Airway inflammation in asthma and perennial allergic rhinitis. Relationship with nonspecific bronchial responsiveness and maximal airway narrowing.

BACKGROUND: Eosinophilic airway inflammation is the hallmark of asthma, but it has also been reported in other conditions such as allergic rhinitis. We have tested whether the analysis of cells and chemicals in sputum can distinguish between patients with mild allergic asthma, those with allergic rhinitis, and healthy controls. The relationship between inflammation markers in sputum and nonspecific bronchial hyperresponsiveness to methacholine (BHR) (PD20 and maximal response plateau [MRP] values) was also evaluated. METHODS: We selected 31 mild asthmatics and 15 rhinitis patients sensitized

Pharmacological stimuli in asthma/urticaria.

BACKGROUND: The use of opioids as analgesics is becoming increasingly widespread, which may have repercussions in patients with urticaria or asthma, as these agents frequently cause adverse reactions. MATERIAL AND METHODS: We present three patients who developed allergic reactions after receiving codeine: two patients who developed acute urticaria, and a third asthmatic patient receiving specific immunotherapy who developed bronchospasm. Skin prick-testing (SPT) and intradermal reaction (IDR) tests with various opioids were performed, followed by controlled oral challenge. Prick tests and IDR

Candida albicans and atopic dermatitis.

The role of sensitization and exposure to Candida albicans in atopic dermatitis (AD) was studied with skin-prick tests, yeast cultures and immunoblotting in 156 young adults with AD attending the Department of Dermatology, University of Turku, during 1983-89. Eighteen patients with allergic rhinitis without eczema and 39 non-atopics were included as controls. Parameters associated with severe AD were simultaneous anti-C. albicans IgE and saprophytic C. albicans growth. A statistically significant correlation between C. albicans sensitization (specific IgE antibodies) and AD symptoms was observ

Lung-Targeted Delivery of Nebulized α‑Asaronol Attenuates Airway Inflammation and Hyperresponsiveness in Murine Asthma Models.

Acorus tatarinowii, a traditional Chinese medicinal herb, is used clinically to treat neurological and respiratory disorders. Our previous work showed that α-asaronol (α-AOL), a low-toxicity natural constituent of A. tatarinowii, exerts robust antiepileptic effects through γ-aminobutyric acid (GABA)-ergic modulation. Given the critical role of the peripheral GABAergic system in asthma pathogenesis and inflammation regulation, the therapeutic potential of α-AOL for asthma warrants investigation. This study evaluated the efficacy, safety, and pulmonary targeting of both nebulized and oral α-AOL.

The toxicity of neodymium substances to soil invertebrates in a boreal soil, and the impacts of soil type, aging, and leaching on metal bioavailability and toxicity.

A clear gap in ecotoxicological data was identified regarding the impacts of the technologically critical lanthanide neodymium (Nd) on soil invertebrates in boreal soil. To address this gap, two inorganic metal salts (NdCl3 and Nd2(SO4)3), an organometallic compound Nd-2-ethylhexanoate (Nd-2-EHA) and its organic component 2-ethylhexanoic acid (2-EHA) were examined. Toxicity exposures were conducted using the earthworm Dendrodrilus rubidus, the collembolan Proisotoma minuta, and the oribatid mite Oppia nitens, in two Canadian boreal soil horizons (i.e., organic and mineral), using standardized

Luteolin Attenuates Asthma Via Inhibiting ALOX15 Mediated Bronchial Epithelial Cell Ferroptosis.

Ferroptosis, an iron-dependent form of cell death driven by lipid peroxidation, plays a critical role in the progression of asthma, yet its therapeutic modulation remains underexplored. Luteolin (Lut), a natural flavonoid known for its anti-inflammatory properties, presents a potential candidate for targeting ferroptosis in asthma. However, the precise mechanisms underlying its therapeutic effects are unclear. This study used a combination of bioinformatics, network pharmacology, molecular docking, and in vitro and in vivo experiments to investigate the role of luteolin in the treatment of fer

Cannabichromeorcin targets cathepsin L to alleviate oxidative stress-Driven airway inflammation in severe asthma.

BACKGROUND: Severe asthma (SA) is characterized by corticosteroid insensitivity, persistent airway inflammation, and excessive oxidative stress, underscoring an urgent unmet therapeutic need. Meroterpenoids derived from Rhododendron capitatum Maxim have demonstrated potent anti-inflammatory properties; however, their therapeutic potential in SA has not been elucidated. METHODS: A meroterpenoid-rich fraction was isolated from R. capitatum Maxim, and cannabichromeorcin (CBCO) was identified as its most abundant constituent. The therapeutic effects of CBCO were evaluated in murine models of SA i

Glehnia littoralis and imperatorin attenuate allergic airway inflammation via mast cell stabilization and transcriptome-defined suppression of FcεRI/TLR-MAPK/NF-κB signaling.

BACKGROUND: Allergic airway inflammation is driven by mast cell activation and IgE-FcεRI signaling. Current pharmacological treatments often present limited efficacy or long-term adverse effects. Glehnia littoralis Fr. Schmidt ex Miq. (GL) is a traditional medicinal herb used for respiratory ailments, but its anti-allergic mechanisms remain largely undefined. PURPOSE: We aimed to evaluate the anti-allergic potential of GL and its bioactive compound, imperatorin, specifically investigating their ability to stabilize mast cells and modulate inflammatory signaling. STUDY DESIGN: This study inte

Unveiling Interleukin-40: A Novel Regulator of Macrophage and B Cell Function in Allergic Asthma.

Allergic asthma is characterized by chronic airway inflammation and heightened type 2 immune responses. Although inhaled corticosteroids are the mainstay of therapy, a subset of patients exhibits suboptimal responses, underscoring the need for new therapeutic targets. In this study, we investigated the role of novel B cell-related interleukin-40 (IL-40) in allergic asthma using patient samples and a house dust mite (HDM)-induced mouse model. Through transcriptomic and immunological profiling, our findings revealed that IL-40 expression was significantly upregulated in both patients with allerg

BRAP Homologue Deficiency Exacerbates Airway Inflammation in Mouse Models of Experimental Asthma-Like Disease.

Bronchial epithelial cells (BECs) play a critical role in asthma pathogenesis through structural and functional alterations that contribute to disease severity. Bombesin receptor-activated protein (BRAP), encoded by the human C6orf89 gene, shares 83% identity with its mouse homologue encoded by BC004004. Both proteins have been identified in bronchial epithelial cells, and our previous studies found that BRAP overexpression alters the biological behaviour of cultured human bronchial epithelial cells, suggesting a potential role in regulating immune homeostasis in the respiratory tract. In this

Characterization of Human Group 9 Innate Lymphoid Cells in Response to Allergen Immunotherapy in Patients With Allergic Rhinitis.

BACKGROUND: Innate lymphoid cells (ILCs) are classically divided into three groups: ILC1, ILC2, and ILC3 to reflect their functional analogy to Th1, Th2, and Th17. There is also an IL-9 single-producing T cell subset, namely the Th9 cell, which plays a dominant role in the onset of allergic diseases compared with traditional Th2 cells. Although a corresponding cell subset of ILCs to different Th cell subsets exists, so far the counterpart of Th9 cells in ILCs has not been reported. OBJECTIVE: In this study, we aimed to report the existence of group 9 innate lymphoid cells (ILC9s) and characte

Adaptive preprocessing and Cascaded Canny Edge Segmentation for cassava disease identification using HyperCapsInception-ResNet-V2-CNN.

INTRODUCTION: Cassava is one of the most widely cultivated crops worldwide, renowned for its rich natural ingredients and numerous nutritional benefits. However, the complex interdependencies among its features often pose challenges in image restoration and segmentation, particularly when identifying disease regions. In previous work, this manifested as higher false positives and misidentification of non-relevant areas, leading to a decline in precision and accuracy. METHODS: To address these issues, this study proposed an efficient artificial intelligence-powered image analysis system that l

Lung proinflammatory microRNA and cytokine expression in a mouse model of allergic inflammation: role of sex chromosome complement and gonadal hormones.

Epigenetic alterations such as dysregulation of miRNAs have been reported to play important roles in interactions between genetic and environmental factors. In this study, we tested the hypothesis that induction of lung inflammation by inhaled allergens triggers a sex-specific miRNA regulation that is dependent on chromosome complement and hormonal milieu. We challenged the four core genotypes (FCGs) model through intranasal sensitization with a house dust mite (HDM) solution (or PBS as a control) for 5 wk. The FCG model allows four combinations of gonads and sex chromosomes: 1) XX mice with o
