Bacteriophage Therapy for Chronic Mastoiditis.

OBJECTIVE: To provide the first description of intratympanic bacteriophage therapy for chronic mastoiditis from multidrug-resistant Pseudomo … of otorrhea, headaches, and hearing impairment. Subsequent cultures showed no growth. CONCLUSIONS: Bacteriophages can enhance antibiotic activity in cases of drug-resistant chronic mastoiditis.

[The sensitivity of Yersinia to bacteriophage Mu].

Various representatives of the genus Yersinia were found to differ in their sensitivity to the lytic action of bacteriophage Mu cts62, which could serve as an auxiliary test for the differentiation of Y. pestis and Y. pseudotuberculosis. Among the strains under study, the causative agents of plague (34 strains) were sensitive to phage Mu cts62, while the causative agents of enteric yersiniosis (42 strains) and pseudotuberculosis (73 strains), except 3 strains with the properties of Y. pestis, were resistant to this phage.

Specificity of bacteriophage Mu excision.

To study the excision of bacteriophage Mu at the DNA sequence level, the Mu-derived phage lambda placMu3 was transposed to the transcribed but non-translated leader region of a plasmid-borne tetracycline (tet) resistance gene. Revertants (excision products) were then selected by Tet+ restoration of Tet+ and characterized. Of 21 independent Tet+ revertants, 17 contained simple deletions of most or all of lambda placMu3, while the other four contained more complex rearrangements in which one end of lambda placMu3 had been transposed, and most of the prophage had been deleted. The deletion endpoi

Interbacterial competition triggers stress-induced CRISPR immunity against bacteriophages.

Bacteria-phage interactions are often studied in relatively simple experimental systems involving a single bacterial strain infected by a single phage.1 In natural environments, however, bacteria typically inhabit complex multispecies communities where species compete for limited resources and ecological niches.2,3,4 Recent studies show that the microbial community context can have profound effects on the evolution of phage resistance.5 For example, Pseudomonas aeruginosa evolves phage resistance primarily through receptor loss when grown in isolation but switches to CRISPR-based immunity in t

PrairieDogTown and Kinny: lytic environmental bacteriophages of Arthrobacter globiformis.

Two bacteriophages, PrairieDogTown and Kinny, were isolated from topsoil using Arthrobacter globiformis as a host. PrairieDogTown has a 36.2-kb genome encoding 52 genes, whereas Kinny has a 56.4-kb genome with 93 genes. Comparative genomic analysis places PrairieDogTown in Cluster FO and Kinny in Cluster AU in the Actinobacteriophage database.

Temperate bacteriophages shape intraspecies competition during Pseudomonas aeruginosa Infection.

Temperate bacteriophages are increasingly recognized as important drivers of microbial evolution. However, their role in mediating competition among members of the same species during infection remains poorly defined. Pseudomonas aeruginosa (Pa), an opportunistic pathogen associated with acute and chronic infections, frequently exhibits clonal dominance within infected individuals, suggesting the existence of effective mechanisms for excluding conspecific competitors. Here, we investigate the contribution of temperate bacteriophages to intraspecies competition among clinical Pa isolates. Scree…

Pf bacteriophages hinder sputum antibiotic diffusion via electrostatic binding.

Despite great progress in the field, chronic Pseudomonas aeruginosa (Pa) infections remain a major cause of morbidity and mortality in patients with cystic fibrosis, necessitating treatment with inhaled antibiotics. Pf phage is a filamentous bacteriophage produced by Pa that has been reported to act as a structural element in Pa biofilms. Pf presence has been associated with resistance to antibiotics and poor outcomes in cystic fibrosis, though the underlying mechanisms are unclear. Here, we have investigated how Pf phages and sputum biopolymers impede antibiotic diffusion using human sputum s

Bacteriophages and their potential for treatment of metabolic diseases.

Recent advances highlight the role of gut virome, particularly phageome, in metabolic disorders such as obesity, type 2 diabetes mellitus, metabolic dysfunction-associated fatty liver disease, and cardiovascular diseases, including hypertension, stroke, coronary heart disease, and hyperlipidemia. While alterations in gut bacteria are well-documented, emerging evidence suggests that changes in gut viruses also contribute to these disorders. Bacteriophages, the most abundant gut viruses, influence bacterial populations through their lytic and lysogenic cycles, potentially modulating the gut ecos…

Microenvironment-Activated Nanozyme-Armed Bacteriophages Efficiently Combat Bacterial Infection.

Bacterial infection is one of the greatest challenges to public health, requiring new therapeutic methods. Herein, an innovative nanozyme-armed phage (phage@palladium (Pd)) system is fabricated for combating bacterial infection. The proposed phage@Pd preserves the function of the phages to achieve precise recognition and adhesion to the host Escherichia coli. In aid of the phages, the ultrasmall Pd nanozymes equipped with conspicuous pH-dependent peroxidase-like activity can generate toxic hydroxyl radical around the bacteria in acidic and hydrogen-peroxide-overexpressed infection microenviron

Investigating the effect of bacteriophages on bacterial FtsZ localisation.

Escherichia coli is one of the most common Gram-negative pathogens and is responsible for infection leading to neonatal meningitis and sepsi … robials. In this study, we have used an in vitro meningitis model system for studying the effect of bacteriophages on FtsZ using fluorescent E. coli EV36/FtsZ-mCherry and K12/FtsZ-mNeon strains. We show localisation of FtsZ to the bacterial cell midbody as a single ring during normal growth conditions, and mislocalisation of FtsZ producing filamentous multi-ring …
