TAF15 mediates ROP16-induced apoptosis and cell cycle arrest in lung cancer.

OBJECTIVE: This study aimed to investigate the effects of the interaction between ROP16 and TAF15 on apoptosis and cell cycle regulation in A549 lung adenocarcinoma cells. METHODS: Lentivirus-infected A549 cells which could overexpress type I, II, and III ROP16, along with an empty vector control group, were established. Potential interacting proteins with ROP16 were identified by using co-immunoprecipitation (Co-IP) and liquid chromatography-mass spectrometry (LC-MS). High-scoring interacting proteins were selected for verification through Venn diagram analysis, scoring, and intensity evalua

Imbalance of APOB Lipoproteins and Large HDL in Type 1 Diabetes Drives Atherosclerosis.

BACKGROUND: Individuals with type 1 diabetes (T1D) generally have normal or even higher HDL (high-density lipoprotein)-cholesterol levels than people without diabetes yet are at increased risk for atherosclerotic cardiovascular disease (CVD). Human HDL is a complex mixture of particles that can vary in cholesterol content by >2-fold. To investigate if specific HDL subspecies contribute to the increased atherosclerosis associated with T1D, we created mouse models of T1D that exhibit human-like HDL subspecies. We also measured HDL subspecies and their association with incident CVD in a cohort of

Differential short-term and long-term metabolic and cytokine responses to infection of severe fever with thrombocytopenia syndrome virus.

INTRODUCTION: Severe fever with thrombocytopenia syndrome (SFTS) is an emerging infectious disease caused by the SFTS virus (SFTSV), which has a wide geographic distribution. The primary clinical manifestations of SFTS are fever and thrombocytopenia, with multiorgan failure being the leading cause of death. While most patients recover with treatment, little is known about the potential long-term metabolic effects of SFTSV infection. OBJECTIVES: This study aimed to shed light on dysregulated metabolic pathways and cytokine responses following SFTSV infection, which pose significant risks to th

Identification of novel autoantigens as potential biomarkers in juvenile idiopathic arthritis associated uveitis.

BACKGROUND: Many children with juvenile idiopathic arthritis (JIA) have autoantibodies, targeting nuclear components (anti-nuclear antibodies, ANA). ANA in JIA is associated with uveitis, an eye inflammation which may cause permanent vision impairment if not detected and treated. However, ANA-testing is neither specific nor sensitive enough to be a clinically reliable predictor of uveitis risk, and the precise autoantigens targeted by ANA in JIA are largely unknown. If identified, specific autoantibodies highly associated with uveitis could be used as biomarkers to facilitate identification of

Evaluation of the effects of newly synthesized metallophthalocyanines on breast cancer cell lines with photodynamic therapy.

In this study, the new phthalonitrile derivative 3-(4-(3-oxobutyl)phenoxy)phthalonitrile (1) and its non-peripheral metallophthalocyanine derivatives [zinc (2), copper (3), cobalt (4), manganese (5), gallium (6), and indium (7)] were synthesized. The newly synthesized phthalocyanines were characterized by standard spectroscopic methods, such as FT-IR, 1H NMR, UV-Vis, fluorescence spectroscopies, and MALDI-TOF spectrometry. Aggregation behaviors of the novel phthalocyanines were investigated by UV-Vis spectroscopy. The effect of pH change on the electronic and emission spectra of the newly synt

In Situ Single Cell Proteomics Reveals Circulating Tumor Cell Heterogeneity during Treatment.

Cancer is a dynamic disease with heterogenic molecular signatures and constantly evolves during the course of the disease. Single cell proteomic analysis could offer a suitable pathway to monitor cancer cell heterogeneity and deliver critical information for the diagnosis, recurrence, and drug-resistant mechanisms in cancer. Current standard techniques for proteomic analysis such as ELISA, mass spectrometry, and Western blots are time-consuming, expensive, and often require fluorescence labeling that fails to provide accurate information about the multiple protein expression changes at the sin

PARP1 Enhances Influenza A Virus Propagation by Facilitating Degradation of Host Type I Interferon Receptor.

Influenza A virus (IAV) utilizes multiple strategies to confront or evade host type I interferon (IFN)-mediated antiviral responses in order to enhance its own propagation within the host. One such strategy is to induce the degradation of type I IFN receptor 1 (IFNAR1) by utilizing viral hemagglutinin (HA). However, the molecular mechanism behind this process is poorly understood. Here, we report that a cellular protein, poly(ADP-ribose) polymerase 1 (PARP1), plays a critical role in mediating IAV HA-induced degradation of IFNAR1. We identified PARP1 as an interacting partner for IAV HA throug

Photocatalytic Performance of CuxO/TiO₂ Deposited by HiPIMS on Polyester under Visible Light LEDs: Oxidants, Ions Effect, and Reactive Oxygen Species Investigation.

In the present study, we propose a new photocatalytic interface prepared by high-power impulse magnetron sputtering (HiPIMS), and investigated for the degradation of Reactive Green 12 (RG12) as target contaminant under visible light light-emitting diodes (LEDs) illumination. The CuxO/TiO₂ nanoparticulate photocatalyst was sequentially sputtered on polyester (PES). The photocatalyst formulation was optimized by investigating the effect of different parameters such as the sputtering time of CuxO, the applied current, and the deposition mode (direct current magnetron sputtering, DCMS or HiPIMS).

Immobilization of polyphosphoesters on poly(ether ether ketone) (PEEK) for facilitating mineral coating.

Poly(ether ether ketone) (PEEK) is an alternative material to metals for orthopedic applications. However, the compatibility of PEEK with hard tissues needs to be improved. To address this issue, this study proposes a novel technique for PEEK surface modifications. A polyphosphodiester macromonomer (PEPMA·Na) was synthesized via the demethylation of polyphosphotriester macromonomer obtained via the ring-opening polymerization of cyclic phosphoesters using 2-hydroxypropyl methacrylamide as the initiator. The surface modification of PEEK was performed via photoinduced and self-initiated graft po

Feasibility of using a novel optically filtered monochromatic portable X-ray fluorescence (MXRF) to quantify lead in wild bird bones.

Lead (Pb) threatens wildlife and humans through neurological, cardiovascular, and renal damage. Conventional tissue Pb assessment relies on costly wet-lab methods such as inductively coupled plasma mass spectrometry (ICP-MS). Monochromatic X-ray fluorescence (MXRF) enables rapid, non-destructive, field-deployable Pb detection with lower detection limits than prior portable XRF systems. We evaluated the feasibility, calibration, attenuation correction, and ICP-MS agreement of a doubly curved crystal (DCC)-enhanced MXRF (X-ray Optical Systems, East Greenbush, NY) for bone Pb quantification in Am

Tfcp2l1 participates in paternal vitamin D deficiency-induced fetal lung maldevelopment in mice.

BACKGROUND: Fetal lung development is crucial for neonatal respiratory function, with insufficient maturation contributing to neonatal respiratory distress syndrome. While maternal vitamin D deficiency (VDD) has been linked to adverse offspring outcomes, the impact of paternal VDD on fetal lung programming remains largely unexplored. This study investigates the effects of paternal VDD on fetal lung development and maturation, aiming to provide a theoretical basis for paternal preconception nutritional intervention and highlight the importance of paternal health in offspring respiratory outcome

LINC00491 promotes nasopharyngeal carcinoma metastasis by binding to purine-rich element binding protein alpha and regulating annexin-A1 to activate the PI3K/AKT pathway.

BACKGROUND: Distant metastasis and locoregional recurrence represent the predominant causes of therapeutic inefficacy in nasopharyngeal carcinoma (NPC), highlighting an urgent need for discovery of novel molecular targets. Current evidence suggests that LINC00491 is involved in the tumorigenesis and progression of pulmonary adenocarcinoma, pancreatic carcinoma, and other malignancies. Nevertheless, its functional role and precise molecular mechanisms in NPC remain to be fully elucidated. METHODS: Gene screening was performed through lncRNA sequencing. LINC00491 expression in NPC specimens was

Novel paracellular marker based on ᴅ-dipeptide structure for highly sensitive quantification with UPLC-MS/MS for in vitro and in vivo blood-brain barrier permeability analysis.

BACKGROUND: The blood-brain barrier (BBB) paracellular permeability must be evaluated in various contexts in vitro and in vivo, including pharmacological evaluation of drug candidates, investigations of pathological changes in disease and model development. However, most available paracellular marker substances are either radioactive, lack sensitivity and thereby require large sample volumes, or they can influence the BBB themselves via osmotic pressure. Moreover, in drug permeability studies, an adequate paracellular marker should be detectable in the same sample as the compound being tested,

Single cell autofluorescence imaging reveals immediate metabolic shifts of neutrophils with activation across biological systems.

INTRODUCTION: Neutrophils are critical innate immune cells that heterogeneously respond to infection and inflammation by performing functions such as oxidative burst and NETosis, which require significant metabolic adaptation. Deeper insights into the single cell diversity of such metabolic changes will help identify regulation of neutrophil functions in health and diseases. Due to their short lifespan and associated technical challenges, the early metabolic processes of neutrophil activation are not completely understood. New tools are needed to measure rapid changes in neutrophil metabolism

Mechanistic insights into aniline-induced liver injury: Role of the mmu_circ_26984/Myh9/NLRP3 axis and modulation by N-acetylcysteine.

Aniline is a widely used chemical. Chronic or high-dose exposure to aniline can lead to hepatocellular damage. Although the hepatic pathogenicity of aniline has been established in previous studies, studies involving pathogenic genes during aniline-induced liver injury are limited. Our study first discovered and identified the role and mechanism underlying a new circRNA mmu_circ_26984 in aniline-induced chemical liver injury. Further, we discuss the protective effect of N-acetylcysteine (NAC) in this pathway. After constructing in vitro and in vivo models of aniline treatment, we screened the
