D-beta-hydroxybutyrate protects dopaminergic SH-SY5Y cells in a rotenone model of Parkinson's disease.

It has been postulated that the pathogenesis of Parkinson's disease (PD) is associated with mitochondrial dysfunction. Rotenone, an inhibitor of mitochondrial complex I, provides models of PD both in vivo and in vitro. We investigated the neuroprotective effect of D-beta-hydroxybutyrate (bHB), a ketone body, against rotenone toxicity by using SH-SY5Y dopaminergic neuroblastoma cells. SH-SY5Y cells, differentiated by all-trans-retinoic acid, were exposed to rotenone at concentrations ranging from 0 to 1,000 nM. We evaluated cellular oxidation reduction by the alamarBlue assay, viability by lact

Somatic and occult germ-line mutations in SDHD, a mitochondrial complex II gene, in nonfamilial pheochromocytoma.

Most pheochromocytomas are sporadic but about 10% are though to be hereditary. Although the etiology of most inherited pheochromocytoma is well known, little is known about the etiology of the more common sporadic tumor. Recently, germ-line mutations of SDHD, a mitochondria complex II gene, were found in patients with hereditary paraganglioma. We sought to determine whether SDHD plays a role in the development of sporadic pheochromocytomas and performed a mutation and deletion analysis of SDHD. Among 18 samples, we identified 4 heterozygous sequence variants (3 germ-line, 1 somatic). One germ-

Does the respiratory rate in sea urchin embryos increase during early development without proliferation of mitochondria?

During early development of the sea urchin, the respiratory rate, enhanced upon fertilization, is maintained up to hatching (pre-hatching period) and then gradually increases to a maximum at the gastrula stage (post-gastrula period). Except for a short duration after fertilization, respiration in embryos is strongly inhibited by CN- and antimycin A. During the whole span of early development, the amounts of proteins, cytochromes and the specific activities of cytochrome c oxidase and reduced nicotinamide adenine dinucleotide (NADH) cytochrome c reductase in mitochondria are practically the sam

Phytogenic supplementation mitigates necrotic enteritis in broilers through modulation of gut microbiota, short-chain fatty acids, and nutrient-related gene expression.

With the withdrawal of antibiotic growth promoters, phytogenic compounds have gained attention as potential alternatives for enhancing gut health and modulating the intestinal microbiota. This study investigated the effects of a phytogenic blend (PHB) containing thymol, oleic acid, capric acid, and refined sunflower fat on cecal short-chain fatty acids (SCFA), jejunal gene expression, and cecal microbiota composition in broiler chickens under necrotic enteritis (NE) challenge. A total of 450 d-old mixed-sex Cobb 500 broiler chicks were randomly allocated to three treatments: non-challenged con

Metabolic reprogramming of tomato roots during rhizobacteria-mediated defense against Erwinia persicina: modulation by gold nanoparticle conjugation.

Rhizobacteria-induced systemic resistance (ISR) is an established strategy for enhancing plant tolerance to biotic stress, yet its metabolic consequences under nanoparticle-assisted delivery remain poorly understood. Here, we investigated metabolic reprogramming in tomato roots (Solanum lycopersicum L.) challenged with the pathogen Erwinia persicina following treatment with PGPR strain Stenotrophomonas rhizophila (Sr) applied either alone or conjugated to phycosynthesized gold nanoparticles using Caulerpa sertularioides. Bionanogold synthesis was confirmed by UV-Visible surface plasmon resonan

Diagnosis and treatment of multisystem amyloidosis associated with SGPL1 mutation: A case report and review of the literature.

RATIONALE: Amyloidosis is a rare, clinically heterogeneous disease, which makes its diagnosis difficult. The relationship between amyloidosis and gene mutations is insufficiently understood. We report a case of sphingosine-1-phosphate lyase 1 (SGPL1) mutation-related amyloidosis, and review the related literature. PATIENT CONCERNS: A 53-year-old man was admitted to our hospital with a 5-month history of renal dysfunction and abdominal distension, aggravated since 2 days. Five months ago, the patient was diagnosed with nephrotic syndrome, with liver dysfunction and abdominal distension. His sy

Carbon source-dependent metabolic states govern redox homeostasis and cofactor biosynthesis in Propionibacterium freudenreichii.

UNLABELLED: Microbial adaptation to fluctuating nutrient and oxygen conditions requires coordinated regulation of the metabolic network to maintain redox homeostasis within physicochemical and energetic constraints. While oxygen-dependent responses in Propionibacterium freudenreichii (PFR) have been characterized at the transcriptomic level, the role of carbon source in defining system-level metabolic states remains unclear. Here, we investigated carbon source-dependent metabolic reprogramming and cofactor biosynthesis in PFR strain DSM 20271T using label-free quantitative proteomics integrate

Human Milk and Infant Gut Microbiome in Association With Infant Fecal Metabolome and Child Blood Pressure.

IMPORTANCE: Infant milk feeding type (eg, human milk vs formula) and infant gut microbes have each been associated with differences in microbial metabolites and childhood blood pressure; however, evidence remains limited regarding how specific infant gut microbes, at a species or strain level, in combination with milk feeding type, shape microbial metabolites and blood pressure. OBJECTIVE: To investigate whether human milk feeding and infant gut microbes, including Bifidobacterium longum subsp infantis (B infantis) and other milk-degrading microbes, are associated with infant fecal metabolite

Relaxin-2 as a modifiable hormonal pathway in knee osteoarthritis: Dose dependent effects on chondrocyte phenotype.

BACKGROUND: Postmenopausal female individuals are disproportionately affected by knee osteoarthritis (KOA), experiencing earlier onset and more severe pathology compared to their male counterparts. Despite this clinical disparity, the molecular mechanisms underlying female-specific vulnerability remain poorly defined. OBJECTIVE: To evaluate the mechanistic role of relaxin-2 in postmenopausal KOA. DESIGN: This was a translational research study that evaluated relaxin-2 using an in vitro postmenopausal human KOA chondrocyte culture model and in silico network medicine simulation. SETTING: Res

Contemporary outcomes after 100 consecutive HeartMate 3 LVAD implantations: a single-centre cohort study from a Swiss tertiary centre.

The extra ocular (eye) muscles are one of the principal tissues involved in the autoimmune-mediated inflammation of Graves' ophthalmopathy (GO). Several eye muscle proteins are targeted by autoantibodies or sensitized T lymphocytes, or both, and include: G2s, which is now identified as the terminal 141 amino acids of the winged-helix transcription factor FOXP1, the flavoprotein (Fp) subunit of the mitochondrial enzyme succinate dehydrogenase, the so-called "64kDa protein", a non-tissue specific membrane protein called 1D and the calcium binding protein calsequestrin. Of these, antibodies again

AI-Driven Quantitative Metabolomics for Early and Precise HIE Diagnosis: Challenges and Solutions.

Adequate cerebral oxygenation is vital for neonatal survival and the establishment of a healthy brain function. A disruption in this critical balance, particularly during the perinatal period, can result in hypoxic-ischemic encephalopathy (HIE)a severe and potentially fatal form of neonatal brain injury caused by diminished oxygen and blood flow to the brain. HIE is a major contributor to neonatal morbidity and mortality worldwide, with an even greater burden in low-resource settings, where delays in diagnosis and limited access to timely intervention exacerbate long-term neurodevelopmental o

Prenatal exposure to microplastics and biomarkers of renal dysfunction in umbilical cord blood: Evidence from a birth cohort in China.

Microplastics (MPs) are ubiquitous environmental contaminants with potential to cross biological barriers, including the placenta. Limited data exist on the human fetal exposure burden and associated health effects. This study investigates the presence and composition of MPs in placental tissue and evaluates their individual and combined (mixture) associations with renal biomarkers in umbilical cord blood, providing novel insights into early-life nephrotoxicity. A total of 1350 pregnant women were recruited from Shengjing Hospital of China Medical University (Shenyang, China) between 2022 and

Mitochondrial damage is associated with an early immune response in inclusion body myositis.

Polymyositis with mitochondrial pathology (PM-Mito) was first identified in 1997 as a subtype of idiopathic inflammatory myopathy. Recent findings demonstrated significant molecular similarities between PM-Mito and inclusion body myositis (IBM), suggesting a trajectory from early to late IBM and prompting the inclusion of PM-Mito as an IBM precursor (early IBM) within the IBM spectrum. Both PM-Mito and IBM show mitochondrial abnormalities, suggesting that mitochondrial disturbance is a crucial element of IBM pathogenesis. The primary objective of this cross-sectional study was to characterize

Serum Metabolomic Production in Parn-Stage Multical Sclerosis:

Immp2l Enhances the Structure and Function of Mitochondrial Gpd2 Dehydrogenase.

'Inner mitochondrial membrane peptidase 2 like' (IMMP2L) is a nuclear-encoded mitochondrial peptidase that has been conserved through evolutionary history, as has its target enzyme, 'mitochondrial glycerol phosphate dehydrogenase 2' (GPD2). IMMP2L is known to cleave the mitochondrial transit peptide from GPD2 and another nuclear-encoded mitochondrial respiratory-related protein, cytochrome C1 (CYC1). However, it is not known whether IMMP2L peptidase activates or alters the activity or respiratory-related functions of GPD2 or CYC1. Previous investigations found compelling evidence of behavioura
