PAX5 modulates vascularization and contributes to neuroendocrine lineage transition during carcinoma progression.

Human carcinomas often gain aggressive characteristics and escape cell type-specific treatment regimens through cryptic shifts in lineage states. However, the underlying mechanisms that govern lineage plasticity in carcinomas are undefined. Here in this study, we found that PAX5, a neural/lymphatic transcription factor, contributed to neuroendocrine (NE) lineage transition. PAX5 was highly expressed in aggressive human NE carcinoma cells and tissues but not in non-NE cancer cells and tissues. Deletion of Pax5 in Rb1fl/fl Trp53fl/fl mice caused a reduction of tumor vessels, loss of NE morpholog

Establishment and characterization of a human juvenile bone marrow-derived mesenchymal stem/stromal cell line under advanced culture conditions for osteogenic differentiation.

Human mesenchymal stem/stromal cells (MSC) from juvenile donors (juvMSC) are crucial for studying bone development and for modeling pediatric skeletal diseases. However, the limited availability of these cells and the lack of physiologically relevant in vitro models hinder preclinical research. To address these issues, we established and characterized a new human bone marrow-derived MSC line under advanced culture conditions. Primary MSC from a 12-year-old donor in good health were immortalized via lentiviral transduction using a library of expansion genes. The resulting clone, C15 juvMSC, ret

Enhanced Efficacy of Sandblasted, Large-Grit, Alkaline-Etched (SLA) Titanium Surfaces Combined with Plasma Treatment and Secretory Leukocyte Protease Inhibitor (SLPI) Coating in Promoting Osteoblast Activity and Reducing Bacterial Adhesion.

The clinical success rate of titanium (Ti) implants is significantly reduced in elderly patients and by implant-associated bacterial infections. Thus, Ti surfaces were modified using sandblasted large-grit alkaline etching (SLA), argon-nonthermal plasma treatment, and recombinant human secretory leukocyte protease inhibitor (rhSLPI) coating. This study evaluated surface characteristics, human fetal osteoblast (hFOB 1.19) activities, and antibacterial efficacy. The modified surfaces exhibited increased roughness and hydrophilicity compared to Ti. The SBPTi-rhSLPI enhanced 7-day protein retentio

Upper limb human-exoskeleton system motion state classification based on semg: application of CNN-BiLSTM-attention model.

This study aims to classify five typical motion states of the human upper limb based on surface electromyography signals, thereby supporting the real-time control system of an assistive upper limb exoskeleton. We propose a deep learning model combining convolutional neural networks, bidirectional long short-term memory networks, and attention mechanism to enhance the accuracy of motion state recognition in complex scenarios. Surface electromyography data were collected from ten participants for the biceps, triceps, and deltoid muscles, covering five representative states: resting, mild activit

Transcriptomic Differences Between Human Trabecular Meshwork Stem Cells and Trabecular Meshwork Cells Reveal Specific Biomarker Profiles.

Glaucoma is a leading cause of irreversible blindness, normally associated with dysfunction and degeneration of the trabecular meshwork (TM) as the primary cause. Trabecular meshwork stem cells (TMSCs) have emerged as promising candidates for TM regeneration toward glaucoma therapies, yet their molecular characteristics remain poorly defined. In this study, we performed a comprehensive transcriptomic comparison of human TMSCs and human TM cells (TMCs) using RNA sequencing and microarray analyses, followed by qPCR validation. A total of 465 differentially expressed genes were identified, with 2

Metabolomics analysis reveals lipid metabolism disorders caused by triphenyltin via upregulation of PPARγ pathway.

Triphenyltin (TPT), a widely utilized organotin compound, serves marine, agricultural, and industrial applications. TPT is commonly found in estuaries and coastal regions. However, recent studies on environmental pollution have identified TPT as a pollutant due to its pronounced pollutant characteristics and potential implications for human health. Research from the past has demonstrated that TPT can cause issues with lipid metabolism. However, the specific metabolites affected and the involvement of the PPAR pathway remain unknown. Herein, male Sprague-Dawley (SD) rats received oral administr

First Spectrofluorimetric Method for Sensitive Determination of Armodafinil in Dosage Forms and Human Plasma: Greenness, Blueness, and Whiteness Assessment.

A novel, sensitive, and reliable spectrofluorimetric method for the determination of a unique antinarcoleptic drug armodafinil in human plasma and dosage forms was developed for the first time. The proposed method is based on the formation of a highly yellow fluorescent derivative through the reaction of the studied drug with acetylacetone and formaldehyde in an acetate buffer (pH 4.5), yielding a strong emission at 485 nm following excitation at 415 nm. The experimental variables that affected the one-pot synthesis were investigated and optimized. The calibration curve demonstrated a linear r

N-Alkane Assimilation by Pseudomonas aeruginosa and Its Interactions with Virulence and Antibiotic Resistance.

Pseudomonas aeruginosa strains with potential for degrading n-alkanes are frequently cultured from hydrocarbon-contaminated sites. The initial hydroxylation step of long-chain n-alkanes is mediated by the chromosomally encoded AlkB1 and AlkB2 alkane hydroxylases. The acquisition of an additional P. putida GPo1-like alkane hydroxylase gene cluster can extend the substrate range assimilated by P. aeruginosa to <C12 n-alkanes. Efficient niche colonization of hydrocarbon-contaminated sites is facilitated by avid iron-uptake systems, such as pyoverdine, and the production of several compounds with

Human umbilical cord mesenchymal stem cells promote steroid-induced osteonecrosis of the femoral head repair by improving microvascular endothelial cell function.

Recently, there has been growing interest in using cell therapy through core decompression (CD) to treat osteonecrosis of the femoral head (ONFH). Our study aimed to investigate the effectiveness and mechanism of human umbilical cord mesenchymal stem cells (hUCMSCs) in treating steroid-induced ONFH. We constructed a steroid-induced ONFH rabbit model as well as dexamethasone (Dex)-treated bone microvascular endothelial cells (BMECs) model of human femoral head. We injected hUCMSCs into the rabbit femoral head via CD. The effects of hUCMSCs on steroid-induced ONFH rabbit model and Dex-treated BM

Host-microbe interactions at the blood-brain barrier through the lens of induced pluripotent stem cell-derived brain-like endothelial cells.

Microbe-induced meningoencephalitis/meningitis is a life-threatening infection of the central nervous system (CNS) that occurs when pathogens are able to cross the blood-brain barrier (BBB) and gain access to the CNS. The BBB consists of highly specialized brain endothelial cells that exhibit specific properties to allow tight regulation of CNS homeostasis and prevent pathogen crossing. However, during meningoencephalitis/meningitis, the BBB fails to protect the CNS. Modeling the BBB remains a challenge due to the specialized characteristics of these cells. In this review, we cover the induced

Salt-Tolerant Plants as Sources of Antiparasitic Agents for Human Use: A Comprehensive Review.

Parasitic diseases, especially those caused by protozoans and helminths, such as malaria, trypanosomiasis, leishmaniasis, Chagas disease, schistosomiasis, onchocerciasis, and lymphatic filariasis, are the cause of millions of morbidities and deaths every year, mainly in tropical regions. Nature has always provided valuable antiparasitic agents, and efforts targeting the identification of antiparasitic drugs from plants have mainly focused on glycophytes. However, salt-tolerant plants (halophytes) have lately attracted the interest of the scientific community due to their medicinal assets, whic

Reemergence of Marburgvirus disease: Update on current control and prevention measures and review of the literature.

In 1967, the very first case of the Marburgvirus disease (MVD) was detected in Germany and Serbia sequentially. Since then, MVD has been considered one of the most serious and deadly infectious diseases in the world with a case-fatality rate between 23% and 90% and a substantial number of recorded deaths. Marburgvirus belongs to the family of Filoviridae (filoviruses), which causes severe viral hemorrhagic fever (VHF). Some major risk factors for human infections are close contact with African fruit bats, MVD-infected non-human primates, and MVD-infected individuals. Currently, there is no vac

3D printed transwell-integrated nose-on-chip model to evaluate effects of air flow-induced mechanical stresses on mucous secretion.

While there are many chip models that simulate the air-tissue interface of the respiratory system, only a few represent the upper respiratory system. These chips are restricted to unidirectional flow patterns that are not comparable to the highly dynamic and variable flow patterns found in the native nasal cavity. Here we describe the development of a tunable nose-on-chip device that mimics the air-mucosa interface and is coupled to an air delivery system that simulates natural breathing patterns through the generation of bi-directional air flow. Additionally, we employ computational modeling

New Retinal Pigment Epithelial Cell Model to Unravel Neuroprotection Sensors of Neurodegeneration in Retinal Disease.

Retinal pigment epithelial (RPE) cells sustain photoreceptor integrity, and when this function is disrupted, retinal degenerations ensue. Herein, we characterize a new cell line from human RPE that we termed ABC. These cells remarkably recapitulate human eye native cells. Distinctive from other epithelia, RPE cells originate from the neural crest and follow a neural development but are terminally differentiated into "epithelial" type, thus sharing characteristics with their neuronal lineages counterparts. Additionally, they form microvilli, tight junctions, and honeycomb packing and express di

Exploring the Immune-Boosting Functions of Vitamins and Minerals as Nutritional Food Bioactive Compounds: A Comprehensive Review.

Food components have long been recognized to play a fundamental role in the growth and development of the human body, conferring protective functionalities against foreign matter that can be severe public health problems. Micronutrients such as vitamins and minerals are essential to the human body, and individuals must meet their daily requirements through dietary sources. Micronutrients act as immunomodulators and protect the host immune response, thus preventing immune evasion by pathogenic organisms. Several experimental investigations have been undertaken to appraise the immunomodulatory f

Impact of human behavior on the spread of African swine fever virus: what every veterinarian should know.

African swine fever virus was first identified and characterized in Africa in the early 1900s, but it has spread exponentially in Europe, Asia, and the Caribbean since 2018. While it is a disease that exclusively affects swine, thus posing no infectious risk to human health, the virus's resiliency and human behavior have facilitated the rapid global dissemination of the virus over the past 4 years. In this Currents in One Health, we will review its epidemiology, viral characteristics, host range, and current prevention strategies; the current perspective on what a response would look like and

Detection of Glaucoma from Fundus Images Using Novel Evolutionary-Based Deep Neural Network.

Glaucoma is an asymptotic condition that damages the optic nerves of a human eye. Glaucoma is frequently caused due to abnormally high pressure in an eye that leads to permanent blindness. Detecting glaucoma at an initial phase has the possibility of curing this disease, but diagnosing accurately is considered as a challenging task. Therefore, this paper proposes a novel method known as a glaucoma detection system that performs the diagnosis of glaucoma by exploiting the prescribed characteristics. The significant intention of this paper involves diagnosing the glaucoma disease present at the

Flavonoids Target Human Herpesviruses That Infect the Nervous System: Mechanisms of Action and Therapeutic Insights.

Human herpesviruses (HHVs) are large DNA viruses with highly infectious characteristics. HHVs can induce lytic and latent infections in their host, and most of these viruses are neurotropic, with the capacity to generate severe and chronic neurological diseases of the peripheral nervous system (PNS) and central nervous system (CNS). Treatment of HHV infections based on strategies that include natural products-derived drugs is one of the most rapidly developing fields of modern medicine. Therefore, in this paper, we lend insights into the recent advances that have been achieved during the past

Retrograde axonal transport of poliovirus and EV71 in motor neurons.

Poliovirus (PV) can spread through neural pathway to the central nervous system and replicates in motor neurons, which leads to poliomyelitis. Enterovirus 71 (EV71), which is closely related to PV, is one of the causative agents of hand-foot-and-mouth disease and can cause severe neurological diseases similar to poliomyelitis. Since PV is similar to EV71 in its motor neurotoxicity, we tried to understand if the results obtained with PV are of general applicability to EV71 and other viruses with similar characteristics. Using microfluidic devices, we demonstrated that both PV capsid and the PV
