Indium-mediated reduction of hydroxylamines to amines.

[reaction: see text] A novel and simple procedure for reduction of hydroxylamines to the corresponding amines by means of indium powder in aqueous media is reported. Applicability to one-pot reactions and isoxazolidine N-O bond reduction is also demonstrated. A catalytic version of the process using 2-5% In in the presence of other metals (Zn, Al) has been successfully developed.

Urinary amines in adults with Tourette's syndrome.

Urinary amines and their metabolites were examined in 32 adults who met DSM-III-R diagnostic criteria for Tourette's Syndrome. These patients were compared with a control group that was of similar age and sexual representation. Analyses revealed significantly lower levels of 3-methoxy-4-hydroxyphenylglycol and serotonin as well as the metabolites of several "trace" amines including indoleacetic acid and m- and p-hydroxyphenylacetic acid. These findings persisted when Tourette's Syndrome patients taking medications were eliminated from the analyses. These data are consistent with reports of neu…

[Biogenic amines in the digesta of swine].

Concentrations of amines were analysed in the digesta of piglets and fattening pigs after refining using an aminoacid-analyser. Feed contained only small amounts of putrescine. In the stomach histamine, tyramine, putrescine and cadaverine were nearly equally distributed, whereas in the colon cadaverine presented the main part. Piglets with 24% CP had in stomach/small intestine/hindgut 4.2/10.2/9.6 mmol, with 18% CP-/5.5/4.9 mmol amines/kg DM digesta. Fattering pigs showed in jejunal digesta from 2.1 to 8.7 mmol, in the caecum 1 to 4.8 mmol and in the colon 0.2 to 2.2 mmol amines/kg DM. Increas…

Amines and choline acetyltransferase in rat intestine.

A biochemical study of the endogenous levels of serotonin (5-HT), noradrenaline (NA) and the activity of choline acetyltransferase (CAT) was carried out in the intestinal tract of the rat. High levels of 5-HT and NA were detected in the caecum and the colon. These anatomical regions also presented the highest activity of CAT. Similar activities of CAT were detected, after dissection, in the mucosa and the muscular layers containing the enteric plexuses. During the day-night cycle, 5-HT and NA amounts showed significant variations as a function of time. Treatment with pargyline (75 mg kg(?1)),

Catalytic Enantioselective Nucleophilic Amination of α-Halo Carbonyl Compounds with Free Amines.

Catalytic enantioselective substitution of the readily available racemic α-halo carbonyl compounds by nitrogen nucleophiles represents one o … symmetric control over the key positively charged intermediates. This protocol not only allows free amines to serve as nucleophiles but also permits different types of carbonyl compounds (ketones, esters, and amides) to participate in the enantioselective C-N bond formation, thereby providing a valuable complement to the known strategies that are limited …

Regioselective copper-catalyzed amination of bromobenzoic acids using aliphatic and aromatic amines.

A chemo- and regioselective copper-catalyzed cross-coupling procedure for amination of 2-bromobenzoic acids is described. The method eliminates the need for acid protection and produces N-aryl and N-alkyl anthranilic acid derivatives in up to 99% yield. N-(1-Pyrene)anthranilic acid has been employed in metal ion-selective fluorosensing. Titration experiments showed that this pyrene-derived amino acid forms an equimolar complex with Hg(II) in water resulting in selective fluorescence quenching even in the presence of other metal ions such as Zn(II) and Cd(II).

A Direct-to-Biology Amine-Acid Amination for Rapid Phenotypic Profiling of Lysosomotropic Natural Products.

Natural products (NPs) are foundational to drug discovery, owing to their diverse molecular scaffolds and novel bioactivities. However, optimizing NPs for optimal bioactivity and improved drug properties is hindered by material scarcity and labor-intensive purification processes. Here, we present an automated ultrahigh-throughput experimentation (ultraHTE) and direct-to-biology (D2B) platform that enables rapid structure-activity relationship (SAR) mapping from miniaturized libraries of synthetic NP analogues, unlocking NPs as a viable starting point for late-stage diversification in drug-lead

Palladium-Catalyzed Amination of Protected Phenylalanine Iodides Using (Hetero)aryl Amines as Nucleophiles.

We report a robust palladium-catalyzed C-N cross-coupling method optimized for selective (hetero)arylation of derivatives containing p-iodophenylalanine (p-I-Phe). The utility of this approach is demonstrated by its use in late-stage modification of clinically relevant peptides in solution-phase. The method preserves the stereochemical integrity of the peptide backbone and exhibits compatibility with side-chain protecting groups. This provides a streamlined synthetic gateway to p-amino-substituted phenylalanines and peptides, expanding the chemical space accessible for peptide-based drug desig

Formal Carbon-Atom Insertion of Primary Aliphatic Amines.

Single carbon-atom insertion represents a powerful molecular-editing strategy for homologation and controlled carbon-chain growth while preserving preexisting functional groups. Despite their ubiquity in bioactive chemical space, primary aliphatic amines─including amino acids─remain challenging substrates for minimal chain expansion. Here, we report a general and iterative strategy that enables formal one-carbon insertion into primary aliphatic amines. The transformation proceeds through in situ generation of triazanium intermediates from commercially available O-diphenylphosphinyl hydroxylami…

Programmable remodelling of carbon-nitrogen connectivity in amines.

Tertiary amines are ubiquitous motifs in biologically active molecules, in which they play central parts in molecular recognition and function1,2. Among these, tertiary benzylamines are particularly prominent in discovery chemistry as readily assembled starting points for lead generation from abundant building blocks3,4, with favourable physicochemical and binding properties. However, despite their accessibility, the core C-N connectivity of these frameworks remains effectively locked5,6, confining diversification to benzylamine derivatives and preventing direct access to structurally distinct…

Copper-Mediated Dehydroamination of Primary Amines for Olefination.

Herein, we introduce a versatile and effective copper-mediated approach for the dehydroamination of a wide range of primary amines encompassing complex bioactive compounds. This method facilitates the transformation of primary amines into olefins under mild conditions, proceeding through a distinctive reaction mechanism. The process involves the formation of the monosubstituted isodiazene intermediate, which undergoes copper-mediated oxidative elimination to yield the corresponding alkene.

Tracking Aromatic Amines from Sources to Surface Waters.

This review examines the environmental occurrence and fate of aromatic amines (AAs), a group of environmental contaminants with possible carcinogenic and mutagenic effects. AAs are known to be partially responsible for the genotoxic traits of industrial wastewater (WW), and AA antioxidants are acutely toxic to some aquatic organisms. Still, there are gaps in the available data on sources, occurrence, transport, and fate in domestic WW and indoor environments, which complicate the prevention of adverse effects in aquatic ecosystems. We review key domestic sources of these compounds, including c…

Potential Role of Heterocyclic Aromatic Amines in Neurodegeneration.

Heterocyclic aromatic amines (HAAs) are mainly formed in the pyrolysis process during high-temperature cooking of meat. Meat consumption is very typical of the western diet, and the amount of meat consumption in the eastern countries is growing rapidly; HAAs represents widespread exposure. HAAs are classified as possible human carcinogens; numerous epidemiological studies have demonstrated regular consumption of meat with HAAs as risk factor for cancers. Specific HAAs have received major attention. For example, 2-amino-1-methyl-6-phenylimidazo[4,5-b] pyridine has been extensively studied as a …
