PPP1R15A Promotes Apoptosis, Autophagy, and Inflammatory Response to Exacerbate Ischemic Stroke Through Activation of TLR4/NF-κB Pathway.

Ischemic stroke (IS) is a prevalent and serious neurological disorder, and is one of the major contributors to mortality and disability worldwide. The activation of autophagy and the inflammatory response following cerebral ischemia are crucial in the progression of IS. Protein phosphatase 1 regulatory subunit 15A (PPP1R15A), as a stress-responsive protein, has been proven to be closely associated with autophagy. Nevertheless, the molecular mechanism of PPP1R15A in IS remains to be fully understood. Our study screened PPP1R15A as a candidate gene by bioinformatics analysis of the differentiall…

MYT1L promotes autophagy to ameliorate amyloid-β pathology and cognitive deficits in Alzheimer's disease via Notch signaling pathway.

Autophagy is essential in the growth and advancement of Alzheimer's disease (AD). Myelin transcription factor 1-like (MYT1L) has been reported to have neuroprotective properties; however, its precise role in AD is still unclear. This study identified a reduction in MYT1L expression in the hippocampal neurons of AD model mice. Interestingly, increasing MYT1L expression in these neurons helped alleviate cognitive impairments in APP/PS1 mice. Additionally, MYT1L overexpression reduced the amyloid-beta (Aβ) burden, promoted autophagy, and provided neuroprotection in the hippocampus of these mice. …

Sinularin induces autophagy-dependent cell death by activating ULK1 and enhancing FOXO3-ATG4A axis in prostate cancer cells.

Sinularin is a natural product extracted from soft coral and is shown to exhibit antitumor effects against multiple human cancers. We previously showed that Sinularin induces apoptotic cell death via stabilizing the FOXO3 protein in prostate cancer cells. In this study, we demonstrated that Sinularin triggers autophagy via two different mechanisms in prostate cancer cells. First, Sinularin reduced the S757 phosphorylation of ULK1 protein, which was mediated by mTOR, leading to ULK1 activation and autophagy initiation. Second, Sinularin enhanced the expression of autophagic protein ATG4A, which…
