Immunohistochemical expression of rabphilin-3A-like (Noc2) in normal and tumor tissues of human endocrine pancreas.

Involvement of rabphilin-3A-like (RPH3AL), or Noc2, the potential effector of Ras-associated binding proteins Rab3A and Rab27A in the regulation of exocytotic processes in the endocrine pancreas has been demonstrated in experimental models. Noc2 expression together with other regulatory molecules of the exocytotic machinery in human tissues, however, has not been studied. We evaluated immunohistochemical expression of the key molecules of the exocytotic machinery, Noc2, Rab3A, Rab27A, and RIM2, together with the characteristic islet cell hormones, insulin and glucagon in normal and endocrine t

Testing different antigen capture ELISA formats for detection of Leptospira spp. in human blood serum.

Leptospirosis is an infectious disease caused by pathogenic spirochetes of the genus Leptospira. The illness is characterized by an acute bacteremic phase followed by an immune phase, in which specific antibodies are found in blood and leptospires are eliminated in urine. Novel diagnostic strategies for use in the acute phase of leptospirosis are needed since clinical manifestations in this phase mimic other feverish tropical diseases. In the present study, mAbs and polyclonal IgY were used in the standardization of three different antigen capture ELISA formats for direct detection of leptospi

[New perspectives in the use of human hepatocytes in the preclinical drug development process].

Human and animal hepatocytes are now widely used for drug metabolism and interaction studies in the drug development process. However, their phenotypic instability and the absence of cell division in primary culture limit their interest for toxicity studies. Hepatoma cell lines are also used but they express very low levels, if any, of cytochromes P450 (CYP) that are essential for metabolism of a number of drugs and other chemicals. A new human hepatoma cell line, named HepaRG, possesses both the metabolic capacity of human hepatocytes in primary culture and the indefinite proliferation potent

Embryology and genetics of primary vesico-ureteric reflux and associated renal dysplasia.

Congenital anomalies of the kidney and urinary tract, as well as primary vesico-ureteric reflux (VUR) and associated renal dysplasia, are the most relevant causes of end-stage renal failure in the pediatric population. In vivo and in vitro experimental studies have allowed the identification of several genes involved both in ureteric bud branching, ureteric elongation and insertion into the bladder, and in nephrogenesis. It has been proposed that both renal and ureteral abnormalities, as well as the associated renal hypo-dysplasia, may derive from a common mechanism as the result of a dysregul

Analysis of a mutant p53 protein arising in a medulloblastoma from a mouse transgenic for the JC virus early region.

BACKGROUND: JC virus (JCV) is a polyomavirus that causes progressive multifocal leukoencephalopathy (PML) in humans and is highly oncogenic in experimental animals. Transgenic mice with JCV T-antigen develop cerebellar tumors, which resemble human medulloblastomas, containing two distinct cell subpopulations, T-antigen positive and negative. In T-negative clones, a novel mutant p53 was detected (p53(mt)). MATERIALS AND METHODS: We have compared p53(mt) to wild-type p53 (p53wt) in p53-null cells. RESULTS: p53(mt) had lost the transcriptional transactivation activity of p53(wt), and unlike p53

Aquaporin expression in normal human kidney and in renal disease.

Aquaporins (AQPs), membrane-inserted water channel proteins, play a highly important role in the reabsorption of water from the renal tubular fluid. Experimentally, both in rats and mice, failure to insert functional AQP molecules into renal tubular membranes leads to nephrogenic diabetes insipidus. In humans, most forms of renal disease lead to a reduction in the water handling capacity of the kidney. AQP distribution in various forms of human renal disease has not been documented. Immunohistochemical studies of biopsy samples from a wide range of renal diseases revealed a substantial and str

The impact of T-ACASI interviewing on reported drug use among men who have sex with men.

Measurements of drug use and other illicit or stigmatized behaviors are subject to nontrivial underreporting biases. During in-person surveys, respondents are more likely to report such behaviors when interviewed using techniques that maximize interviewee privacy, e.g., use of paper SAQs and audio-CASI rather than questioning by human interviewers. Until recently, respondents in telephone surveys could not be offered similar privacy. A new technology, telephone audio computer-assisted self-interviewing (T-ACASI) overcomes this limitation of telephone surveys by allowing respondents to respond

Single and multiple transgenic mice as models for Alzheimer's disease.

Transgenic mice expressing in brain different mutant forms of the Amyloid Precursor Protein, develop functional, cognitive and pathological defects which resemble or are reminiscent of symptoms observed in Alzheimer's disease (AD) patients. The late development of amyloid plaques in aging transgenic APP mice is needed to warrant that the earlier behavioural and cognitive defects are informative for the human disorder. We describe and discuss our work, the rationale behind the approach and the techniques used to generate these APP transgenic mice, including specific experimental problems. The A

Twenty-seven years studying the human neurotoxicity of methylmercury exposure.

Research at the University of Rochester (U of R) has been focused on mercury for nearly half a century. Initially studies focused on dosimetry, especially the accuracy of measuring exposure, and experimental work with animal models. Clinical studies in human populations started when the U of R mercury group was asked to assist with dosimetry in the Iraq epidemic of 1971-1972. Initial clinical studies described the effects of methylmercury (MeHg) poisoning on adults and children. A dose-response curve for prenatal exposure was determined and it suggested that relatively low exposures might be h

Do NSAIDs prevent colorectal cancer?

There is increasing evidence to suggest that acetylsalicylic acid (ASA) and other nonsteroidal anti-inflammatory drugs (NSAIDs) reduce the risk of colorectal cancer. This observation is supported by animal studies that show fewer tumours per animal and fewer animals with tumours after administration of several different NSAIDs. Studies in humans consistently support this hypothesis. Intervention data from familial adenomatosis coli establish that the process of human colonic adenoma polyp formation is affected. Supportive evidence comes from 21 of 23 human studies - both case-control and cohor

Increased excitability of the human corticospinal system with hyperventilation.

OBJECTIVES: Hyperventilation is effective in inducing generalized spike-wave discharges in patients with absence seizures and improves visual function and normalizes visual function in patients with multiple sclerosis. Hyperventilation increases the excitability of cutaneous and motor axons. In experimental animals, hyperventilation increases excitability of hippocampal neurons. There is however no direct evidence of a hyperventilation-induced increase in neuronal excitability within the central nervous system in humans. In this study we determined the effects of hyperventilation on the human

Presence of potent transcriptional activation domains in the predisposing tuberous sclerosis (Tsc2) gene product of the Eker rat model.

The Eker rat hereditary renal carcinoma is an excellent example of Mendelian dominant predisposition to a specific cancer in an experimental animal. We recently reported that a germline insertion in the rat homologue of the human tuberous sclerosis gene (TSC2) gives rise to dominantly inherited cancer in the Eker rat model, as well as a tumor suppressor nature for the Tsc2 gene function. We also showed a strong conservation between the rat and human gene products. The molecular function of the Tsc2 gene product (called "tuberin" in the human case) is not yet understood, although it contains a

Characterization of human IgG subclasses within intraocular compartments.

Multiple factors regulate the distribution of the human immunoglobulin (Ig) G1-4 subclasses within different compartments of the human eye. An experimental protocol was developed for analysis of IgG1-4 using a combination of high pressure liquid chromatography (HPLC) chromatofocusing and an enzyme-linked immunosorbent assay (ELISA). IgG in the human cornea, aqueous humor, and vitreous was extracted and fractionated by chromatofocusing in a pH 9.0-6.0 gradient. A monoclonal antibody based capture-type ELISA was used to determine the IgG1-4 subclass distribution within the fractions. Histograms

A Review: Mast Cells as Targets for Glucocorticoid Action.

PURPOSE OF REVIEW: This review aims to summarize the established and potential targets of glucocorticoids (GCs) on mast cells, which are implicated in the pathophysiology of various allergic and inflammatory diseases including asthma, allergic rhinitis, and atopic dermatitis. We sought to answer the following questions: (1) How do glucocorticoids inhibit the release of mediators from mast cells? (2) What are the effects of glucocorticoids on mast cell signaling molecules? (3) By what mechanisms do glucocorticoids induce mast cell apoptosis? and (4) What are the genomic and non-genomic mechanis

Diversifying Selection Analysis Predicts Antigenic Evolution of 2009 Pandemic H1N1 Influenza A Virus in Humans.

UNLABELLED: Although a large number of immune epitopes have been identified in the influenza A virus (IAV) hemagglutinin (HA) protein using various experimental systems, it is unclear which are involved in protective immunity to natural infection in humans. We developed a data mining approach analyzing natural H1N1 human isolates to identify HA protein regions that may be targeted by the human immune system and can predict the evolution of IAV. We identified 16 amino acid sites experiencing diversifying selection during the evolution of prepandemic seasonal H1N1 strains and found that 11 sites

Deep cervical lymphaticovenous anastomosis for Alzheimer disease: A critical narrative assessment and translational framework.

Deep cervical lymphaticovenous anastomosis (dcLVA) has been proposed to augment cerebral waste clearance in Alzheimer disease. This critical narrative assessment examines the mechanistic premise, available human reports, registered studies, and governance implications. Experimental loss-of-function and gain-of-function studies support a role for meningeal lymphatics in brain-fluid homeostasis, but they do not establish a rate-limiting cervical outflow abnormality in human Alzheimer disease. The clinical reports identified through this narrative search were uncontrolled single-arm investigation

Applying Spatial Transcriptomics to the Human Heart: A Practical Guide for Translational Studies.

Spatial transcriptomics and single-cell methods are changing the questions we can ask about the human cardiovascular system. In this review, we share lessons learned from applying spatial profiling to human cardiovascular samples. We walk through experimental considerations, sampling, fixation, and ribonucleic acid quality control, then compare sequencing-based and imaging-based platforms within a 2-stage "discovery then validation" framework. We propose a heart-aware analytical pipeline: spatially informed quality control, normalization respecting the outsized transcriptome of cardiomyocytes,

Force-moment mechanics of wiggling in connector insertion.

Humans instinctively use both pushing and wiggling motions when inserting objects into sockets, connectors, or mechanical interfaces that resist fitting, often without understanding the rationale behind these actions. Despite their apparent simplicity, such tasks involve complex mechanical interactions that are challenging to describe mathematically. In this paper, we present a simplified two-leg mathematical model that clarifies the fundamental principles governing the combined pushing-wiggling during assembly. The model integrates forces, moments, frictional reactions, and other uncertain ef

Type I interferon controls vertical transmission and fetoplacental infection of Oropouche virus.

The current outbreak of the emerging arthropod-transmitted Oropouche virus (OROV) in South America has been epidemiologically linked to vertical transmissions, microcephaly, and stillbirths. Nevertheless, the impact of OROV infection during pregnancy has not been experimentally evaluated. To address how OROV infection might impact pregnancy outcome, we performed experiments in human cells and mice. Studies in cell cultures showed that the human trophoblast cell lines BeWo and JEG-3 are permissive to OROV infection (strain BeAn19991) and develop type I interferon (IFN)-dependent antiviral respo

A practical toolbox for modelling fibrosis in vitro.

Fibrosis is a widespread disease implicated in millions of deaths worldwide and impacting diverse organs, including the heart, lung, kidney, liver and synovium. Characterized by excessive extracellular matrix deposition, fibrosis leads to tissue scarring and dysfunction, ultimately resulting in organ failure. The development of faithful experimental models of fibrosis is challenged by its complexity, limiting the discovery of effective therapeutics. Human in vitro tissue models are emerging as powerful systems that capture the pathophysiology of fibrosis to uncover disease mechanisms, identify
