Cytotoxicity of monoclonal antibodies to Trypanosoma brucei.

Monoclonal antibodies (McAbs) were raised against Metacyclic Variable Antigen Types (M-VATs) of the AnTAR 1 and ETAR 1 serodemes of Trypanosoma brucei. Two dominant M-VATs, one from each serodeme, were labelled by two of the McAbs using the indirect immunofluorescence technique. These McAbs were of the IgM class, and labelled exposed epitopes on living trypanosomes. They showed lytic activity in vitro towards their respective homologous VAT trypanosomes, both in the presence and absence of complement. In vivo, the McAbs promoted lysis and clearance of trypanosomes from the bloodstream of infec

Trypanosoma (Nannomonas) congolense: analysis by fluorescein-conjugated plant lectins of surface saccharides of cloned variant antigen types differing in infectivity for mice.

Surface saccharides of 4 cloned VATs (variant antigen types) of Trypanosoma (Nannomonas) congolense, AmNats (Amherst Nannomonas antigen types) 1.1, 1.2, 2.1, and 3.1, derived from 3 different stocks, were compared by fluorescein-conjugated, plant lectins using a quantitative fluorescence method. It was ascertained by the ID63 assay that the 4 AmNats differed in their infectivity for mice. The lectins employed for AmNats 1.1, 2.1, and 3.1 were concanavalin A (Con A), wheat germ agglutinin (WGA), soybean agglutinin (SBA), garden pea agglutinin (GPA), and gorse seed (Ulex europaeus) agglutinin (U

Histopathological findings in mini-pigs infected with different strains of Trypanosoma brucei.

Two pigs infected with Trypanosoma brucei gambiense developed low parasitemia which became undetectable after 6 months; on autopsy 13 months after infection they showed no histopathological alterations. --Five of six pigs infected with a Trypanosoma brucei brucei strain from the Ivory Coast developed low parasitemia (up to about antilog 6.5 per ml of blood) which became progressively lower but was detectable up to one year after infection. On autopsy they showed interstitial myocarditis and meningo-encephalitis; during the course of the infection, the first became milder, the second more inten

Regulation of the growth and differentiation of Trypanosoma (Trypanozoon) brucei brucei in resistant (C57Bl/6) and susceptible (C3H/He) mice.

While Trypanosoma brucei brucei GUTat 3 were equally infective for C3H/He and for C57Bl/6 mice at doses ranging from 5 to 5 x 10(3) organisms and had similar prepatent periods in both strains of mice, infected C57Bl/6 mice displayed lower parasitaemia, shorter times to parasite wave remission and survived for a longer time than infected C3H/He mice. Parasite growth and differentiation rates and host immune responses were similar for the first 5 days in both strains of mice after infection with 10(3) T.b.brucei GUTat 3 but, thereafter, parasite differentiation proceeded more rapidly and specifi

Infections with blood parasites in the small British rodents Apodemus sylvaticus, Clethrionomys glareolus and Microtus agrestis.

Three populations of small wild British rodents were studied by capture-recapture methods over a period of 3 years, a fourth group was studied for 1 year and a fifth was sampled annually for 4 years. Blood smears were taken from 3 species of rodents: the woodmouse Apodemus sylvaticus, the bank vole Clethrionomys glareolus (and an island sub-species C.g. skomerensis) and the short-tailed vole Microtus agrestis. The smears were examined microscopically. Four genera of haemoparasites Babesia, Hepatozoon, Trypanosoma and Grahamella were detected. Babesia was absent from C.g. skomerensis, Hepatozoo

Characterization of novel Trypanosoma species from birds and amphibians in the Colombian dry forest using an integrative taxonomic approach.

Trypanosomatids are flagellated protozoa that infect a wide range of hosts. While Trypanosoma and Leishmania are extensively studied due to their medical importance, information regarding species infecting wildlife remains scarce. This study aimed to characterize trypanosomatids from Colombian amphibians, birds, and mammals using an integrative taxonomic approach. We sampled 46 birds, 18 reptiles and amphibians, and 124 mammals within the tropical dry forest of Tolima, Colombia. Detection was performed employing novel Hsp70 (671 bp) and 18S rRNA (1,400-1,900 bp) primers. Positive PCR products

Differential Virulence and Pathogenicity of Canine Drug-Resistant Trypanosoma brucei brucei Field Isolates in Experimental Murine Infection.

PURPOSE: African animal trypanosomosis remains a significant veterinary and economic threat in sub-Saharan Africa, worsened by rising multidrug resistance. In dogs, Trypanosoma brucei brucei infections are often acute and fatal, yet the relationship between drug resistance and pathogenicity remains poorly defined. This study aimed to compare the virulence and pathogenicity of three canine multidrug-resistant T. b. brucei field isolates from Southeast Nigeria using an experimental murine model. METHODS: Twenty Sprague Dawley rats were randomly assigned to four groups (n = 5). Groups I-III rece

Survey on the prevalence of trypanosomosis in small ruminants and on tsetse distribution in Dedesa and Bedele districts, Buno Bedele Zone, Ethiopia.

BACKGROUND: African animal trypanosomosis (AAT) is one of the most serious diseases with ongoing detrimental effects on animal health and food production. METHODS: A cross-sectional study was conducted in the Bedele and Dedesa districts of Buno Bedele Zone, Southwest Ethiopia, to determine the prevalence of trypanosomosis and its vector distributions in small ruminants. Blood samples collected from a total of 384 small ruminants were examined for trypanosomosis via hematological analysis. For the tsetse distribution survey, 72 traps were deployed in the designated districts. RESULTS: Among t

A new automated chilled adult release system for the aerial distribution of sterile male tsetse flies.

Tsetse eradication continues to be a top priority for African governments including that of Senegal, which embarked on a project to eliminate Glossina palpalis gambiensis from the Niayes area, following an area-wide integrated pest management approach with an SIT component. A successful SIT programme requires competitive sterile males of high biological quality. This may be hampered by handling processes including irradiation and the release mechanisms, necessitating continued improvement of these processes, to maintain the quality of flies. A new prototype of an automated chilled adult releas

In vitro evaluation of arylsubstituted imidazoles derivatives as antiprotozoal agents and docking studies on sterol 14α-demethylase (CYP51) from Trypanosoma cruzi, Leishmania infantum, and Trypanosoma brucei.

There is an urgent need to discover and develop new drugs to combat parasitic diseases as Chagas disease (Trypanosoma cruzi), sleeping sickness (Trypanosoma brucei), and leishmaniasis (Leishmania ssp.). These diseases are considered among the 13 most unattended diseases worldwide according to the WHO. In the present work, the synthesis of 14 arylsubstituted imidazoles and its molecular docking onto sterol 14α-demethylase (CYP51) was executed. In addition, the compounds, antiprotozoal activity against T. brucei, T. cruzi, Trypanosoma brucei rhodesiense, and Leishmania infantum was evaluated. In

Trypanosoma brucei infection protects mice against malaria.

Sleeping sickness and malaria are parasitic diseases with overlapping geographical distributions in sub-Saharan Africa. We hypothesized that the immune response elicited by an infection with Trypanosoma brucei, the etiological agent of sleeping sickness, would inhibit a subsequent infection by Plasmodium, the malaria parasite, decreasing the severity of its associated pathology. To investigate this, we established a new co-infection model in which mice were initially infected with T. brucei, followed by administration of P. berghei sporozoites. We observed that a primary infection by T. brucei

Oligopeptide Signaling through TbGPR89 Drives Trypanosome Quorum Sensing.

Trypanosome parasites control their virulence and spread by using quorum sensing (QS) to generate transmissible "stumpy forms" in their host bloodstream. However, the QS signal "stumpy induction factor" (SIF) and its reception mechanism are unknown. Although trypanosomes lack G protein-coupled receptor signaling, we have identified a surface GPR89-family protein that regulates stumpy formation. TbGPR89 is expressed on bloodstream "slender form" trypanosomes, which receive the SIF signal, and when ectopically expressed, TbGPR89 drives stumpy formation in a SIF-pathway-dependent process. Increased genetic diversity and prevalence of co-infection with Trypanosoma spp. in koalas (Phascolarctos cinereus) and their ticks identified using next-generation sequencing (NGS).

Infections with Trypanosoma spp. have been associated with poor health and decreased survival of koalas (Phascolarctos cinereus), particularly in the presence of concurrent pathogens such as Chlamydia and koala retrovirus. The present study describes the application of a next-generation sequencing (NGS)-based assay to characterise the prevalence and genetic diversity of trypanosome communities in koalas and two native species of ticks (Ixodes holocyclus and I. tasmani) removed from koala hosts. Among 168 koalas tested, 32.2% (95% CI: 25.2-39.8%) were positive for at least one Trypanosoma sp. P
