The incidence of extragenital and gynecologic diseases, surgical procedures and menstrual, gestational and family histories have been evaluated in 50 female carriers of balanced chromosomal rearrangements (index group) and 33 women without karyotype abnormalities (control group). This study provided criteria for karyotype testing in women. Counseling guidelines were recommended for female carriage of chromosomal anomalies. These recommendations depend on karyotypic presentations of the anomalies in a carrier: alternatives are prenatal diagnosis of the fetal karyotype and adequate contraception

Peripheral blood lymphocytes of patients undergoing radiation therapy for pelvic tumours have been examined for the presence of dicentric and centric ring chromosomes. Blood samples were taken, by venipuncture, prior to the first radiotherapy session and 24 h after radiotherapy sessions to allow the mixing of the irradiated lymphocytes in the circulating blood. The yield of dicentrics and centric rings was best fitted by a straight line which, according the maximum likelihood method, corresponds to Y = 1.77 +/- 0.0003 10(-2) D. On this basis the dose inducing ten dicentrics or rings is 5.62 Gy

The incidence of characteristic nonrandom chromosome aberrations in malignant cells of unselected, consecutively studied, and previously untreated patients from various geographic regions has been surveyed. The results demonstrate significant geographic heterogeneity in the distribution of the specific aberrations considered among the four malignant disorders studied in a sufficient number to permit conclusions, viz., acute nonlymphocytic leukemia, chronic myeloid leukemia, polycythemia vera, and myelodysplastic syndrome.

We describe two cases of sonographic abnormalities associated with unusual chromosomal aberrations. Case 1 presented with a cystic hygroma at 12 weeks' gestation. Cytogenetic analysis revealed an unbalanced complex chromosome rearrangement implicating chromosomes 6, 13 and 21 (karyotype: 47,XX,t(6;21;14)(q14;q21;q21)mat,+21) and corresponding to a complete trisomy 21. The frequencies of chromosome aberrations in 135 workers from nuclear- power plants were compared with those in 135 age-matched controls. A total of 135,000 cells was scored. A Monte Carlo simulation computer program for radiation-produced chromosome aberrations, based on the breakage-and-reunion model, was extended to include proximity effects due to localization of chromosomes and limited range for break-break interactions. Blood cell lymphocyte chromosomes from untreated (UT) and clinically-cured (CC) patients with paracoccidioidomycosis and from healthy (control) people (CO) were studied. The frequency of aneuploid cells in the UT patients was higher than in the CC and CO individuals. Cytogenetic analysis of short-term cultures of one non-secreting and one prolactin-producing pituitary adenoma revealed simple clonal numerical abnormalities in both tumors. The karyotype of the non-secreting adenoma was 48,XX, +4, +9[42]/49,XX, +4, +9, +20[2]/46,XX[6]. In the prolactin-secreting adenoma, three aberrant clones were detected, giving the karyotype 45,X, -Y[20]/47,XY, +Y[6]/45,XY, -21[3]/46,XY[21]. The cytogenetic findings of an alpha-fetoprotein (AFP)-producing hepatoblastoma in a 14-month-old girl are reported. Ten of 36 tumor cells were chromosomally abnormal, yielding the composite karyotype 47,XX,2q +, t(3;5)(p25;q31),dup(4)(q12q26), +20. Three of the chromosomal abnormalities (2q+, break at 4q12, +20) have been reported previously and might be of pathogenetic significance.

We studied 150 women with primary/secondary amenorrhea and/or oligomenorrhea; in 61 cases we found an abnormal karyotype (i.e. 40.7%): 51 cases in the first group of 110 patients with amenorrhea (i.e. 46%) and 10 cases in the second group of 40 patients with oligomenorrhea and/or secondary amenorrhea (i.e. 25%). The yield of chromosome aberrations (dicentric chromosomes, ring chromosomes) induced in lymphocytes of four radiotherapy patients was determined. In each case this yield was drastically increased when compared with the spontaneous incidence, the dicentric frequencies increasing by the following factors: 604, 525, 600, and 725. Data on the distribution of dicentrics and acentrics observed when human lymphocytes are cultured for 48 h after irradiation by X-rays, gamma-rays, and neutrons are presented. Analysis shows that for dicentrics, the observed distribution for X-rays, gamma-rays, and fission neutrons may be described by Poisson statistics but for higher energy neutrons overdispersion is observed.
