Acquired motility of Babesia microti-infected red blood cells.

Babesia microti is an intraerythrocytic protozoan parasite and the main causative agent of human babesiosis in the United States. While extensive research has focused on the prevalence of this vector-borne pathogen in natural populations, increases of human cases and clinical manifestation, and pathogen structure, little is known about the movements of B. microti within vertebrate red blood cells (RBCs). RBCs are nonmotile due to their lack of cellular structures for active movement. Here, we report a phenomenon in which B. microti-infected RBCs exhibit an acquired motility compared to uninfec…

Inhibitory Effects of Fosmidomycin Against Babesia microti in vitro.

Babesia microti, the main pathogen causing human babesiosis, has been reported to exhibit resistance to the traditional treatment of azithromycin + atovaquone and clindamycin + quinine, suggesting the necessity of developing new drugs. The methylerythritol 4-phosphate (MEP) pathway, a unique pathway in apicomplexan parasites, was shown to play a crucial function in the growth of Plasmodium falciparum. In the MEP pathway, 1-deoxy-D-xylulose 5-phosphate reductoisomerase (DXR) is a rate-limiting enzyme and fosmidomycin (FSM) is a reported inhibitor for this enzyme. DXR has been shown as an antima…

Complete Molecular and Immunoprotective Characterization of Babesia microti Enolase.

The apicomplexan Babesia microti is the primary causative agent of human babesiosis, one of the most broadly distributed tick-borne diseases worldwide. B. microti undergoes a complex lifecycle within both the mammalian host and the tick vector, and employs several different specific molecular mechanisms to enter host cells. Enolase, the key glycolytic enzyme in intracellular glucose metabolism, can also be expressed on the parasite's outer surface, binds to human plasminogen, and coordinates apicomplexan parasite invasion of host cells, however, it lacks sorting sequences or lipoprotein anchor…

First Report of Babesia microti-Caused Babesiosis in Spain.

Babesiosis is an emerging zoonosis now found in several areas of the world. Using PCR and indirect immunofluorescence assay, we have diagnosed the first case of human babesiosis caused by Babesia microti in Spain. Diagnosis was delayed because of the nonspecific clinical symptoms that occurred in an immunocompetent patient.

Babesia microti in rodents and raccoons from northeast Florida.

Human babesiosis in the United States is caused most commonly by the intraerythrocytic protozoan parasite, Babesia microti . Although a few reports have described evidence of Babesia species in animals in Florida, to date Babesia microti specifically has not been reported from Florida or most other southern states. To determine if the organism is present in vertebrates in the region, small mammals were trapped and sampled at 2 sites in northeastern Florida, and DNA extracts from blood samples were screened for B. microti DNA via PCR assays targeting portions of the nuclear small subunit rRNA (…

Azotemia and mortality among Babesia microti-like infected dogs.

Babesia microti-like piroplasms are a recently recognized cause of illness in dogs in northwest Spain. Our objective was to describe the clinical characteristics and investigate the risk factors for azotemia and death among 58 B microti-like infected dogs. Twenty-one of the 58 (36%) dogs were azotemic at the time that the infection was diagnosed. The case fatality rate during the following week was 22%. Dogs with azotemia at the time of diagnosis were 10 times (95% CI, 3.26-28.8) more likely to die during the following week. Azotemia was the main cause of death for B microti-like infected dogs…

Immunogenicity of recombinant Babesia microti hsp70 homologue in mice.

A Babesia microti hsp70 homologue was cloned, expressed in a prokaryotic system and tested in a pilot study for protection against lethal challenge. Results showed that 30% of the mice inoculated with recombinant protein (rBm hsp70) survived challenge, while all of the controls died. Evidence of antibody response to the hsp homologue was detected by Western blot analysis, but no protection was imparted through passive transfer of convalescent serum. Passively transferred spleen cells, from mice which survived challenge, also failed to impart protection. The mechanism of partial immunity sugges…

Immunological relationships of Long Island isolates of Babesia microti.

Studies to detect strain differences among two rodent-derived and one human-derived Babesia microti isolates from Long Island were undertaken, using various methods. Superinfection experiments using the homologous and heterologous isolates showed cross-protection. All hamsters were resistant to superinfection challenges of increasing dosages of both the homologous and heterologous isolates. Attempts to infect other laboratory animals with the Long Island isolates of B. microti were successful in intact and splenectomized Sprague-Dawley rats and questionable in Swiss mice. Nylar and CFW mice as…

Babesia microti: morphology, distribution and host relationship in Germany.

A short review has been given about recent studies on Babesia microti in natural animal host's near Munich (Southern Germany). An infected area has been studied in order to elucidate the outdoor relationship between the local strains and their preferred hosts, the European field vole Microtus agrestis. The seasonal variation of the parasites prevalence in voles shows a rise in the early summertime (71% of the catches infected). Roundish forms predominate in the erythrocytes. Multiplication never takes place by binary fission, but in "Maltese cross" form or budding-like. Normally the parasite i…

A conserved protein of Babesia microti elicits partial protection against Babesia and Plasmodium infection.

BACKGROUND: The protozoan parasite Babesia microti that causes the zoonotic disease babesiosis resides in the erythrocytes of its mammalian host during its life-cycle. No effective vaccines are currently available to prevent Babesia microti infections. METHODS: We previously identified a highly seroactive antigen, named Bm8, as a B. microti conserved erythrocyte membrane-associated antigen, by high-throughput protein chip screening. Bioinformatic and phylogenetic analysis showed that this membrane-associated protein is conserved among apicomplexan hemoprotozoa, such as members of genera Babes…

Transmission of Babesia microti Parasites by Solid Organ Transplantation.

Babesia microti, an intraerythrocytic parasite, is tickborne in nature. In contrast to transmission by blood transfusion, which has been well documented, transmission associated with solid organ transplantation has not been reported. We describe parasitologically confirmed cases of babesiosis diagnosed ≈8 weeks posttransplantation in 2 recipients of renal allografts from an organ donor who was multiply transfused on the day he died from traumatic injuries. The organ donor and recipients had no identified risk factors for tickborne infection. Antibodies against B. microti parasites were not det…

Seroprevalence of Babesia microti infection in Canadian blood donors.

BACKGROUND: Human babesiosis, caused by the intraerythrocytic protozoan parasite Babesia microti, is primarily transmitted by tick bites and is also transmitted by transfusion. Infections have been identified in U.S. blood donors close to Canadian borders. We aimed to assess the risk of transfusion-transmitted babesiosis in Canada by examining infections in ticks and seroprevalence in blood donors. STUDY DESIGN AND METHODS: Passive surveillance (receipt of ticks submitted by the public) was used to identify regions for tick drag sampling (active surveillance, 2009-2014). All ticks were tested…

Reservoir competence of wildlife host species for Babesia microti.

Human babesiosis is an increasing health concern in the northeastern United States, where the causal agent, Babesia microti, is spread through the bite of infected Ixodes scapularis ticks. We sampled 10 mammal and 4 bird species within a vertebrate host community in southeastern New York to quantify reservoir competence (mean percentage of ticks infected by an individual host) using real-time PCR. We found reservoir competence levels >17% in white-footed mice (Peromyscus leucopus), raccoons (Procyon lotor), short-tailed shrews (Blarina brevicauda), and eastern chipmunks (Tamias striatus), and …

Acute fulminating babesiosis in hamsters infected with Babesia microti.

In this study, Babesia microti (ATCC30222) from mice was adapted to golden hamsters. The parasite was passaged to immunosuppressed and then adapted to normal hamsters. When 30 normal hamsters were inoculated with this strain, parasitaemia increased to 74% of erythrocytes by day 7 and 70% of the hamsters died. By day 12, parasitaemia extended to 90%, with 97% mortality. Hearts and kidneys from infected animals were enlarged. Histopathology revealed acute myocarditis, hepatitis, pneumonitis, glomerulonephritis and splenomegaly. Giemsa, Acridine Orange and Rhodamine staining of the parasite were …

Reservoir competence of white-footed mice for Babesia microti.

Although the white-footed mouse, Peromyscus leucopus Rafinesque, has been incriminated as the main reservoir of the agent of human babesiosis, Babesia microti Franca, a quantitative demonstration of reservoir competence has not been presented. Mice captured within an intensely zoonotic site served as host for laboratory-reared larval Ixodes dammini Spielman, Clifford, Piesman, and Corwin, and the resulting nymphal ticks were assayed for evidence of salivary gland infection by the piroplasm. Solely 25% of the mice were patently parasitemic on thin blood smears, but virtually all mice infected t…

Babesia microti, human babesiosis, and Borrelia burgdorferi in Connecticut.

Babesia microti was isolated from a white-footed mouse (Peromyscus leucopus) that was captured in southeastern Connecticut in 1988, when the first human case of babesiosis acquired in Connecticut was recognized. To date, 13 cases of babesiosis have been reported in Connecticut, the largest number of human cases reported on the mainland United States. Two of nine patients quiried remembered a prior tick bite. Since Babesia parasites are known to be vectored only by ticks, we surmise that 12 of these infections were acquired via tick bites; 1 was obtained by blood transfusion (the patient was 46…

Reduced dietary protein content suppresses infection with Babesia microti.

The influence of acute dietary protein restriction on the development of Babesia microti infection in the mouse model was investigated. Female mice consuming a diet either devoid of protein or adequate with respect to protein were infected with B. microti-parasitized erythrocytes and sacrificed 7 days later. Absence of dietary protein resulted in a delay in the onset of infection and a significantly reduced peak parasitemia. Non-specific antibody responses to heterologous erythrocytes and specific anti-babesial antibody titers were impaired in mice consuming the protein-free diets, suggesting …
