The ganglion cell response to optic nerve injury in the cat: differential responses revealed by neurofibrillar staining.

The early responses of cat retinal ganglion cells to axotomy have been examined using neurofibrillar and Nissl-stained wholemounts. We were interested to learn whether the enhanced neurofilament expression, seen in a number of neuronal systems, was also present in different neuronal populations of the cat retina and could be used to study the distribution of these cells. We found that beta ganglion cells degenerate very rapidly after axotomy with the nuclei becoming pyknotic within a few days. Few beta cells showed increased neurofibrillar staining of the dendrites. The cell body degenerated p…

Forebrain damage following prenatal exposure to low-dose X-irradiation.

Exposure of fetal rats to X-irradiation on gestational day 15 resulted postnatally in dose-related effects on body weight, growth of forebrain structures, and branching of dendrites of caudate neurons. Rats were followed for 4 months postnatally after 125, 75, 50, or 25 R whole-body irradiation to the dam. Significant decreases in body weight were present at birth after the three high doses and continued as long as 4 months after 125 or 75 R. Decreased thickness of the cerebral cortex and decreased area of the caudate nucleus were also seen. Cortical thickness was reduced by 125 R to half the …

The development of the olfactory mucosa in the mouse: electron microscopy.

The development of the olfactory epithelium from the 10th day of gestation of postnatal life has been examined electron miscroscopically in the mouse. At 10 days' gestation the epithelium is already differentiated into dark and pale cells, the former representing embryonic stem cells and the latter the developing receptors. Axons are also visible at this stage. At 11 days the first signs of dendrite formation appear, and at 12 days spheroidal terminal swellings containing numerous microtubules are present at the apices of receptor dendrites. Centriole clusters also appear in the receptor cell …

Subcellular interactions of neuropeptide Y and corticotropin-releasing factor in the central nucleus of the amygdala in the mouse.

Neuropeptide Y (NPY) is ubiquitously distributed throughout the central nervous system. Recognized as a mediator of stress resilience, NPY h … lver labeling for CRF showed that NPY-labeled axon terminals (NPY-t) form synapses with CRF-labeled dendrites (CRF-d). Semi-quantitative analysis revealed that 163 of NPY-t directly target CRF-d. In addition, approximately 85% of NPY-t form symmetric synapses with CRF-d while approximately 1% form asymmetric synapses. These findings provide the first ultras …

Interfacial self-healing engineered bifunctional quasi-solid electrolyte for high-performance lithium-sulfur batteries.

Given their low cost and high energy density, lithium‑sulfur batteries (LSBs) are an appealing alternative for next-generation battery systems. However, the shuttle effect of polysulfides and the growth of lithium dendrites during battery operation severely hinder the practical application of LSBs. Here, we propose an interfacial self-healing strategy to prepare the bifunctional Polyvinylidene fluoride-hexafluoropropylene (PVDF-HFP)/Graphene oxide (GO)/Copper benzene-1,3,5-tricarboxylate (HKUST-1)/Borate bond Quasi-Solid-State Electrolyte (PHGKB QSSE), which significantly improves the electroc…

The Role of RNA Binding Proteins for Local mRNA Translation: Implications in Neurological Disorders.

As neurons are one of the most highly polarized cells in our body, they require sophisticated cellular mechanisms to maintain protein homeostasis in their subcellular compartments such as axons and dendrites. When neuronal protein homeostasis is disturbed due to genetic mutations or deletions, this often results in degeneration of neurons leading to devastating outcome such as spinal muscular atrophy (SMA), amyotrophic lateral sclerosis (ALS), and fragile X syndrome (FXS). Ribonucleoprotein (RNP) complexes are macromolecular complexes composed of RNA binding proteins (RBPs) and their target RN…

Kymolyzer, a Semi-Autonomous Kymography Tool to Analyze Intracellular Motility.

The movement of intracellular cargo, such as transcripts, proteins, and organelles, is fundamental to cellular function. Neurons, due to their long axons and dendrites, are particularly dependent on proper intracellular trafficking and vulnerable to defects in the movement of intracellular cargo that are noted in neurodegenerative and neurodevelopmental disorders. Accurate quantification of intracellular transport is therefore needed for studying the mechanisms of cargo trafficking, the influence of mutations, and the effects of potentially therapeutic pharmaceuticals. In this article, we intr…

Melanin accumulation was more in the IGH patients as compared to the controls. Alterations of striatal indirect pathway neurons precede motor deficits in two mouse models of Huntington's disease.

Alterations that specifically occurred at symptomatic ages included increased calcium release by back-propagating action potentials in proximal dendrites, due to enhanced engagemen …

Ultrastructural relationships between terminals immunoreactive for enkephalin, GABA, or both transmitters in the rat ventral tegmental area.

Enk-labeled terminals formed either symmetric or asymmetric synapses on small or large unlabeled dendrites. [Morphologic characteristics of locus coeruleus neurons after selective chemical damage to the catecholaminergic system of the brain].

The initial part of the noradrenergic cerebral system--the locus caeruleus neurons--has been studied light and electron microscopically at various time after injection of 6- hydrodopamine (6-OHDA), a substance possessing a selective neurotoxic effect on the catecholaminergic mediatory system. Intracisternal injection of 6-OHDA (300 mcg) produces a number of reactive rearrangements in the neurons and large dendrites. Nevertheless, the death of the neural cells is not observed even by the 36th day.

Modified Dendritic cell-based T-cell expansion protocol and single-cell multi-omics allow for the selection of the most expanded and in vitro-effective clonotype via profiling of thousands of MAGE-A3-specific T-cells.

INTRODUCTION: Adoptive cell therapy using TCR-engineered T-cells is one of the most effective strategies against tumor cells. The TCR T-cell approach has been well tested against a variety of blood neoplasms but is yet to be deeply tested against solid tumors. Among solid tumors, cancer-testis antigens are the most prominent targets for tumor-specific therapy, as they are usually found on cells that lie behind blood-tissue barriers. METHODS: We have employed a novel efficient protocol for MAGE-A3-specific T-cell clonal expansion, performed single-cell multi-omic analysis of the expanded T-cel

Case Report: In Situ Vaccination by Autologous CD16+ Dendritic Cells and Anti-PD-L 1 Antibody Synergized With Radiotherapy To Boost T Cells-Mediated Antitumor Efficacy In A Psoriatic Patient With Cutaneous Squamous Cell Carcinoma.

The combination of radiotherapy and immunotherapy improves the survival rate of patients with malignancies developed through escape from T-cell-mediated immune surveillance. Immune checkpoint inhibitors, such as anti-programmed cell death protein-ligand 1 (anti-PD-L1) antibody, are used to rescue exhausted T cells. Simultaneously, dendritic cells (DCs) which are antigen-presenting cells that can initiate T-cell activation, are used to induce a tumor-specific immune response. However, the synergistic antitumor efficacy of the aforementioned combinational immunotherapy with intratumoral injectio
