Efficient vitamin A production in Lipomyces starkeyi through metabolic engineering of the β-carotene and retinoid pathways.

BACKGROUND: Vitamin A, an essential micronutrient, is critical for vision, immune function, and cellular differentiation. Traditional vitamin A production methods, primarily based on chemical synthesis, pose significant environmental challenges. Microbial fermentation offers a sustainable alternative, but microbial production of vitamin A has yet to match chemical synthesis in terms of yield and cost-effectiveness. Oleaginous yeasts, such as Lipomyces starkeyi, which can synthesize high levels of acetyl-CoA and lipids, represent promising platforms for producing high-value lipophilic compounds

Thermophysical and Rheological Characteristics of CO2 Hydrate Slurries for Cold Thermal Energy Storage Applications and Engineering Perspectives.

Carbon dioxide (CO2) hydrate slurries have emerged as promising candidates for cold thermal energy storage (CTES) and refrigeration systems due to their high latent heat, controllable flow behavior, and environmentally friendly nature. These slurries are formed by dispersing solid CO2 hydrate particles in a liquid phase, forming a multiphase system with tunable thermophysical and rheological properties. The performance of these slurries is dependent on nucleation kinetics, particle sizes and their distribution, solid content, and thermal transport characteristics under flow conditions. This re

The fitness landscape of a form II rubisco in a photosynthetic bacterium guides engineering of oxygen tolerance.

Ribulose-1,5-bisphosphate carboxylase/oxygenase (rubisco) is an important but challenging protein engineering target. Fast and selective rubiscos could enhance photosynthesis in plants and accelerate biobased production processes. To facilitate engineering of rubisco, we applied an in vivo screen that couples rubisco activity to growth rate of the photoautotrophic cyanobacterium Synechocystis sp. PCC 6803. We screened a barcoded mutagenesis library of the form II rubisco from Gallionella sp. containing 15,000 single-site and multi-site variants. Exchanges in loop 6 near the active site, at the…

Porous Glycine-Modified LaFeO3 for Highly Selective Ethanol Sensing: Band Gap Engineering Driven by Organic-Inorganic Hybridization.

The demand for highly selective ethanol sensors for reliable environmental and industrial monitoring remains a critical challenge. Herein, we demonstrate a facile hydrothermal strategy to synthesize porous glycine-LaFeO3 (Gly@LFO) composites, where organic-inorganic hybridization drives significant band gap narrowing. This electronic restructuring, combined with a porous morphology (optimal at a 1:1 Gly-to-LFO ratio, sample L3), creates abundant active sites and facilitates rapid gas diffusion as confirmed by SEM, EDS, and XPS analysis. First-principles calculations elucidate the underlying me

Rational Engineering of Copper Ion-Coordinated Molecularly Imprinted Polymers for Synergistic Enhancement of Protein Recognition and Separation.

By creating tailor-made binding sites, molecularly imprinted polymers (MIPs) function as synthetic antibodies, offering comparable specificity with enhanced stability and lower cost. While molecular imprinting technology has achieved significant success with small molecules, its application to macromolecules such as proteins remains challenging. This is primarily due to the common use of aqueous solutions for protein imprinting, where key interactions like hydrogen bonding and electrostatic forces are significantly weakened. To address these limitations, this study reports the rational design

Graphene-Skinned Fiber with Fine-Tunable Electrical Resistance via Radical and Substrate Engineering for Electromagnetic-Thermal Fabric.

This work demonstrates a radical-manipulation strategy for synthesizing graphene (Gr)-skinned SiO2 fabric via low-pressure chemical vapor deposition using methanol precursor. Controlled pyrolysis at high temperature regulated C1/C2/C6 radical ratios, enabling microstructure engineering. Substrate effects governed bilayer evolution. SiO2 imposed lower adsorption energy of C1 and higher diffusion barriers of radical compared to Gr, promoting edge defects in subsurface G1-type Gr layers, whereas reduced substrate constraints facilitated low-defect G2-type Gr growth on top of G1-type Gr. Synergist…

Engineering the Enzyme-Support Interface: Arginine-Directed Anchoring of Peroxygenase on a β-Ketoester Covalent Organic Framework.

Controlling the conformation and orientation of enzymes at solid interfaces is a central challenge in the design of functional biohybrid materials, and the outcome depends sensitively on the chemistry and geometry of the enzyme-support interface. Here, a β-ketoester-functionalized keto-enamine covalent organic framework (COF) is developed as a structurally defined support for arginine-directed immobilization of unspecific peroxygenase (UPO). The crystalline periodicity and post-synthetic addressability of the COF lattice enable precise placement of anchoring groups at the enzyme-support interf

Safeguarding VSMC Contractile Phenotype With In Situ circRNA-mediated Endothelial Olaratumab Engineering to Prevent Vascular Graft Stenosis.

Small-diameter vascular grafts (SDVGs) often fail due to restenosis driven by endothelial cell (EC)-derived PDGF-BB, which shifts vascular smooth muscle cells (VSMCs) toward a synthetic phenotype. Despite mechanistic insights, durable, localized, and cell-specific control of this crosstalk remains elusive. Here, we developed a circRNA-based in situ antibody engineering strategy to functionalize SDVGs, reprogramming ECs into local biofactories that secrete Olaratumab (Ola), a PDGFR-α-neutralizing antibody, to precisely modulate EC-VSMC signaling. In vitro, in situ Ola engineering reversed PDGF-…
