Negative correlation between grey matter in the hippocampus and caudate nucleus in healthy aging.

Neurobiological changes that occur with aging include a reduction in function and volume of the hippocampus. These changes were associated with corresponding memory deficits in navigation tasks. However, navigation can involve different strategies that are dependent on the hippocampus and caudate nucleus. The proportion of people using hippocampus-dependent spatial strategies decreases across the lifespan. As such, the decrease in spatial strategies, and corresponding increase in caudate nucleus-dependent response strategies with age, may play a role in the observed neurobiological changes in …

Structural alterations in the caudate nucleus and precuneus in un-medicated anorexia nervosa patients.

Anorexia nervosa (AN) is a mental disorder characterized by a preoccupation with thinness and may be associated with brain structural alteration. The aim of the study was to examine the brain structural alteration in AN patients, including subcortical structure volume and cortical thickness. Thirty-five un-medicated AN patients and 20 matched healthy controls underwent structural magnetic resonance imaging brain scans. High resolution structural images were acquired on a SIEMENS 3T scanner and preprocessed using FreeSurfer software. Subcortical structure volume and cortical thickness were comp

Identification of an ATP-sensitive K+ channel in spiny neurons of rat caudate nucleus.

On the somata of GABAergic spiny neurons in the caudate nucleus of the rat an ATP-sensitive K+ channel (KATP-channel) was identified. The KATP-currents in cell-attached patches were activated both by energy-depleting conditions (200 microM cyanide) and by diazoxide (300 microM) and were reversibly blocked by tolbutamide (EC50 = 5 microM). In inside-out patch membranes both ATP (1 mM) and its non-hydrolyzable analog AMP-PNP (adenylylimidodiphosphate; EC50 = 27 microM) reversibly inhibited channel activity. These results demonstrate that the KATP-channel in spiny neurons displays properties char…

The neuronal mechanism underlying parkinsonism and dyskinesia, and differential roles of the putamen and caudate nucleus.

The basal ganglia were manipulated pharmacologically by using GABA antagonists and also 1-methyl-14-phenyl-1,2,3,6-tetrahydropyridine (MPTP) in rats, cats, and monkeys, and neuronal activities were recorded from the basal ganglia. It was concluded that (a) in parkinsonism, neuronal activities of the caudate nucleus decreased, those of the pars reticulata of the substantia nigra and internal segment of the globus pallidus increased, and those of the ventromedial nucleus of the thalamus depressed markedly; (b) in dyskinesia, periodical discharges of the caudate-putamen complex were the primary e…

Effect of xylocaine and carbachol applied to the caudate nucleus of rats on barbiturate sleeping time.

Since lesion of subcortical structures may affect the barbiturate sleeping time (bST), we decided with the present study to elucidate the role of the caudate nucleus in the determination of central sensitivity to barbiturates. Rats implanted with a cannula in the caudate nucleus of one or both sides, as well as with a jugular cannula, were utilized. Intravenous injections of sodium pentobarbital (40 mg/kg), intracerebral injections of Xylocaine (0.04 microgram), carbachol (0.2 micrograms) or artificial cerebrospinal fluid (ACF) into the caudate nucleus were performed. Both Xylocaine and carbac…

Caudate nucleus-dependent navigational strategies are associated with increased use of addictive drugs.

This study aimed to investigate the relationship between navigational strategies and the use of abused substances in a sample of healthy young adults. Navigational strategies were assessed with the 4-on-8 virtual maze (4/8VM), a task previously shown to dissociate between hippocampal-dependent spatial navigational strategies and caudate nucleus-dependent stimulus-response navigational strategies. Spatial strategies involve learning the spatial relationships between the landmarks in an environment, while response learning strategies involve learning a rigid set of stimulus-response type associa…

[Subcortical infarcts (caudate nucleus) in a case of bilateral anterior cerebral artery occlusion].

The authors describe a patient with bilateral anterior cerebral artery (ACA) occlusion. CT and MRI revealed bilateral encephalomalacia in the regions supplied by Heubner arteries and/or by perforating branches of ACA. The patient presented mainly with frontal symptomatology resulting from caudate nuclei lesion. Frontal symptomatology due to caudate impairment is discussed in the sense of frontal-subcortical circuits: lateral orbitofrontal and anterior cingulate ones. We emphasise a similarity of behavioural and cognitive disorders in early Huntington's disease and in frontal lobe lesion.

Caudate nucleus pathology in Parkinson's disease: ultrastructural and biochemical findings in biopsy material.

Ultrastructural and biochemical properties of caudate nucleus (CN) biopsies in two patients with advanced Parkinson's disease (PD) were compared with three CN specimens removed during surgery for intracranial tumors. An additional two specimens from neurologically intact patients (59 and 86 years old) were removed during autopsy (performed 3 and 4 h post mortem, respectively) for electron microscopic studies. Dopamine levels in PD were reduced to less than 15% of control values. Both PD patients showed frequent dystrophic neurites and transsynaptic degeneration of neurons and neuritic processe…

Direct projection from the visual associative cortex to the caudate nucleus in the feline brain.

Recent morphological and physiological studies support the assumption that the extrageniculate ascending tectofugal pathways send visual projection to the caudate nucleus (CN) in amniotes. In the present study we investigate the anatomical connection between the visual associative cortex along the anterior ectosylvian sulcus (AES) and the CN in adult domestic cats. An anterograde tracer - fluoro-dextrane-amine - was injected into the AES cortex. The distribution of labeled axons was not uniform in the CN. The majority of labeled axons and terminal like puncta was found only in a limited area i…

[Effect of chronic alcoholic intoxication in the rat on the structural organization of the caudate nucleus in their progeny].

By means of light (Nissl and Golgi), electron microscopy, as well as using morphometry, structure of neurons and interneuronal connections of the nucleus caudatus has been studied in 21-day-old rats reproduced by chronically alcoholized parents. As demonstrated the investigations, in young rats, physically underdeveloped, there are some signs of a delayed maturation in neurons, dendrites and synapses. Certain distrophic and reparative shifts are observed in all experimental animals. The distrophic changes of neural structures in the nucleus caudatus preponderate over the reparative ones, and i

Response of local blood flow in the caudate nucleus of the cat to intraventricular administration of carbachol.

The effect of perfusion of the cerebral ventricles with artificial cerebrospinal fluid containing carbachol on the blood flow in the caudate nucleus of the cat and the possibility to inhibit this effect by anticholinergic drugs was studied by means of the hydrogen clearance technique. After a control period during which both lateral ventricles were perfused with artificial CSF of identical composition, the drug under study was added on one side (experimental side) while the other side continued to be perfused with the control artificial CSF (control side). The blood flow on the experimental si…

Abnormal resting-state functional connectivity of the left caudate nucleus in obsessive-compulsive disorder.

Altered brain activities in the cortico-striato-thalamocortical (CSTC) circuitry are implicated in the pathophysiology of obsessive-compulsi … circuitry but increased functional connectivity in other brain regions. Furthermore, decreased left caudate nucleus-thalamus connectivity within the CSTC circuitry was positively correlated with the illness duration of OCD. This study provides additional evidence that CSTC circuitry may play an essential role and alteration of large-scale brain networks may be in …

Decreased diffusivity in the caudate nucleus of presymptomatic huntington disease gene carriers: which explanation?

BACKGROUND AND PURPOSE: The neostriatum is known to be affected in HD. In this work, our aim was to determine whether microstructural and vo … ic MR imaging and DWIs were acquired, and statistical analyses were performed on the volumes of the caudate nucleus and putamen and on the corresponding MD measurements. RESULTS: Neostriatal volumes were significantly smaller in both presymptomatic HD gene carriers and symptomatic patients with respect to controls. However, whereas the diffusivity in the caudate …

Reduction of caudate nucleus volumes in neuroleptic-naïve female subjects with schizotypal personality disorder.

BACKGROUND: The caudate nucleus might contribute to the psychopathological and cognitive deficits observed in schizotypal personality disorder (SPD), a schizophrenia spectrum disorder. Here we focused on female patients, because this group is underrepresented in studies of SPD and schizophrenia, and we might learn more about the caudate and clinical and cognitive impairments that are unique to female patients diagnosed with SPD. METHODS: Magnetic resonance imaging scans, obtained on a 1.5-T magnet with 1.5-mm contiguous slices, were used to measure the caudate in 32 neuroleptic-naïve women wi…

Spasmodic torticollis-like posture and caudate nucleus monoamine in cats: in vivo dialysis study.

In order to study the monoamine mechanism in the caudate nucleus of experimental spasmodic torticollis, the left side of the ventromedial tegmentum (VMT) was electrically destroyed in cats, and in vivo brain dialysis in the caudate nucleus was performed for determining the changes in monoamines and their metabolites. Six out of 12 animals showed the torticollis-like posture in which the head became deviated towards the side of the lesion by rotating around the longitudinal axis. In 4 of these 6 cats, this posture continued for 3 d until brain dialysis was performed. The extracellular 5-HIAA le…

Autoantibodies against neurofilaments in Creutzfeldt-Jakob disease: immunofluorescence in sections of human caudate nucleus.

Heterospecific autoantibodies to axonal neurofilaments of neurons in sera from five patients with Creutzfeldt-Jakob disease were detected with the use of cryostat section from normal human brain caudate nucleus. The antibody, visualized by the indirect immunofluorescence technique, was shown to bind complement. This allowed to achieve brighter staining either by the anticomplementary method alone or by a combination of both anti-C3 and anti-IgG conjugates

Relative sparing of dopaminergic terminals in the caudate nucleus is a feature of rest tremor in Parkinson's disease.

Resting tremor (RT) is a Parkinson's disease (PD) symptom with an unclear relationship to the dopaminergic system. We analysed data from 432 subjects from the Parkinson's Progression Markers Initiative, 57 additional PD patients and controls and 86 subjects referred for dopamine transporter single-photon emission computed tomography (DaT-SPECT). Caudate binding ratio (CBR), but not putamen binding ratio, was higher in RT patients. Furthermore, higher baseline CBR was linked to RT development. In the smaller cohorts, a 4-6 Hz oscillation-based metric from inertial sensors correlated with RT amp

Diffusion-Weighted MRI Findings of Caudate Nucleus and Putamen in Patients With Obsessive-Compulsive Disorder.

Objective The purpose of this study was to establish the diffusion-weighted magnetic resonance imaging (DW-MRI) findings of the caudate nucleus and putamen in patients with obsessive-compulsive disorder (OCD) and to obtain new information on the etiopathogenesis of OCD, which is still unclear. Methods The study comprised 20 patients with OCDs and 20 healthy volunteers. In these cases, DW-MRI and diffusion-weighted echo-planar images (DW-EPI) at b600 and b1000 gradient values were taken and the measurements were made using the apparent diffusion coefficient (ADC) maps of each group at b600 and …
