Acid-base regulation in the toad Bufo marinus during environmental hypoxia.

Gas exchange and the correlated changes in blood and tissue metabolic and acid-base status were investigated during long term exposure of the toad Bufo marinus to graded levels of hypoxia. During hypoxia, PCO2 values in blood and tissues fell, leading to a transient alkalosis in the extracellular but not in the intracellular space. A reduction in blood perfusion of the skin during hypoxia may explain why PCO2 was low in sartorius muscle under normoxia, but approached the PCO2 values found in other tissues (gastrocnemius muscle, ventricle) under hypoxia. At PO2 values below the critical PO2, la

Asymmetric effects of avermectin insecticides on testicular morphology and function in Bufo gargarizans.

Avermectins (AVMs) are widely used insecticides that pose potential risks to non-target organisms through environmental exposure. However, their effects on amphibian reproductive systems remain poorly understood. This study conducted a 14-day exposure experiment on the Asiatic toads (Bufo gargarizans) through food (0.36 ng/d), water (0.02 μg/L), and combined treatments to investigate the asymmetric toxic effects of AVM on testicular morphology and function in this species. Results showed that AVM exposure significantly reduced sperm motility and induced histopathological damage in testes, with

Photodynamic toxicity and its prevention by antioxidative agents in Bufo arenarum embryos.

In this work we describe an experimental model to evaluate the photodynamic toxicity on amphibian embryos, as well as the protective effect of antioxidants against the lethal oxidative stress induced by photosensitization. Bufo arenarum embryos were treated with 10 mg/l methylene blue (MB) in AMPHITOX solution for 72 h and then irradiated with a red laser or white light for variable times. Both light sources affected the survival of MB-treated animals and lethal effects occurred within the initial 12 h post-irradiation. For white light irradiation, the most effective phototoxic condition in ou

The basal midbrain as a region modulating the level of alerting in the toad, Bufo paracnemis.

Reversible thiopental blockade of sites located in the basal midbrain of conscious toads bearing chronically implanted cannulae induced a response in which the animal withdrew to the bottom of the experimental box with flexed head and forelegs. Partial or total eyelid closure, myosis, and reduced ventilatory movements also occurred. No significant changes in arterial pressure levels or heart rate occurred and the reflex wiping response to the application of a painful stimulus (acid) to the skin was also unaffected. However, the somatic and visceral responses to nonpainful stimuli were reduced.

Aerobic and anaerobic metabolism of the crystalline lens of a poikilotherm; the toad Bufo marinus.

1. The O2 consumption and lactic acid production of the lens of the toad Bufo marinus was measured under various conditions. The energy consumption was very low compared to other amphibian tissues and about 20% of that previously described in the rat lens. About 80% was derived from oxidative metabolism, which is the converse of that seen in mammals. 2. Cyanide abolished O2 consumption and increased lactate production 20-30 times ("Pasteur effect"). 3. Under aerobic conditions about 40% of the energy requirements of the lens are related to the presence of Na in the bathing-media but this was n

Methimazole disrupted skeletal ossification and muscle fiber transition in Bufo gargarizans larvae.

Methimazole (MMI) is an emerging endocrine disrupting chemical (EDC) due to its increasing use in the treatment of thyrotoxicosis (hyperthyroidism), but its potential impact on amphibian development remains largely unexplored. In the present study, the effects of 8 mg/L MMI and 1 μg/L thyroxine (T4) exposure on skeletal ossification and muscle development in Bufo gargarizans tadpoles were comprehensively investigated by double skeletal staining, histological analysis and RNA sequencing. Our results indicated that MMI treatment down-regulated the expression levels of ossification-related genes

Effects of temperature and oxygen availability on circulating catecholamines in the toad Bufo marinus.

The release of catecholamines during hypoxia has received limited attention in amphibians and the adrenergic regulation of cardio-pulmonary functions is, therefore, not well understood at the organismic level. To describe the changes in plasma catecholamine concentrations, we exposed toads (Bufo marinus) to different levels of hypoxia at two temperatures (15 and 25 degrees C). In addition, blood oxygen binding properties were determined in vitro at 15 and 25 degrees C at two different pH values. Hypoxia elicited a significant increase in plasma catecholamines (adrenaline and noradrenaline) at

Responses to circulatory pressures, and conduction velocity, of pulmocutaneous baroreceptors in Bufo marinus.

1. Baroreceptor activity was recorded within the recurrent laryngeal branch of the toad vagus in forty-four preparations. The receptive fields of the receptors were located in the pulmocutaneous artery (p.c.a.), generally within 5 mm of its separation from the truncus. However, the most easily recorded afferents in this nerve were mechanoreceptors which responded to punctate stimulation of the lip of the glottis. 2. The conduction velocities of p.c.a. baroreceptor and mechanoreceptive glottal afferents recorded in the recurrent laryngeal nerve ranged from 0.3-0.7 (0.5 +/- 0.1) m s-1 and 2.2-14

Membranes during yolk-platelet development in oocytes of the toad Bufo marinus.

Oocytes of the toad Bufo marinus have been studied by means of thin section and particularly freeze-fracture electron microscopy to characterize the cytoplasmic membranes around the yolk organelle, and the storage of yolk material in precursors and platelets. This appears to be a previously unknown type of yolk-platelet formation. During yolk-organelle development from the primordial precursor to the bi-partite fully grown yolk platelet, numerous lipoid droplets are attached to the periphery of the platelet, indicating an intense uptake of lipids. As is typical for amphibians, the fully grown

Comparative developmental toxicity of nickel to Gastrophryne carolinensis, Bufo terrestris, and Xenopus laevis.

The early embryo-larval developmental toxicity of nickel (Ni) to 3 amphibian species, Xenopus laevis (South African clawed frog), Bufo terrestris (southern toad), and Gastrophryne carolinensis (eastern narrow-mouthed toad), was evaluated using a modified FETAX model. Studies were initiated from late blastulae stage (Nieuwkoop and Faber [NF] stage 10 or Gosner stage 12) and completed at a common embryological-based test termination point, which represented the completion of the major stages of organogenesis (NF stage 46 for Xenopus or Gosner stage 26 for the toads). Results indicated that, in t

Vitamin A mediated limb deformities in the common Indian toad, Bufo melanostictus (Schneider).

Several types of limb deformities were induced by vitamin A in B. melanostictus. These ranged from total suppression of all the limbs (ectromelia) to partial development of either the forelimb or the hindlimb or both (mesomelia) to reduction or absence of digits in either the forelimbs or hindlimbs or both and absence of long bones in either the forelimbs or hindlimbs or both (phocomelia) or duplication of the hindlimbs (polymelia). All the limb abnormalities were induced in the developing limbs of the tail amputated tadpoles of B. melanostictus following vitamin A treatment, which is all the

Social isolation, rather than kinship, influences association behavior in toad tadpoles (Bufo formosus).

Experiences of social isolation alter the developing social behavior of animals. Association preference is an important behavioral tendency for amphibian larvae (tadpoles) that exhibit social aggregation as a means of predator avoidance. For tadpoles that exhibit social aggregation, the learning period during exposure to conspecifics could be crucial. This study investigated how water quality and social experience influence association preferences to unfamiliar conspecifics in Bufo formosus (Eastern-Japanese common toad) tadpoles. Using a 2 × 2 factorial design, subject tadpoles were reared ei

Comparative transcriptomes of three different skin sites for the Asiatic toad (Bufo gargarizans).

BACKGROUND: It has been proved that human voltage-dependent anion channel 2 (VDAC2) plays a significant role in sperm function and male fertility. This study was primarily aimed at exploring whether VDAC2 is a risk factor for idiopathic male infertility. RESULTS: We determined a significantly increased risk of idiopathic infertility with abnormal semen parameters in association with the variant rs2804535 and a decreased risk of idiopathic infertility with abnormal semen parameters in association with the variant rs11001334. However, among subjects with normal semen parameters, no significant

Hindlimb extensor muscle function during jumping and swimming in the toad (Bufo marinus).

Many anurans use their hindlimbs to generate propulsive forces during both jumping and swimming. To investigate the musculoskeletal dynamics and motor output underlying locomotion in such physically different environments, we examined patterns of muscle strain and activity using sonomicrometry and electromyography, respectively, during jumping and swimming in the toad Bufo marinus. We measured strain and electromyographic (EMG) activity in four hindlimb muscles: the semimembranosus, a hip extensor; the plantaris, an ankle extensor; and the gluteus and cruralis, two knee extensors. During jumpi

Effects of Separate and Combined Exposure of Cadmium and Lead on the Endochondral Ossification in Bufo gargarizans.

Cadmium (Cd) and lead (Pb) are ubiquitous in aquatic environments and most studies have examined the potential effects of Cd or Pb alone on aquatic organisms. In the present study, chronic effects of Cd and Pb, alone and in combination, on Bufo gargarizans were investigated by exposing embryos to these contaminants throughout metamorphosis. Significant reductions in body mass and snout-to-vent length were observed in B. gargarizans at Gosner stage 42 (Gs 42) and Gs 46 exposed to a Cd/Pb mixture. Single and combined exposure with Cd and Pb induced histological alterations of the thyroid gland c

Exposure to nitrate alters the histopathology and gene expression in the liver of Bufo gargarizans tadpoles.

Nitrate is a ubiquitous component in aquatic environment and the concentrations of anthropogenic nitrate-nitrogen (NO3N) can exceed 25 mg/L in surface waters and 100 mg/L in ground waters. The exceed nitrate has adverse effects on survival, development, and metamorphosis of amphibian. Liver is the hub of many biological processes, including lipid metabolism and bile salts secretion. However, there is little information about the effects of nitrate on the liver in amphibians during metamorphosis. In this study, B. gargarizans was exposed to different concentrations of nitrate from embryo to met
