Neural Network Prediction of Response Factors for Extractables and Leachables in Pharmaceuticals and Medical Devices.

Ensuring the safety of patients using pharmaceuticals and medical devices through chemical characterization requires accurate estimation of extractables and leachables to ensure tolerable risk from unintentional exposure to these chemicals. However, this is complicated by variability in chemical responses, particularly with mass spectrometry methods. High-quality relative response factor predictions provide the opportunity for both expedited and more accurate quantitation in extractables and leachables analyses. In-silico prediction models were developed to test a wide variety of compounds, wh

End-to-end deep learning methods for genetic risk prediction of schizophrenia.

Schizophrenia is a highly heritable psychiatric disorder with a complex genetic basis. While recent GWAS and whole exome sequencing studies have identified numerous risk loci, existing clinical prediction models rely primarily on linear assumptions, limiting their ability to capture complex, nonlinear genetic effects such as epistasis. In this study, we apply end-to-end genome interpretation neural network models to predict schizophrenia risk using whole exome sequencing data in a cohort of 6,135 cases and 6,245 controls. We show that nonlinear neural networks significantly outperform conventi

Dynamic Prediction of Clinical Endpoints in Survival Analysis With Machine and Deep Learning: A Systematic Review and Regulatory Perspective.

Accurate prediction of time-to-event outcomes is essential for clinical decision-making and model-informed drug development (MIDD). Conventional survival models may inadequately represent complex longitudinal patient trajectories, time-dependent treatments, and evolving biomarker or pharmacokinetic/pharmacodynamic (PK/PD) profiles. Recent advances in machine learning (ML) and deep learning (DL) have enabled dynamic survival models that update individualized risk predictions as new longitudinal information becomes available. This report systematically reviews dynamic survival modeling framework

Physiological Data Analysis Framework for Pain Prediction in Physical Rehabilitation.

Predicting pain in physical rehabilitation is challenging due to subjectivity, patient variability, and self-report bias, especially in telerehabilitation. This study aims to determine whether machine-learning models based on heart rate (HR), heart rate variability (HRV), and peripheral oxygen saturation (SpO2) can reliably detect clinically meaningful pain during real rehabilitation sessions, including home-based settings where self-report is least reliable; we hypothesized that these low-cost, non-invasive markers carry sufficient information to flag low-to-moderate pain episodes without rel

Hybrid feature selection with novel deep learning model for COVID-19 risk prediction.

In the past five years, the COVID-19 epidemic has created unparalleled difficulties throughout the world. The COVID-19 virus has not entirely disappeared all over the world, and it is still spreading with new transformations and new variants throughout the globe. Hence, the requirement of exact and appropriate risk prediction can emphasize the care of patients and effectively allocate the resources for improving their lifespan. The prediction of risk factors for COVID-19 is highly needed for minimizing these hazards. As a result, this work develops the Fuzzy-Deep Kronecker Network + Deep Recur

A Bayesian Learning Model for Joint Risk Prediction of Alcohol and Cannabis Use Disorders.

Substance use disorders (SUDs) are a serious public health concern in the United States. Alcohol and cannabis are two of the most widely used substances. For adolescent/youth users of alcohol or cannabis, we propose a joint Bayesian learning model to predict their risks of developing alcohol use disorder (AUD) and cannabis use disorder (CUD) in adulthood based on their personal risk factors. The model is trained on nationally representative longitudinal data from Add Health ( n = 12,503 $$ n=\mathrm{12,503} $$ ). It consists of sub-models that predict the two SUDs for three groups of users-t

Incorporating global-local tissue changes to predict future breast cancer from longitudinal screening mammograms.

Early detection of breast cancer (BC) through mammography screening is critical for reducing mortality and improving patient outcomes. However, full-population-based, age-driven screening might not lead to optimal resource use and may enlarge screening associated harms in low risk women. Accurate and interpretable BC risk prediction is essential to improve strategies and make screening more personalized. Although recent deep learning models have shown promise in leveraging mammograms for risk stratification, challenges remain in interpretable modeling of temporal changes, efficiently capturing

Development of an interpretable machine learning model for predicting 4-year chronic kidney disease risk in elderly hypertensive patients.

INTRODUCTION: Age and hypertension are key drivers of renal impairment, predisposing older hypertensive adults to faster kidney function decline and higher mortality. We aim to develop an interpretable machinelearning model to predict 4-year chronic kidney disease (CKD) risk in this population. METHODS: Our study incorporated 4,142 hypertensive patients from the Health and Retirement Study (HRS) 2010 and 2012 cohorts for model development and internal validation, with additional temporal validation performed within the HRS 2006 and 2008 cohorts. External validation was conducted using three d

Structural brain imaging biomarkers for predicting seizure recurrence after a first unprovoked seizure.

OBJECTIVES: Predicting seizure recurrence following a first unprovoked seizure (FUS) remains a significant clinical challenge, especially when routine clinical magnetic resonance imaging (MRI) and EEG do not reveal abnormalities diagnostic of epilepsy. Here, we incorporate quantitative structural MRI-derived biomarkers into prediction models for seizure recurrence at 12 months and identify brain structural features that are predictive of seizure recurrence. METHODS: We analyzed a retrospective, multicenter cohort of 197 adult patients with FUS, comprising 83 with seizure recurrence and 114 wi

Predicting recurrence within 5 years in Early-Stage lung adenocarcinoma with micropapillary and solid patterns.

BACKGROUND: Micropapillary (MP) and solid (S) pathological patterns are recognized as high-risk patterns. Even early-stage invasive adenocarcinoma (IAC) with MP/S pathological patterns carry a high risk of postoperative recurrence. This study aimed to construct and validate machine learning (ML) models to predict recurrence within 5 years in patients with IAC with MP/S pathological components and tumor diameter ≤ 3 cm without lymph node metastasis and distant metastasis. METHODS: We retrospectively analyzed 974 patients from two centers. Univariate and multivariate logistic regression analyse

Real-World Application of a Machine Learning-Based Early Recurrence Model for Guiding Adjuvant Chemotherapy Use in Patients with Gallbladder Cancer.

BACKGROUND/AIMS: Although recent randomized controlled trials have reported the efficacy of adjuvant chemotherapy for resected biliary tract cancer, discrepancies remain between recommendations and real-world practice. Therefore, we aimed to assess the efficacy of adjuvant chemotherapy in patients with gallbladder cancer and used machine learning-based risk stratification to predict recurrence to avoid unnecessary chemotherapy. METHODS: Patients who underwent surgery between 2005 and 2022 and were histologically diagnosed with stage 2 or above gallbladder cancer were included. The patients we

Coagulation dysfunction as an independent predictor of early pregnancy loss in women with polycystic ovary syndrome: a machine learning-based retrospective cohort study.

OBJECTIVE: Current studies on adverse pregnancy outcomes in polycystic ovary syndrome (PCOS) have primarily focused on metabolic and endocrine abnormalities, whereas systematic investigations addressing live birth outcomes and coagulation dysfunction remain limited. In addition, there is a lack of comprehensive tools for early pregnancy risk stratification. This study aimed to explore the associations between early pregnancy indicators and early pregnancy loss (EPL) in women with PCOS, identify key risk factors, and subsequently develop a clinically interpretable predictive model to facilitate

Are Limited Electronic Medical Record Follow-Up Data Sufficiently Useful for Validating the Performance of Survival Prediction Models?

PURPOSE: Machine learning models that predict survival time for patients are increasingly used for clinical decision support. Validating model performance in deployment is important but challenging because the only timely source of follow-up/death data is the electronic medical record (EMR), which is known to undercapture deaths, resulting in informative censoring. We examined whether model performance evaluation using EMR data alone can distinguish between low- and high-quality models using high-quality cancer registry data combined with EMR data as a comparison to calculate model performance

Machine learning forecasts of laser intensity evolution through underwater turbulence.

This experimental study investigates whether the temporal evolution of received laser-beam intensity can be forecast from intensity images alone under controlled underwater turbulence. Conventional adaptive-optics systems commonly rely on present wavefront measurements and are therefore reactive to measured turbulence-induced distortions. We examine a different prerequisite problem: whether established deep-learning components can be assembled into short-horizon predictors of experimentally measured intensity evolution, without phase measurements or iterative wavefront reconstruction. Synchron

Automated Measurement of Depigmentation Extent with a New AI Tool Applied to the Example of Vitiligo.

INTRODUCTION: We have developed a digital algorithm to assess skin pigmentation, specifically an artificial intelligence-based image analysis tool that segments photographed lesions and then scores them by Facial Vitiligo Area Scoring Index (F-VASI), in place of trained site investigators. Vitiligo, the disease used in this exemplary demonstration of the algorithm, is a chronic, acquired, immune-mediated depigmentation disease characterized by white macules and/or patches of skin. The F-VASI is a clinician-reported outcome that relies on manual assessment of affected body surface area (BSA) an
