The Role of Fibro/adipogenic Progenitors (FAPs) in Sarcopenia: Mechanisms and Potential Therapeutic Strategies.

Sarcopenia is an age-related degenerative disease of skeletal muscles, characterized by progressively declined muscle mass, strength, and physical function. The prevalence of sarcopenia ranges from 11% to 50% among the global elderly population. The etiology of sarcopenia is multifactorial, including protein homeostasis imbalance, mitochondrial dysfunction, muscle satellite cells (MuSCs) dysfunction, chronic low-grade inflammation, and muscle interstitial remodeling. FAPs are mesenchymal-derived progenitor cells, playing crucial roles in maintaining muscle homeostasis and in the progression of

Mechanosensitive Ion Channel PIEZO1 Suppresses BMP2-Induced Ossification of the Annulus Fibrosus Cells.

OBJECTIVE: Major cause of low-back pain is intervertebral disc degeneration (IVDD), with mechanical stress playing a crucial role in its progression. A mechanosensitive ion channel, PIEZO1, is involved in various musculoskeletal tissues, but its role in the annulus fibrosus (AF) remains unclear. This study aimed to elucidate the function of PIEZO1 in AF cells under mechanical stimulation. METHODS: Primary rat AF cells were subjected to cyclic tensile strain (CTS) at low (2%) and high (12%) strain levels to investigate strain-dependent effects on osteogenic gene expression. We evaluated the ef

Molecular Mechanisms of Electric Signals Generated at Corneal Wounds.

Over 150 years ago, it was demonstrated that wounds produce electric fields and currents. Recent research provides strong evidence that these electric fields send a powerful signal to guide the migration of cells and epithelial sheets to heal. However, the mechanisms by which wounds generate and regulate the electric signals are not known. Here, we demonstrate that cornea wound electric currents are an actively regulated wound response. Using a systematic and hierarchical drug screening strategy, starting with general inhibitors for channels/pumps of sodium, chloride, potassium, and calcium ch

Imaging manifestations of malignancy in inflammatory bowel disease: patterns, pitfalls and pearls.

Patients with inflammatory bowel disease (IBD) are at increased risk of gastrointestinal and hepatobiliary malignancies arising in the setting of chronic inflammation, dysplasia, stricturing disease, and fistulizing complications. Although endoscopic surveillance remains the cornerstone for dysplasia detection, imaging plays an increasingly important complementary role in identifying invasive malignancy, evaluating transmural and extra-intestinal disease, and detecting lesions in segments inaccessible or poorly amenable to endoscopic assessment. Distinguishing malignant transformation from chr

AI-Driven Analysis Unveils Functional Dynamics of Müller Cells in Retinal Autoimmune Inflammation.

Müller cells are the most abundant glial cell type in the human and mouse retina, playing a crucial role in maintaining retinal homeostasis. However, many aspects of Müller cell function remain poorly characterized. In this study, we reanalyzed a single-cell RNA-seq (scRNA-seq) dataset from Aire-/- mice, focusing on Müller cells and T cells. We identified nine distinct Müller cell subgroups and five T cell subgroups, with activated Müller cells comprising the majority of the Müller cells in the inflamed retina. Using SCassist, an AI-based workflow assistant for single-cell analysis, we created

Interplay of PRMTs and Identification of Biomarkers Through Machine Learning Algorithms in Pan-Cancer, Highlighting PRMT3 as a Biomarker in Pancreatic Cancer.

Protein arginine methylation was a common post-translational modification, playing a key role in many biological processes and disease. But the regulatory mechanisms of protein arginine methyltransferases (PRMTs) in cancer were not well understood. This study aimed to examine the influence of PRMTs across various cancers and identify potential PRMTs-related prognostic biomarkers. Three machine learning algorithms were employed to identify potential PRMTs biomarkers across cancers. Additionally, drug prediction and molecular docking studies were carried out to discover new therapeutic targets.

Targeting biofilm formation in Candida albicans with halogenated pyrrolopyrimidine derivatives.

Growing concern over environmental contaminants, including pharmaceuticals and antifungal agents, highlights their role in promoting resistance and biofilm formation by microorganisms. Antifungal resistance, especially in drug-resistant Candida spp., poses a global threat, worsened by the widespread use of antifungal agents in both clinical applications and environmental contamination. This study investigates the antibiofilm properties of various halogenated pyrrolo pyrimidine derivatives, specifically 4-chloro-5-iodo-7H-pyrrolo[2,3-d]pyrimidine (10) and 2,4-dichloro-5-iodo-7H-pyrrolo[2,3-d]py

PDE10A as a novel diagnostic and therapeutic target in cancer: insights and challenges.

Phosphodiesterase 10A (PDE10A) is a dual-substrate enzyme that hydrolyzes both cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP), playing a critical role in regulating intracellular signaling pathways. While its function has been extensively studied in the central nervous system, emerging evidence highlights its broader physiological and pathological relevance, including its involvement in cancer. Functionally, it modulates key signaling pathways such as cAMP/protein kinase A (PKA) and cGMP/protein kinase G (PKG), influencing cell proliferation, differentiation, a

Divergent somatic mutation patterns among human cerebellar neuron types.

Neurons in the human brain accumulate somatic mutations with age. However, it is largely unknown how somatic mutation rates and patterns vary among the brain's diverse types of neurons. Characterizing this variability is critical for elucidating the role of genome integrity in human brain function and disease. Moreover, the significant physiological differences among the brain's cell types provides an opportunity to learn more general underlying factors that determine mutation rates and patterns. Here, we utilized high-fidelity duplex DNA sequencing to profile somatic mutation processes across

Non-muscle myosin IIC predominantly expressed in the slow-twitch skeletal muscles impedes age-related muscle weakness.

Skeletal muscle expresses three types of non-muscle myosin (NM) II in addition to skeletal type myosin. While immature myoblasts have been reported to express NMIIA and NMIIB, playing roles in cell morphology, the specific localization and function of NMIIC in skeletal muscle cells remain unclear. In this study, we aimed to investigate the expression pattern and the physiological role of NMIIC in skeletal muscle. NMIIC was specifically expressed in the slow-twitch muscles such as soleus, which primarily consists of type I and type IIa fibers, and its expression increased as muscle differentiat

Extracellular vesicles, syntaxin 2 and SNAP23 in the uterine microenvironment of the rat.

IN BRIEF: During uterine receptivity, exocytosis from uterine epithelial cells (UECs) contributes to uterine fluid composition, playing a role in communication with an implanting blastocyst. SNAREs, a family of proteins involved in exocytosis, are increased in the receptive uterine epithelium of the rat. ABSTRACT: Uterine luminal fluid is composed of secretions from the uterine luminal and glandular epithelial cells. The fluid composition plays a role in cell-to-cell communication between the receptive endometrium and an invading blastocyst. Part of this fluid is released from the epithelial
