth factor receptor activation: an upstream signal for transition of quiescent astrocytes into reactive astrocytes after neural injury.

Modulating the behaviors of reactive astrocytes is a potential therapeutic strategy for neurodegenerative diseases. We found that upregulation and activation of the epidermal growth factor receptor (EGFR) occur in astrocytes after different injuries in optic nerves in vivo. Activation of EGFR regulates genes and cellular processes representing most major markers of reactive astrocytes and genes related with glaucomatous optic neuropathy and other neural disorders. These results suggest that activation of EGFR is a common, regulatory pathway that triggers quiescent astrocytes into reactive astr

Gut virome plays an extended role with bacteriome in neurological health and disease.

The gut-brain axis (GBA) is a complex two-way communication system that links the gastrointestinal tract and the central nervous system (CNS) through neural, immune, hormonal, and microbial pathways. The microbiota-gut-brain axis (MGBA), a more specific concept, focuses on how gut microorganisms, including bacteria, viruses, and other microbes, modulate this communication and influence neurological health. This comprehensive review examines the intricate mechanisms through which gut microorganisms modulate neural function and contribute to neurological health and disease pathogenesis. The gut

Transplacental Transfer of Oxytocin and Its Impact on Neonatal Cord Blood and In Vitro Retinal Cell Activity.

The development of fetal organs can be impacted by systemic changes in maternal circulation, with the placenta playing a pivotal role in maintaining pregnancy homeostasis and nutrient exchange. In clinical obstetrics, oxytocin (OXT) is commonly used to induce labor. To explore the potential role of OXT in the placental homeostasis of OXT, we compared OXT levels in neonatal cord blood among neonates (23-42 weeks gestation) whose mothers either received prenatal OXT or experienced spontaneous labor. Our previous research revealed that the oxytocin receptor (OXTR), essential in forming the blood-

Autonomic Response Latency Revisited: Assessment of Repeatability in Healthy Subjects.

BACKGROUND: Autonomic function assessment provides valuable information regarding the status of the autonomic nervous system. The time lag between the onset of orthostasis and the peak/nadir of heart rate response is a surrogate of the integrity of underlying autonomic neural pathways. Autonomic response latency (ARL) is a relatively novel yet underreported parameter in this context. Test-retest repeatability of this parameter has not been evaluated previously. MATERIALS AND METHODS: We recruited 31 healthy adults (17 males and 14 females; 29.00 ± 5.44 years) and subjected them to postural ch…

A focus on the multiple interfaces between action and perception and their neural correlates.

Successful behaviour relies on the appropriate interplay between action and perception. The well-established dorsal and ventral stream theories depicted two distinct functional pathways for the processes of action and perception, respectively. In physiological conditions, the two pathways closely cooperate in order to produce successful adaptive behaviour. As the coupling between perception and action exists, this requires an interface that is responsible for a common reading of the two functions. Several studies have proposed different types of perception and action interfaces, suggesting the

Conserved and lineage-specific mechanisms drive chromatophore differentiation in reptiles.

The mechanisms by which novel differentiation pathways evolve to produce new cell types are still not fully understood. Chromatophores, the pigmented cells in the skin, offer an ideal paradigm because each type independently develops from neural crest cells to produce a distinct colour using well-characterised biosynthetic pathways. Here we show, using single-cell gene expression analyses, that canonical chromatophores develop in the embryonic skin of corn snakes and bearded dragon lizards. Yet, we identify previously undescribed chromatophore subtypes in the bearded dragon. These populations

Distinct perceptual and conceptual representations of natural actions along the lateral and dorsal visual streams.

Actions are the building blocks of our dynamic visual world, yet the neural computations supporting action perception are not well understood. How does perceptual and conceptual information unfold in the brain when we observe what others are doing? We collected EEG and fMRI data while participants viewed short videos and sentences depicting naturalistic actions. Using representational similarity analysis, we found distinct conceptual representations along the ventral, dorsal, and lateral pathways, with the target of actions specifically encoded in lateral occipitotemporal cortex (LOTC) and pos

Gut Microbiota and Metabolites: Orchestrating Depression Pathogenesis Through the Microbiota-Gut-Brain Axis's Neural, Immune, and Metabolic Routes.

Depression (major depressive disorder, MDD) is a globally prevalent, highly disabling, and complex mental disorder whose pathogenesis has not been fully elucidated. In recent years, the role of the gut microbiota in depression via the "microbiota-gut-brain axis" (MGB axis) has attracted increasing attention. A large body of evidence indicates that the gut microbiota and its metabolites can engage in bidirectional communication with the central nervous system through three core pathways-neural, immune, and metabolic-thereby profoundly influencing the onset and progression of depression. This ar

Genome-wide association meta-analyses over one million individuals identify 54 loci associated with urinary incontinence and its subtypes.

Urinary incontinence (UI) markedly reduces quality of life, and its genetic basis remains to be elucidated. Here, we performed GWAS meta-analyses of four UI phenotypes, using hospital-based data from 1,045,436 individuals from the Trøndelag Health Study (HUNT), UK Biobank, FinnGen, and the Michigan Genomics Initiative, and self-reported UI from 56,957 females from HUNT and the Nurses' Health Study. We identified 54 (52 novels) genetic loci associated with at least one UI phenotype. Analyses revealed tissue-specific involvement, with connective and muscular tissues for stress UI, and neural tis

Spatiotemporally Precise Chemical Neuromodulation through MOF-Mediated Near-Infrared Control of Endogenous Signaling.

Chemical neuromodulation offers a physiologically relevant approach to manipulate neural functions, yet current methods suffer from poor spatiotemporal precision, cargo instability, or requirements for genetic modification. Here, we report a near-infrared (NIR) light-controlled platform for on-demand nitric oxide (NO) delivery to neurons, using upconversion nanoparticles (UCNPs) coated with metal-organic framework shells covalently loaded with photolabile NO donors (UCNP@MOF-NO). The core-shell architecture exploits NIR-to-UV upconversion to photolytically release NO with spatiotemporal precis

Unravelling the brain resilience following stroke: From injury to rewiring of the brain through pathway activation, drug targets, and therapeutic interventions.

Synaptic plasticity is a neuron's intrinsic ability to make new connections throughout life. The morphology and function of synapses are highly susceptible to any pathological condition. Ischemic stroke is a cerebrovascular event that affects various brain regions, resulting in the loss of neural networks. Stroke can alter both structural and functional plasticity of synapses, leading to long-term functional disability. Upon ischemic insult, numerous glutamate-mediated synaptic destruction pathways and glial-mediated phagocytic activity are triggered, resulting in excessive synapse loss, alter

Epigenetic dysregulation and the etiology of spina bifida.

Spina bifida is a complex and multifactorial congenital defect driven by both genetic and environmental factors. As such, epigenetic studies of spina bifida present an opportunity to study the joint contribution of both genes and the environment in the development of this disorder. This review focuses specifically on epigenetic research that may help us to understand the ways in which dysregulation of the epigenome and downstream cellular processes can confer increased risk of spina bifida. To do so, we discuss the epigenetic regulation of genes linked to spina bifida risk among children born
