Platelet activating factor-induced shock and intestinal necrosis in the rat: role of endogenous platelet-activating factor and effect of saline infusion.

BACKGROUND AND METHODS: The mechanism of ischemic bowel necrosis induced by platelet-activating factor is unclear. Since intestinal hypoperfusion is observed after platelet-activating factor injection, we hypothesized that mesenteric vasoconstriction is the mechanism of bowel injury. The present study investigated the effects of saline infusion on platelet activating factor-induced bowel necrosis and its mechanism. Male Sprague-Dawley rats were divided into four groups: group A consisted of sham-operated rats; group B received platelet-activating factor (1.5 micrograms/kg iv); group C received

Genome reorganisation and expansion shape 3D genome architecture and define a distinct regulatory landscape in coleoid cephalopods.

How genomic changes translate into organismal novelties is often confounded by the multi-layered nature of genome architecture and the long evolutionary timescales over which molecular changes can accumulate. Coleoid cephalopods (squid, cuttlefish, and octopus) provide a unique system to study these processes due to a large-scale chromosomal rearrangement in the coleoid ancestor that gave rise to highly modified karyotypes, followed by lineage-specific repeat expansions, fusions, and translocations. How these events have shaped gene regulatory patterns underlying the evolution of coleoid innov

Necessary, Legendary and Detrimental Components of Human Colorectal Organoid Culture Medium: Raising Awareness to Reduce Experimental Bugs.

The creation of a specific culture medium for colorectal organoids in 2011 heralded a new era in human primary cultures by enabling the indefinite expansion of normal and pathological epithelial organoids. The original formula has been used ever since, with only minor, lab-specific modifications. The goal of culturing organoids from different tissues has relied on saving and propagating the pluripotent stem cell. The "magic bullet" and all its subsequent derivatives have pursued this goal. Consequently, agonist and antagonist signals are chronically activated in the organoid medium, forcing or

Ambient ionization mass spectrometry for rapid screening of illegal adulteration in traditional Chinese medicines: a review.

Illegal adulteration of traditional Chinese medicines has become a critical global safety concern, driven by the covert addition of synthetic pharmaceuticals such as phosphodiesterase type five inhibitors, anorectic agents, nonsteroidal anti-inflammatory drugs, corticosteroids, and sedatives. This review critically examines the rapid expansion of ambient ionization mass spectrometry technologies and evaluates their performance in detecting diverse adulterants across powders, pills, decoctions, creams, and botanical tissues. Techniques including desorption electrospray ionization, direct analys

Multiomics Reveals NDRG1-Driven Oral Squamous Cell Carcinoma Progression: Competitive Endogenous RNAs Regulation and Proangiogenic Microenvironment Remodelling.

Oral squamous cell carcinoma (OSCC) is characterized by high aggressiveness. This study aims to elucidate the role of NDRG1 in the evolutionary heterogeneity and spatial microenvironmental remodelling of OSCC. By integrating bulk transcriptomics, single-cell RNA sequencing (scRNA-seq), and spatial transcriptomics (ST), complemented by in vivo and in vitro functional assays, we systematically explored the regulatory logic of the MSTRG.47889/miR-1299/NDRG1 axis. The MSTRG.47889/miR-1299/NDRG1 ceRNA regulatory axis was identified and validated, demonstrating its significant role in promoting OSCC

Rewiring cellular immunotherapy: From CAR evolution to circuit innovations across diseases.

Chimeric antigen receptor (CAR)-based immunotherapy has evolved from early linear receptor designs into increasingly sophisticated biological signal processing systems. While conventional CAR-T cell therapies have achieved remarkable clinical success, particularly in hematologic malignancies, their broader application is constrained by limited scalability, manufacturing complexity, and treatment-related toxicities. These challenges have driven the expansion of CAR engineering beyond T cells to alternative immune cell types, including natural killer (NK) cells and macrophages, as well as the di

Synapse-like specializations at dopamine release sites orchestrate efficient and precise neuromodulatory signaling.

Neuromodulators are generally understood to act through expansive projections that broadcast signals relatively indiscriminately, in stark contrast to the precise, synapse-bound organization of fast neurotransmitters like glutamate and GABA. This dichotomy has left the local architecture of neuromodulatory release and receptor engagement largely obscure. Here, using dopamine as a model system, we reveal that neuromodulatory signaling can be guided by precise molecular specificity. We show that dopamine axons exhibit a tropism for neurons expressing D1 or D2 dopamine receptors. Single-bouton-re

Organoids: Current Applications and Future Directions.

Organoids are three-dimensional multicellular structures derived from stem cells or primary tissues that recapitulate key structural and functional features of native organs. Advances in stem cell biology, biomaterials, and bioengineering have established organoids as powerful platforms for studying human development and disease mechanisms, drug discovery, precision medicine, and regenerative medicine. However, current organoid models are limited by incomplete maturation, insufficient vascularization, inadequate immune and neural integration, and poor reproducibility, restricting their physiol

Ubiquitin-Dependent Protein Quality Control in Polyglutamine Diseases.

Polyglutamine diseases comprise a family of nine age-dependent disorders caused by CAG triplet-repeat expansions that produce misfolded proteins with elongated glutamine tracts. Although these diseases share proteotoxic stress and engagement of protein quality control pathways as responses to their presence and disruptive activities, they differ in protein context, normal function, localization, interactomes and selective vulnerability. Here, we review how expanded polyglutamine proteins engage ubiquitin-dependent protein quality control across Huntington's disease, dentatorubral-pallidoluysia

Antisense Oligonucleotide-Mediated Degradation of Toxic RNA in Myotonic Dystrophy Type 1.

Myotonic dystrophy (DM) exists in two forms, type 1 (DM1) and type 2 (DM2), both inherited in an autosomal dominant manner and caused by toxic gain-of-function RNAs produced from unstable repeat expansions. In DM1, the mutation lies in the DMPK gene, whereas in DM2 it involves the CNBP gene. These repeats are inherently unstable, with a strong tendency to further expand in somatic tissues and across generations, driving disease progression and anticipation. Despite arising from distinct genetic defects, DM1 and DM2 share a common pathogenic pathway-RNA-mediated toxicity-which accounts for thei

Genome-wide identification and evolution of the RCP gene family in soybean uncovers stress-responsive candidates.

Abiotic stress has a significant impact on soybean growth, development, and yield. Proteins containing the Regulator of Chromosome Condensation 1 (RCC1) domain, known as RCPs, play important roles in plant stress responses. However, systematic analysis of the RCP gene family in soybean remains limited. In this study, a total of 52 GmRCP genes were identified in the soybean genome and classified into nine phylogenetic clades. Members within the same clade exhibited similar domain architectures. Collinearity analysis suggested that segmental duplication served as the major driver for the expansi

The Arabidopsis ARF3-AIP1/2-SAP18 module specifies the root stem cell niche in response to auxin.

In the root apical meristem, the stem cell niche (SCN) comprises a mitotically inactive quiescent center (QC) and adjacent, mitotically active stem cells that divide to form root tissues. Auxin dynamics are essential for specification and maintenance of the root SCN; however, the underlying mechanisms remain to be explored. Here, we report that Arabidopsis (Arabidopsis thaliana) AUXIN RESPONSE FACTOR3 (ARF3), ARF3-INTERACTING PROTEIN1/2 (AIP1/2), and SIN3-ASSOCIATED POLYPEPTIDE OF 18 KDA (SAP18) form a protein complex that specifies root SCN cell fate in response to auxin level. Harnessing endophytes and Multi-Omics for sustainable Colchicine biosynthesis.

Gloriosa superba, an endangered medicinal plant, serves as the principal natural source of colchicine, a vital alkaloid used for treating gout, arthritis, cancer, and various inflammatory disorders. However, its conventional extraction from plant tissues is constrained by low yield, ecological degradation, and conservation concerns, necessitating sustainable production alternatives. Emerging evidence indicates that colchicine biosynthesis is not solely plant-autonomous but is strongly influenced by endophytic microorganisms that function as active metabolic partners. Endophytic fungi and bacte

ModelistsGCN: a multimodal graph convolutional network framework for single-cell spatial transcriptomic cell typing.

Spatial transcriptomics technologies currently face a trade-off between spatial and molecular resolution. High-resolution single-cell methods such as MERFISH and Expansion Sequencing (ExSeq) resolve individual cells but typically profile limited gene panels, restricting the direct application of transcriptome-based cell typing strategies developed for single-cell RNA sequencing. Consequently, accurate cell-type identification in spatial single-cell data remains challenging when transcriptomic coverage per cell is sparse. Here, we introduce ModelistsGCN, a semi-supervised multimodal graph convo

Blood feeding-induced transcriptomic changes in the hard tick Ixodes persulcatus.

INTRODUCTION: Ticks are hematophagous ectoparasites that must overcome significant physiological challenges during blood feeding. These include managing oxidative stress, detoxifying host-derived molecules, and reallocating energy to support digestion, tissue remodeling, and reproduction. METHODS: In this study, we conducted a de novo transcriptome assembly and genome-wide transcriptional profiling of female Ixodes persulcatus ticks at three key feeding stages: unfed, semi-engorged, and fully engorged. Functional annotation and Gene Ontology (GO) enrichment analyses were conducted to characte

Single-Cell Transcriptomic Profiling Uncovers a Metastasis-Associated MUCL3+ Signet-Ring Cell Subpopulation in Gastric Cancer.

Background: Gastric signet-ring cell carcinoma (GSRCC) is an aggressive gastric cancer subtype with abundant mucin production and high metastatic propensity. However, scarcity of specific biomarkers has impeded clinical diagnosis and mechanistic research. This study systematically compares GSRCC and gastric adenocarcinoma (AC) to identify biomarkers and elucidate molecular basis of GSRCC's aggressive behavior. Methods: We performed single-cell RNA sequencing (scRNA-seq) on surgically resected primary GC tissues, validating our findings using public datasets and functional experiments. Results:

Enhanced virulence and neuroinvasion of contemporary Oropouche virus strains in the AG129 mouse model.

Oropouche virus (OROV) is an orthobunyavirus undergoing dramatic re-emergence and rapid geographic expansion across Central and South America, coinciding with the emergence of reassortant lineages carrying novel segment combinations and adaptive mutations. Despite extensive genomic and epidemiological characterization, the biological mechanisms driving OROV resurgence during 2023-2024 remain unclear. Here, we show that contemporary reassortant OROV isolates exhibit markedly enhanced virulence in the AG129 mouse model compared with the historical prototype strain OROVBeAn19991. Two reassortant

Genome-wide characterization of the TGF-β superfamily identifies bmp15, gdf9, and gsdf as sex-biased candidate regulators of gonadal differentiation in the synchronous hermaphrodite Plectropomus leopardus.

The transforming growth factor-β (TGF-β) superfamily plays conserved roles in vertebrate reproduction and gonadal sex differentiation. However, its genomic repertoire and sex-biased expression patterns remain unclear in the leopard coral grouper (Plectropomus leopardus), a species with synchronous hermaphroditism. Here, we performed a genome-wide identification of the TGF-β superfamily, identifying 42 genes from the chromosome-level genome. Phylogenetic and synteny analyses indicated that segmental duplication under purifying selection contributed to family expansion. Expression profiling acro

The Power of MUC2 in Mucinous Adenocarcinomas.

<p>Background: In cancers, key prognostic determinants such as clinical stage and mutational load are well recognized. However, mucinous adenocarcinomas (MACs) display distinct biological behavior as a large proportion of their tumor volume consists of extracellular mucin, predominantly the MUC2 protein, rather than neoplastic cells. MUC2 provides a protective barrier for cancer cells against external factors, including chemotherapeutic agents and cytotoxic lymphocytes, while its anti-inflammatory and tumor-suppressive properties paradoxically support tumor persistence and immune evasion.
