Is ridge preservation/augmentation at periodontally compromised extraction sockets safe? A retrospective study.

OBJECTIVES: This study aimed to evaluate the safety of ridge preservation/augmentation procedures when performed at compromised extraction sockets. METHODS: Patients subject to ridge preservation/augmentation at periodontally compromised sockets at Seoul National University Dental Hospital (SNUDH) were evaluated in a chart review. Tooth extractions due to acute infection were not included in our study as chronically formed lesions are the only lesions that can be detected from radiographic images. If inflammatory symptoms persisted following ridge preservation/augmentation and antimicrobial a

Effects of ovariectomy on turnover of alveolar bone in the healed extraction socket in rat edentulous mandible.

OBJECTIVE: The number of patients with postmenopausal osteoporosis receiving dental implants because of edentulism is increasing. Since osseointegration around implants requires formation and maintenance of new bone, knowledge of how ovariectomy (OVX) affects turnover of mandibular and maxillary bone is required. In the present study, we investigated the effects of OVX on turnover of alveolar bone in the healed extraction socket of the rat left mandibular incisor. METHODS: The molars and the incisor on left side in 6-month-old Sprague-Dawley female rats (n=38) were extracted and left to heal

A retrospective clinical and radiologic study of teeth re-implanted following traumatic avulsion.

Re-implantation is the recommended therapeutic procedure following traumatic exarticulation of teeth though its long-term prognosis remains controversial. The purpose of the following study was to evaluate the periodontal healing of 33 reimplanted incisors lost after trauma. The sample, which included 24 upper and 9 lower incisors, was divided in two groups: 15 teeth were reimplanted within 1 hour (=Group A) and 21 teeth after 3 hours or more (=Group B). The reimplanted teeth were followed for different radiographic evaluation periods up to 5 years (mean=2-9 years). The results showed a high r

Subgingival odontoclastic resorptive lesions. Classification, treatment, and results in 58 cats.

A commonly recognized dental problem in cats is the resorption of tooth structure and subsequent loss of the tooth. These tooth defects are often very painful, because the sensitive dentin layer is exposed. The destruction of the tooth through odontoclastic resorption is considered a consequence of inflammatory resorption, probably secondary to periodontal inflammation. Because these resorptive lesions are progressive in nature, it is best to stage this progression of resorption in order to address treatment planning. The purpose of this study was to evaluate a group of 58 cats with resorptive

Histological differences between teeth with adult periodontitis and prepubertal periodontitis.

Thirty-nine permanent teeth from three siblings aged 11 to 14 years were extracted because of generalized prepubertal periodontitis (PP) and studied histologically with light microscopy. Twenty healthy teeth (HT) removed for orthodontic procedures from 10 children of similar ages and 22 teeth affected by adult periodontitis (AP) extracted from patients over 50 years old were used as controls. The cementum of the PP teeth was much thicker than that of the healthy teeth. A high percentage of resorption, unrelated to inflammation, was found in all the teeth studied. In 24 PP teeth the cementum wa

The Aymara of western Bolivia: III. Occlusion, pathology, and characteristics of the dentition.

With the objective of contributing to the understanding of factors influencing the characteristics of dentition, dental observations from 429 children and adult highland people of western Bolivia are described. Various age, sex, ethnic, and village differences are observed in malocclusion, dental caries, periodontal disease, dental morphology, and anomalies. The findings revealed a population with relatively good occlusion, low frequency of dental pathology, and dental characteristics in part consistent with those of other similar Amerindian populations.

A pleiotropic recurrent dominant ITPR3 variant causes a complex multisystemic disease.

Inositol 1,4,5-trisphosphate (IP3) receptor type 1 (ITPR1), 2 (ITPR2), and 3 (ITPR3) encode the IP3 receptor (IP3R), a key player in intracellular calcium release. In four unrelated patients, we report that an identical ITPR3 de novo variant-NM_002224.3:c.7570C>T, p.Arg2524Cys-causes, through a dominant-negative effect, a complex multisystemic disorder with immunodeficiency. This leads to defective calcium homeostasis, mitochondrial malfunction, CD4+ lymphopenia, a quasi-absence of naïve CD4+ and CD8+ cells, an increase in memory cells, and a distinct TCR repertoire. The calcium defect was rec

Hydrogels promote periodontal regeneration.

Periodontal defects involve the damage and loss of periodontal tissue, primarily caused by periodontitis. This inflammatory disease, resulting from various factors, can lead to irreversible harm to the tissues supporting the teeth if not treated effectively, potentially resulting in tooth loss or loosening. Such outcomes significantly impact a patient's facial appearance and their ability to eat and speak. Current clinical treatments for periodontitis, including surgery, root planing, and various types of curettage, as well as local antibiotic injections, aim to mitigate symptoms and halt dise

Mechanisms of bone remodeling and therapeutic strategies in chronic apical periodontitis.

Chronic periapical periodontitis (CAP) is a typical oral disease in which periodontal inflammation caused by an odontogenic infection eventually leads to bone loss. Uncontrolled infections often lead to extensive bone loss around the root tip, which ultimately leads to tooth loss. The main clinical issue in the treatment of periapical periodontitis is the repair of jawbone defects, and infection control is the first priority. However, the oral cavity is an open environment, and the distribution of microorganisms through the mouth in jawbone defects is inevitable. The subversion of host cell me

In vivo evaluation of chitosan based local delivery systems for atorvastatin in treatment of periodontitis.

Periodontitis is a local inflammatory disease initiated by bacteria accumulation and results in cytokine mediated alveolar bone resorption and tissue destruction. In this study, the effect of locally delivered atorvastatin (2% w/v) containing chitosan formulations in the treatment of periodontitis was evaluated in rats with ligature induced periodontitis. The levels of interleukin-1beta (IL-1β), IL-6, IL-8, IL-10, transforming growth factor-β1 (TGF-β1), TGF-β2 and TGF-β3 were measured after treatment with formulations. Histomorphometric analysis included the measurements of the area of alveola

TRPV4 channel activity is modulated by direct interaction of the ankyrin domain to PI(4,5)P₂.

Mutations in the ankyrin repeat domain (ARD) of TRPV4 are responsible for several channelopathies, including Charcot-Marie-Tooth disease type 2C and congenital distal and scapuloperoneal spinal muscular atrophy. However, the molecular pathogenesis mediated by these mutations remains elusive, mainly due to limited understanding of the TRPV4 ARD function. Here we show that phosphoinositide binding to the TRPV4 ARD leads to suppression of the channel activity. Among the phosphoinositides, phosphatidylinositol-4,5-bisphosphate (PI(4,5)P2) most potently binds to the TRPV4 ARD. The crystal structure

[Translational Medicine and its application in stem cell research for periodontal regeneration].

Translational Medicine is an evolutional concept that encompasses the rapid translation of basic research for use in clinical disease diagnosis, prevention, treatment and finally in public health promotion. It follows the idea "from bench to bedside and back", and hence relies on cooperation between laboratory research and clinical care. Translation process is a complex process that requires both research and non-research activities. During the past ten years, there has been intense interest in the development of new clinical procedures, therapeutic molecules, and prototypes based on translati

Local complement-targeted intervention in periodontitis: proof-of-concept using a C5a receptor (CD88) antagonist.

When excessively activated or deregulated, complement becomes a major link between infection and inflammatory pathology including periodontitis. This oral inflammatory disease is associated with a dysbiotic microbiota, leads to the destruction of bone and other tooth-supporting structures, and exerts an adverse impact on systemic health. We have previously shown that mice deficient either in complement C5a receptor (C5aR; CD88) or TLR2 are highly and similarly resistant to periodontitis, suggesting that a cross-talk between the two receptors may be involved in the disease process. In this pape

Comparative proteomic analysis of whole saliva from chronic periodontitis patients.

Chronic periodontal disease is a chronic inflammatory process affecting tooth supporting tissues in the presence of pathogenic bacterial biofilm. There is some evidence for changes in the protein composition of whole saliva from chronic periodontitis patients, but there have been no studies using a proteomic approach. Hence, the aim of this study was to compare the protein profiles of unstimulated whole saliva from patients with periodontitis and healthy subjects by two complementary approaches (2D-gel electrophoresis and liquid chromatography). Protein spots of interest were analyzed by MALDI

New approaches to enhanced remineralization of tooth enamel.

Dental caries is a highly prevalent diet-related disease and is a major public health problem. A goal of modern dentistry is to manage non-cavitated caries lesions non-invasively through remineralization in an attempt to prevent disease progression and improve aesthetics, strength, and function. Remineralization is defined as the process whereby calcium and phosphate ions are supplied from a source external to the tooth to promote ion deposition into crystal voids in demineralized enamel, to produce net mineral gain. Recently, a range of novel calcium-phosphate-based remineralization delivery
