Gastrointestinal microbiota alteration induced by Mucor circinelloides in a murine model.

Mucor circinelloides is a pathogenic fungus and etiologic agent of mucormycosis. In 2013, cases of gastrointestinal illness after yogurt consumption were reported to the US FDA, and the producer found that its products were contaminated with Mucor. A previous study found that the Mucor strain isolated from an open contaminated yogurt exhibited virulence in a murine systemic infection model and showed that this strain is capable of surviving passage through the gastrointestinal tract of mice. In this study, we isolated another Mucor strain from an unopened yogurt that is closely related but dis

Quantifying Enteropathogen Contamination along Chicken Value Chains in Maputo, Mozambique: A Multidisciplinary and Mixed-Methods Approach to Identifying High Exposure Settings.

BACKGROUND: Small-scale poultry production is widespread and increasing in low- and middle-income countries (LMICs). Exposure to enteropathogens in poultry feces increases the hazard of human infection and related sequela, and the burden of disease due to enteric infection in children <5 y in particular is substantial. Yet, the containment and management of poultry-associated fecal waste in informal settings in LMICs is largely unregulated. OBJECTIVES: To improve the understanding of potential exposures to enteropathogens carried by chickens, we used mixed methods to map and quantify microbia

Mercury poisoning in women and infants inhabiting the Gangetic plains of Bihar: risk assessment.

Mercury is a persistent, bio-accumulative, and hazardous contaminant. When released into the environment, it accumulates in water sediments, converting it to poisonous methylmercury that enters the food chain. The present study was carried out in habitations from the 11 districts of Bihar (India). In the study, n = 224 lactating women and their infants n = 172 infants participated. After obtaining the written informed consent, their breast milk, urine, and blood were collected for mercury estimation. The breastmilk content was measured in n = 181 subjects, in which 74% women had their breastmi

Electrochemical identification of riboflavin-mediated redox adaptation in Escherichia coli under mild heat stress.

Understanding the stress response mechanisms of Escherichia coli (E. coli) is essential for controlling its contamination in food systems. In this study, the adaptive response of E. coli to mild heat treatment (MHT) was investigated from an electrochemical perspective, with a focus on energy metabolism and electron transport processes. A distinct oxidation peak at -0.45 V (vs Ag/AgCl) was identified and attributed to riboflavin (RF) through differential pulse voltammetry (DPV), HPLC, and LC-MS/MS analyses. Under MHT condition, the RF-associated electrochemical signal was significantly enhanced

Achieving Highly Sensitive In Situ Detection of Trace Starch with a Hydrogel-Functionalized MZI Optical Fiber Biosensor.

To address the critical concern about trace starch residues in cleaning water within the food industry, which can lead to cross-contamination and water eutrophication, a label-free optical fiber biosensor designed for high-sensitivity, specific in situ detection of starch was proposed. The sensor was constructed as an Multimode-No-core-Multimode (MNM) structure by cascading multimode fiber (MMF) and no-core fiber (NCF), which was functionalized with a hydrogel-based sensing layer. A key innovation involved the covalent immobilization of α-amylase onto the NCF surface via a glutaraldehyde-CS cr

Revealing spatiotemporal distribution of aflatoxin B1-related health burdens to optimize monitoring in peanut supply chains.

Aflatoxin B1 (AFB1), one of the most hazardous environmental and foodborne mycotoxins, is a major contributor to the global burden of hepatocellular carcinoma owing to its potent hepatocarcinogenicity and persistence throughout food systems. Although risk assessment and monitoring programs for aflatoxins have been established worldwide, the dynamic patterns of AFB1-related disease burden from production to consumption have been largely overlooked, limiting the development of truly risk-based monitoring strategies. Here, we present an integrated framework that quantifies seasonal and regional o

HSI-AgriFoodAnomaly, a hyperspectral dataset for foreign object detection in agri-food inspection.

Ensuring food safety in agri-food production requires reliable inspection data for detecting foreign objects and other contamination risks. Hyperspectral imaging (HSI) is a relevant non-destructive modality for this purpose because it combines spatial information with detailed spectral responses. This Data Descriptor presents HSI-AgriFoodAnomaly, an open hyperspectral image dataset acquired under conveyor-based industrial-like conditions for foreign object annotation in an oat, white-chocolate and dark-chocolate mixture. The dataset contains 147 calibrated hyperspectral cubes, associated red-g

Determination of Pyrrolizidine Alkaloids and Their N‑Oxides in Bee Pollen by Accelerated Solvent Extraction Followed by Magnetic Covalent Organic Framework-Based Offline Purification and Ultra-High-Performance Liquid Chromatography-Tandem Mass Spectrometry.

Pyrrolizidine alkaloids (PAs) and their N-oxides (PANOs) are naturally occurring toxic plant metabolites that have attracted widespread attention due to their potential food safety hazards. In this study, an efficient analytical method for determining PAs/PANOs in bee pollen was established by combining accelerated solvent extraction (ASE) with ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). A magnetic covalent organic framework material (Fe3O4@COF-TAPB-TFP) was synthesized and used as an adsorbent for offline purification to reduce matrix interference and

Sustainable cellulose acetate beads impregnated with eugenol, thymol, β-citronellol, and their mixtures via supercritical carbon dioxide for microbial and pest insect control.

Post-harvest losses of legumes caused by microbial contamination and pest insects represent a major challenge for food security and sustainable storage systems. In this study, biodegradable and bio-based cellulose acetate beads (CAB) were developed as functional materials for bean protection using an environmentally benign supercritical impregnation process. Eugenol, thymol, β-citronellol, and their mixtures were incorporated into CAB, achieving loadings of up to 7.5 wt% within a 3 h process. Impregnation resulted in a pronounced reduction in average pore size, from approximately 0.5 to 0.2 μm

Climate-stressed crop mycobiomes as pre-harvest sentinels of human mycotoxin exposure: an AI-enabled One Health framework linking dysbiosis, exposomics, and toxicopathology.

Climate change is altering the conditions under which crop-associated fungi colonize plants and produce mycotoxins. Current control relies on both pre-harvest management and post-harvest testing; however, field-level fungal-community changes are rarely used to guide where and when testing should be intensified. This Review critically compares evidence linking weather and crop stress, fungal-community composition, toxigenic strain activity, contamination during harvest and storage, human biomarkers, and organ-specific effects. On this basis, we propose the Crop Mycobiome-Exposome-Toxicopatholog

Precise, Sensitive Detection of Viable Foodborne Pathogenic Bacteria with a 6-Order Dynamic Range via Digital Rolling Circle Amplification.

The presence of viable pathogenic bacteria in food can lead to serious foodborne diseases, thus posing a risk to human health. Here, we develop a digital rolling circle amplification (dRCA) assay that enables the precise and sensitive quantification of viable foodborne pathogenic bacteria. Directly targeting pathogenic RNAs via a ligation-based padlock probe allows for precisely discriminating viable bacteria from dead one. The one-target-one-amplicon characteristic of dRCA enables high sensitivity and a broad quantitative detection range, conferring a detection limit of 10 CFU/mL and a dynami

Everything but the kitchen sink: The use of multiple hypothesis generation methods to investigate an outbreak of Salmonella Enteritidis associated with frozen profiteroles and eclairs.

In December 2018, an outbreak of Salmonella Enteritidis infections was identified in Canada by whole-genome sequencing (WGS). An investigation was initiated to identify the source of the illnesses, which proved challenging and complex. Microbiological hypothesis generation methods included comparisons of Salmonella isolate sequence data to historical domestic outbreaks and international repositories. Epidemiological hypothesis generation methods included routine case interviews, open-ended centralized re-interviewing, thematic analysis of open-ended interview data, collection of purchase recor

Quantitative and Fast Sterility Assurance Testing of Surfaces by Enumeration of Germinable Endospores.

A fast Endospore Germinability Assay (EGA) was validated with traditional plate counts to enumerate single endospore germination events for monitoring surface sterilization. The assay is based on a time-gated luminescence microscopy technique enabling visualization and enumeration of individual germinating endospores. Germinating endospores release calcium dipicolinate to form highly luminescent terbium dipicolinate complexes surrounding each germinating endospore. EGA and heterotrophic plate counting (HPC) were used to evaluate the swab/rinse recovery efficiency of endospores from stainless s

Discharge of swine wastes risks water quality and food safety: Antibiotics and antibiotic resistance genes from swine sources to the receiving environments.

Swine feedlots are widely considered as a potential hotspot for promoting the dissemination of antibiotic resistance genes (ARGs) in the environment. ARGs could enter the environment via discharge of animal wastes, thus resulting in contamination of soil, water, and food. We investigated the dissemination and diversification of 22 ARGs conferring resistance to sulfonamides, tetracyclines, chloramphenicols, and macrolides as well as the occurrence of 18 corresponding antibiotics from three swine feedlots to the receiving water, soil environments and vegetables. Most ARGs and antibiotics survive
