D-Amino acids in mammalian endocrine tissues.

D-Aspartate, D-serine and D-alanine are a regular occurrence in mammalian endocrine tissues, though in amounts varying with the type of gland. The pituitary gland, pineal gland, thyroid, adrenal glands and testis contain relatively large amounts of D-aspartate in all species examined. D-alanine is relatively abundant in the pituitary gland and pancreas. High levels of D-serine characterize the hypothalamus. D-leucine, D-proline and D-glutamate are generally low. The current knowledge of physiological roles of D-amino acids in endocrine tissues is far from exhaustive, yet the topic is attractin…

Amino Acids in Circulatory Function and Health.

Cardiovascular disease is the major cause of global mortality and disability. Abundant evidence indicates that amino acids play a fundamental role in cardiovascular physiology and pathology. Decades of research established the importance of L-arginine in promoting vascular health through the generation of the gas nitric oxide. More recently, L-glutamine, L-tryptophan, and L-cysteine have also been shown to modulate vascular function via the formation of a myriad of metabolites, including a number of gases (ammonia, carbon monoxide, hydrogen sulfide, and sulfur dioxide). These amino acids and t…

Hydration of copper(II) amino acids complexes.

Hydration of the copper(II) bis-complexes with glycine, serine, lysine, and aspartic acid was studied by DFT and MD simulation methods. The distances between copper(II) and water molecules in the 1st and 2nd coordination shells, the average number of water molecules and their mean residence times in the hydration shells were calculated. Good agreement was observed between the values obtained and those found by DFT and NMR relaxation methods. Influence of the functional groups of the ligands and the cis-trans isomerism of the complexes on the structural and dynamical parameters of the hydration

Branched-Chain Amino Acids and Brain Metabolism.

This review aims to provide a historical reference of branched-chain amino acid (BCAA) metabolism and provide a link between peripheral and central nervous system (CNS) metabolism of BCAAs. Leucine, isoleucine, and valine (Leu, Ile, and Val) are unlike most other essential amino acids (AA), being transaminated initially in extrahepatic tissues, and requiring interorgan or intertissue shuttling for complete catabolism. Within the periphery, BCAAs are essential AAs and are required for protein synthesis, and are key nitrogen donors in the form of Glu, Gln, and Ala. Leucine is an activator of the…

Antiirritant properties of polyols and amino acids.

BACKGROUND: Antiirritants are used in cosmetic products to prevent or to treat skin irritations that arise during daily life. Data were published earlier on the efficacy of the best-known humectant, glycerol, in reducing irritation. OBJECTIVE: The aim of the present study was to examine the effects of different polyols (including glycerol, xylitol, and mannitol) and the amino acids taurine and glycine on sodium lauryl sulfate (SLS)-induced skin irritation. METHODS: Healthy adult volunteers were patch-tested with 0.1% SLS in the presence or absence of one or another polyol or amino acid. Skin…

Branched-chain amino acids and brain function.

Branched-chain amino acids (BCAAs) influence brain function by modifying large, neutral amino acid (LNAA) transport at the blood-brain barrier. Transport is shared by several LNAAs, notably the BCAAs and the aromatic amino acids (ArAAs), and is competitive. Consequently, when plasma BCAA concentrations rise, which can occur in response to food ingestion or BCAA administration, or with the onset of certain metabolic diseases (e.g., uncontrolled diabetes), brain BCAA concentrations rise, and ArAA concentrations decline. Such effects occur acutely and chronically. Such reductions in brain ArAA co…

Changes of essential amino acids in thyropathies.

Composition of free amino acids in serum of patients with thyrotoxicosis and hypothyroidism was studied by gas chromatography. In thyrotoxicosis the level of serum amino acids, especially the essential ones and several glucogenic amino acids, increased. The reverse was found to a lesser extent in hypothyroidism. Thyrotoxicosis exerted an increase and hypothyroidism a decrease in the value of the ratio of the sum of all essential amino acids versus the sum of all nonessential amino acids. The elevation of glucogenic amino acids in thyrotoxicosis is discussed with respect to hepatic function and…

Free amino acids in human tonsillar tissue.

Free amino acids in the tonsils of 20 individuals were measured column chromatographically. Those always found in readily detectable amounts included O-phosphoserine, taurine, O-phosphoethanolamine, aspartic acid, hydroxyproline, threonine, serine, glutamic acid, proline, glycine, alanine, alpha-amino-n-butyric acid, valine, cystine, methionine, isoleucine, leucine, tyrosine, phenylalanine, ornithine, gamma-amino-butyric acid, lysine, histidine, and arginine. Results were compared for three clinical pathological groups and for four age groups. Some abnormal values may result from the pathologi…

D-amino acid oxidase suppresses hepatocellular carcinoma via oxidizing D-amino acids.

BACKGROUND: Patient-derived organoids provide a powerful platform for elucidating mechanisms of drug resistance and tumor evolution in hepatocellular carcinoma (HCC) and identifying novel therapeutic targets. METHODS: Transcriptomic sequencing was used to compare gene expression patterns between organoid-forming and non-forming HCC tissues, as well as between sorafenib-resistant organoids and sorafenib-sensitive counterparts. The TCGA-liver hepatocellular carcinoma (LIHC) cohort was used to screen for the key molecular drivers of HCC evolution from the overlapping differentially expressed gen

Increased pine root exudation of phenolic acids and amino acids under drought.

Belowground carbon (C) allocation by trees, particularly through root exudation, is a key pathway influencing long-term C storage in forest … sition, increasing several metabolites, mainly phenolic acids (e.g., gallic acid and caffeic acid), amino acids and key osmoprotectants such as trehalose, proline and betaine. Overall, these observations show that pines strongly increased their root exudation under drought and that exudate metabolites shifted towards more specialized metabolites linked to str …

Photofragmentation of C,N-protected alpha-amino acids: comparing tert-leucine with sulfur-containing amino acids methionine and cysteine.

The photochemical fingerprint for the N-acetyl methyl ester of the aliphatic amino acid tert-leucine 1 was investigated. This reaction path was compared with the electron transfer active amino acids methionine (N-acetyl methyl ester derivative 2a as well as the methyl ester derivative 2b) and the cysteine derivatives 3a and 3b (penicillamine derivative). Photofragmenation of the ester group dominated the photolysis of 1, whereas loss of methylmercaptane was observed for 2a and 3a. Vinylglycine derivatives 11 and 14 were formed to a minor extent. The gem-dimethylated compound 3b gave a solvent-…
