A novel magnetic-driven tissue retraction device for minimally invasive surgery.

INTRODUCTION: The purpose of this work is to design and validate an innovative magnetic-based device for tissue retraction for minimally invasive surgery. MATERIAL AND METHODS: An intra-abdominal magnetic module is coupled with an extracorporeal system for establishing a stable attraction, and consequently a reliable tissue retraction. Once the retractor has been inserted into the abdomen, tissue retraction is not constrained by a fixed access port, thus guaranteeing a more flexible, safer and less invasive operation. The intra-abdominal unit is composed of an axial permanent magnet attached

Nanofibrous nerve guidance conduits decorated with decellularized matrix hydrogel facilitate peripheral nerve injury repair.

Rationale: Peripheral nerve injury (PNI) is a great challenge for regenerative medicine. Nerve autograft is the gold standard for clinical PNI repair. Due to its significant drawbacks, artificial nerve guidance conduits (NGCs) have drawn much attention as replacement therapies. We developed a combinatorial NGC consisting of longitudinally aligned electrospun nanofibers and porcine decellularized nerve matrix hydrogel (pDNM gel). The in vivo capacity for facilitating nerve tissue regeneration and functional recovery was evaluated in a rat sciatic nerve defect model. Methods: Poly (L-lactic acid

Effects of connective tissue growth factor on the regulation of elastogenesis in human umbilical cord-derived mesenchymal stem cells.

BACKGROUND: A key to clinical microtia reconstruction is construct flexibility. The most significant current limitation to engineered elastic cartilage is maintaining an elastic phenotype, which is principally dependent on elastin production (although other parameters, including maintenance of a ratio above 1 for collagens II to I, minimizing collagen X content, and presence of adequate matrix fibrillin for elastin binding, all play supporting roles). Connective tissue growth factor (CTGF), a compound secreted by chondrocytes, has been shown to promote an elastic phenotype in mature rabbit cho

Sources of adult mesenchymal stem cells applicable for musculoskeletal applications - a systematic review of the literature.

Mesenchymal stem cells (MSCs) were first discovered by Friedenstein and his colleagues in 1976 from bone marrow. The unique property of these cells was their potential to develop into fibroblastic colony forming cells. Since Friedenstein's discovery of these cells the interest in adult MSCs has been progressively growing. Nowadays MSCs are defined as undeveloped biological cells capable of proliferation, self renewal and regenerating tissues. All these properties of MSCs have been discovered in the past 35 years. MSCs can play a crucial role in tissue engineering, organogenesis, gene therapy, …

[Polishing up functional dermatopathology, dermometrology and mechanobiology].

Dermatopathology is a tool using the microscopic examination for diagnosing cutaneous diseases. The frontiers of its field of interest were extended by exploring the physiopathology of skin functional properties. An example is provided by the evaluation of alterations of skin suppleness in various connective tissue disorders. This approach takes profit from both conventional histology and histochemistry. Immunohistology targeting dermal dendrocytes reveals their various shapes reflecting the modifications in tensegrity under the control of mechanobiology. The latter property is an indicator of

Microinjection molded PLA/ZnO-TiO2 composites with in situ fibrillation: achieving mechanical matching cortical bone and enhanced osteoinduction.

Polylactic acid (PLA) has emerged as a cornerstone material in bone tissue engineering due to its biocompatibility and processability. Nevertheless, critical limitations persist, including thermal deformation susceptibility and inadequate osteoinductive capacity, which collectively hinder its clinical translation for load-bearing applications. Herein, we engineer a microinjection molded PLA@ZnO-TiO2 composite bone fixation plate that simultaneously addresses these challenges through innovative interfacial engineering. The high shear stress during microinjection processing induces in situ fibri…

Transposon-derived genome editors in plants: from compact nucleases to large-fragment integration and regeneration strategies.

CRISPR/Cas technology has been transformative for genome editing, enabling the precise integration of large DNA fragments-a capability essential for advanced genome (re)writing in plants, such as for trait stacking and pathway engineering. However, several new RNA-guided systems derived from transposable elements (TEs) have shown promise for plant genome editing, thereby enriching the toolkit available for plant engineering. This review comprehensively analyzes these emerging TE-based editors, including OMEGA nucleases (TnpB, IscB, Fanzor), CRISPR-associated transposases (CASTs), and R2 retrot

The evolving landscape of antibody-based cancer therapies: From monospecific to multi-specific and beyond.

The landscape of antibody-based cancer therapies has evolved dramatically since the first monoclonal antibody (mAb) was approved for cancer treatment three decades ago. Advances in recombinant DNA and molecular engineering have transformed monospecific mAbs into multi-specific and multi-modal constructs, revolutionizing oncology. Unlike traditional mAbs, next-generation antibodies, including bispecific antibodies, antibody-drug conjugates (ADCs), and radionuclide-antibody conjugates (RACs), enable synergistic targeting of tumor-associated antigens (TAAs), immunomodulatory receptors, and immune

Harnessing exosomes for advanced osteoarthritis therapy.

Exosomes are nanosized, lipid membrane vesicles secreted by cells, facilitating intercellular communication by transferring cargo from parent to recipient cells. This capability enables biological crosstalk across multiple tissues and cells. Extensive research has been conducted on their role in the pathogenesis of degenerative musculoskeletal diseases such as osteoarthritis (OA), a chronic and painful joint disease that particularly affects cartilage. Currently, no effective treatment exists for OA. Given that exosomes naturally modulate synovial joint inflammation and facilitate cartilage ma

Engineering approaches for RNA-based and cell-based osteoarthritis therapies.

Osteoarthritis (OA) is a chronic, debilitating disease that substantially impairs the quality of life of affected individuals. The underlying mechanisms of OA are diverse and are becoming increasingly understood at the systemic, tissue, cellular and gene levels. However, the pharmacological therapies available remain limited, owing to drug delivery barriers, and consist mainly of broadly immunosuppressive regimens, such as corticosteroids, that provide only short-term palliative benefits and do not alter disease progression. Engineered RNA-based and cell-based therapies developed with syntheti
