First Anti-Inflammatory Peptide AnmTX Sco 9a-1 from the Swimming Sea Anemone Stomphia coccinea.

A novel peptide AnmTX Sco 9a-1 with the β-hairpin fold was isolated from the swimming sea anemone Stomphia coccinea (Actinostolidae family). The peptide consists of 28 amino acid residues, including modified hydroxyproline residue, and its measured molecular mass is 2960 Da. The peptide was not toxic on mice; however, it stimulated their exploratory motivation and active search behavior, and demonstrated an anti-anxiety effect. AnmTX Sco 9a-1 at doses of 0.1 and 1 mg/kg reduced the volume of edema during 24 h better than the nonsteroidal anti-inflammatory drug, Diclofenac, at dose of 1 mg/kg i…

Inducing Complete Polyp Regeneration from the Aboral Physa of the Starlet Sea Anemone Nematostella vectensis.

Cnidarians, and specifically Hydra, were the first animals shown to regenerate damaged or severed structures, and indeed such studies arguab … mmon features and/or species or context-specific cellular and molecular mechanisms. The starlet sea anemone, Nematostella vectensis, is an emerging model organism for regeneration. Like Hydra, Nematostella is a member of the ancient phylum, cnidaria, but within the class anthozoa, a sister clade to the hydrozoa that is evolutionarily more basal. Thus aspe …

Acid-sensing ion channel (ASIC) structure and function: Insights from spider, snake and sea anemone venoms.

Acid-sensing ion channels (ASICs) are proton-activated cation channels that are expressed in a variety of neuronal and non-neuronal tissues. … ors described from animal venoms, with those from snakes and spiders favouring ASIC1, while the sea anemones preferentially target ASIC3. Some modulators, such as the prototypical ASIC1 modulator PcTx1 have been studied in great detail, while some of the newer members of the club remain largely unstudied. Here we review the current state of knowledge on v …

Calitoxin, a neurotoxic peptide from the sea anemone Calliactis parasitica: amino acid sequence and electrophysiological properties.

We have isolated a new toxin, calitoxin (CLX), from the sea anemone Calliactis parasitica whose amino acid sequence differs greatly from that of other sea anemone toxins. The polypeptide chain contains 46 amino acid residues, with a molecular mass of 4886 Da and an isoelectric point at pH 5.4. The amino acid sequence determined by Edman degradation of the reduced, S-carboxymethylated polypeptide chain and tryptic and chymotryptic peptides is Ile-Glu-Cys-Lys-Cys-Glu-Gly-Asp-Ala-Pro-Asp-Leu-Ser-His-Met-Thr-Gly-Thr- Val-Tyr - Phe-Ser-Cys-Lys-Gly-Gly-Asp-Gly-Ser-Trp-Ser-Lys-Cys-Asn-Thr-Tyr-Thr-Ala…

Is a Commensal Here, a Pathogen There? Genotype Specific Responses by the Sea Anemone Nematostella vectensis to Vibrio Species.

Animals rely on diverse molecular mechanisms to maintain beneficial microbial associations while also defending against pathogens, yet the extent to which these responses vary among genotypes of a single species remains poorly understood. Using individuals from different locations of the sea anemone Nematostella vectensis, we compared transcriptional responses to a commensal (Vibrio diabolicus) and pathogen (Vibrio coralliilyticus) bacterium. We find striking genotype-specific divergence: individuals from Nova Scotia mounted a strong transcriptional response to V. diabolicus, whereas the North…

The disruption of a symbiotic sea anemone by light pollution: Non-linear effects on zooxanthellae and molecular indicators.

Artificial Light at Night (ALAN) is a pervasive stressor that may affect coastal organisms, particularly sessile forms associated with photosynthetic symbionts. We examined the effects of ALAN upon the symbiotic snakelocks anemone (Anemonia viridis)'s relationship with photosynthetic zooxanthellae, as well as molecular indicators of oxidative stress and metabolism. Anemones were exposed to natural daylight/night or either mild or strong ALAN intensities for four weeks, before quantifying zooxanthellae, superoxide dismutase (SOD) enzymes, and respiration rates. In comparison to natural conditio…

Swimming sea anemones of Puget Sound: swimming of Actinostola new species in response to Stomphia coccinea.

Swimming as a response of the sea anemone Actinostola new species can be elicited as a result of contact with the submarginal surface of another swimming sea anemone Stomphia coccinea. However, Stomphia does not swim as a result of contact with Actinostola. In all other known respects, swimming is caused in both species by the same stimuli, including certain starfishes, a nudibranch, and electrical stimuli. No agent that causes Actinostola to swim has been detected in extracts, rubbings, or dried matter from Stomphia.

Diversity and Bioprospection of Functional Proteins from Sea Anemone Heteractis magnifica Based on Multi-Omics Approach.

Sea anemone venom has attracted increasing attention in biomedical research due to its multifarious compounds with biological activities. Although the venom is predominantly made up of proteins, the diversity and complexity of these proteins remain poorly understood. In this work, the proteins derived from the tentacle and column of Heteractis magnifica were investigated by integrating transcriptomic and proteomic technologies. A total of 3573 protein sequences from transcriptome databases were identified and clustered into nine functional categories. We also performed proteomic analysis on th…

Chronic, low concentration pesticide exposure alters reproduction and behavior in the intertidal sea anemone, Anthopleura elegantissima.

Widespread pesticide and herbicide use paired with frequent transport away from application sites has led to pesticide presence in nearly all terrestrial and aquatic environments globally. Pesticides have unintentional toxic effects on non-target organisms by interfering with cellular processes, behavior, feeding, reproduction, and disrupting endocrine processes. The aggregating anemone, Anthopleura elegantissima, is an important species along the North American Pacific coast due to its symbiotic relationships that contribute to high productivity, and its clonal abundance that structures the r…

The clownfish-hosting sea anemones (Anthozoa: Actiniaria): updated nomenclature, biogeography, and practical field guide.

Ten described species of sea anemones (Anthozoa: Hexacorallia: Actiniaria) serve as hosts to charismatic clownfishes (or anemonefishes) on coral reefs throughout the tropical Indo-West Pacific. Although not diverse in number, the clownfish-hosting sea anemones have large biogeographic ranges, exhibit extensive intraspecific phenotypic appearances, and have been surrounded by a great deal of historical and contemporary taxonomic and nomenclatural confusion. We believe these factors have created challenges for field scientists making real-time species-level identifications of host sea anemones. …

Neurotoxin localization to ectodermal gland cells uncovers an alternative mechanism of venom delivery in sea anemones.

Jellyfish, hydras, corals and sea anemones (phylum Cnidaria) are known for their venomous stinging cells, nematocytes, used for prey and defence. Here we show, however, that the potent Type I neurotoxin of the sea anemone Nematostella vectensis, Nv1, is confined to ectodermal gland cells rather than nematocytes. We demonstrate massive Nv1 secretion upon encounter with a crustacean prey. Concomitant discharge of nematocysts probably pierces the prey, expediting toxin penetration. Toxin efficiency in sea water is further demonstrated by the rapid paralysis of fish or crustacean larvae upon appli…

Equinatoxin II: How a Cationic Pore-Forming Sea Anemone Toxin Drives Nodal Swelling of Myelinated Nerve Fibers.

This study was performed to elucidate the mechanism underpinning the nodal swelling induced by equinatoxin II (EqtII), a cation-selective pore-forming toxin derived from the sea anemone Actinia equina. Experiments were conducted using frog myelinated nerve fibers as a model system. Application of EqtII led to an approximately two-fold increase in the nodal volume of myelinated axons, but only when extracellular Ca2+ was present. Replacing extracellular Cl- with isethionate had no measurable effect on this response, whereas substitution of NaCl with either sucrose or LiCl, an established Na+/Ca…

A variable residue in the pore of Kv1 channels is critical for the high affinity of blockers from sea anemones and scorpions.

Animal toxins are associated with well defined selectivity profiles; however the molecular basis for this property is not understood. To address this issue we refined our previous three-dimensional models of the complex between the sea anemone toxin BgK and the S5-S6 region of Kv1.1 (Gilquin, B., Racape, J., Wrisch, A., Visan, V., Lecoq, A., Grissmer, S., Ménez, A., and Gasparini, S. Modulation of Kv3 subfamily potassium currents by the sea anemone toxin BDS: significance for CNS and biophysical studies.

Kv3 potassium channels, with their ultra-rapid gating and high activation threshold, are essential for high-frequency firing in many CNS neurons. Significantly, the Kv3.4 subunit has been implicated in the major CNS disorders Parkinson's and Alzheimer's diseases, and it is claimed that selectively targeting this subunit will have therapeutic utility. Previous work suggested that BDS toxins ("blood depressing substance," from the sea anemone Anemonia sulcata) were specific blockers for rapidly inactivating Kv3.4 channels, and consequently these toxins are increasingly used as diagnostic agents …

Structural conservation of the pores of calcium-activated and voltage-gated potassium channels determined by a sea anemone toxin.

The structurally defined sea anemone peptide toxins ShK and BgK potently block the intermediate conductance, Ca(2+)-activated potassium channel IKCa1, a well recognized therapeutic target present in erythrocytes, human T-lymphocytes, and the colon. The well characterized voltage-gated Kv1.3 channel in human T-lymphocytes is also blocked by both peptides, although ShK has a approximately 1,000-fold greater affinity for Kv1.3 than IKCa1. To gain insight into the architecture of the toxin receptor in IKCa1, we used alanine-scanning in combination with mutant cycle analyses to map the ShK-IKCa1 in…

Isolation, Structural Elucidation, and Biological Evaluation of Pyrrole-Based Alkaloids from Sea Anemone-Associated Streptomyces sp. S1502.

Three new pyrrole alkaloids, streptopyrroles D-F (1-3), along with four known analogs (4-7) were isolated from Sea Anemone-Associated Streptomyces sp. S1502 via an OSMAC (One Strain Many Compounds)-based strategy. Their structures were elucidated through comprehensive spectroscopic analyses, including HRESIMS and 1D/2D NMR experiments (COSY, HSQC, and HMBC), and further confirmed by X-ray crystallography. Biological evaluation identified streptopyrrole (4) as an anti-MRSA (methicillin-resistant Staphylococcus aureus) agent, while 4 and 6 displayed broad-spectrum cytotoxicity and good selectivi…

Sea Anemone Kunitz Peptide HCIQ2c1: Structure, Modulation of TRPA1 Channel, and Suppression of Nociceptive Reaction In Vivo.

TRPA1 is a homotetrameric non-selective calcium-permeable channel. It contributes to chemical and temperature sensitivity, acute pain sensation, and development of inflammation. HCIQ2c1 is a peptide from the sea anemone Heteractis magnifica that inhibits serine proteases. Here, we showed that HCIQ2c1 significantly reduces AITC- and capsaicin-induced pain and inflammation in mice. Electrophysiology recordings in Xenopus oocytes expressing rat TRPA1 channel revealed that HCIQ2c1 binds to open TRPA1 and prevents its transition to closed and inhibitor-insensitive 'hyperactivated' states.
