Discovery of Cinnamic Acid Derivatives as Potent Anti-H. pylori Agents.

Antibiotics are currently used for the treatment of Helicobacter pylori (H. pylori), which is confirmed to be the major cause of gastric disorders. However, the long-term consumption of antibiotics has already caused antibiotic resistance and side effects in vivo. Therefore, there is an emerging need for searching for safe and effective anti-H. pylori agents. Inspired by the excellent bioactivities of cinnamic acid, a series of cinnamic acid derivatives (compounds 1-30) were synthesized and determined for H. pylori inhibition. The initial screening revealed that compound 23, a 2,4-dinitro cinn

Structure-Activity Relationships of Cinnamate Ester Analogues as Potent Antiprotozoal Agents.

Protozoal infections are still a global health problem, threatening the lives of millions of people around the world, mainly in impoverished tropical and sub-tropical regions. Thus, in view of the lack of efficient therapies and increasing resistances against existing drugs, this study describes the antiprotozoal potential of synthetic cinnamate ester analogues and their structure-activity relationships. In general, Leishmania donovani and Trypanosoma brucei were quite susceptible to the compounds in a structure-dependent manner. Detailed analysis revealed a key role of the substitution patter

The Neuroprotective Effects of Cinnamic Aldehyde in an MPTP Mouse Model of Parkinson's Disease.

Cinnamic aldehyde (CA), a key flavor compound in cinnamon essential oil, has been identified as an anti-oxidant, anti-angiogenic, and anti-inflammatory material. Recently, the neuroprotective effects of CA have been reported in various neurodegenerative disorders, including Parkinson's disease (PD). In neurons, autophagy is tightly regulated, and consequently, the dysregulation of autophagy may induce neurodegenerative disorders. In the present study, we found that the selective dopaminergic neuronal death in the substantia nigra of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) mouse mod

Corrosion Protection of Mild Steel in Acidic Environments by Cetrimonium Cinnamates in Solution and Added to a Low Volatile Organic Compound Coating.

Protection of mild steel from acidic solutions used in the industry by environmentally friendly methods is an area of need. This work explores the anticorrosive properties of cetrimonium cinnamate compounds for mild steel in acid solutions as an additive in solution and as a pigment in a low-volatility organic compound (VOC) coating. Immersion tests show that protection is considerably enhanced after 24 h, at pH 1 i corr for the control being 330 μA/cm2 compared to 4.3 μA/cm2 for CTA-MeOcinn, suggesting synergy between the cetrimonium cation and cinnamate anion systems. Electrochemical Three-component Synthesis of Alkenesulfonates from Cinnamic Acids, SO2, and Alcohols.

A novel, electrochemical three-component reaction for the synthesis of alkyl alkenesulfonates from cinnamic acids, SO2, and alkyl alcohols is reported. This metal-free process employs inexpensive and readily available graphite electrodes in combination with easy-to-use stock solutions of SO2 and enables a straightforward construction of the styrene sulfonate scaffold via a decarboxylative transformation. Mechanistic studies indicate a pseudo-Kolbe type reaction. This novel reaction pathway enables a regioselective synthesis of alkenesulfonates from substituted cinnamic acids without double-bon

Therapeutic effects of cinnamic acid in a rat model of collagenase-induced Achilles tendinopathy.

BACKGROUND: Achilles tendinopathy is a common degenerative disorder characterized by pain, impaired function, and poor tissue regeneration. Effective treatment remains a challenge due to the tendon's limited healing capacity. Cinnamic acid, a naturally occurring phenolic compound, has demonstrated anti-inflammatory and antioxidant effects in various tissue repair models. However, its therapeutic potential in tendon injury remains unexplored, and this study represents the first study to evaluate its effects in an experimental Achilles tendinopathy model. METHODS: In this experimental study, te

Cinnamic Derivatives as Antitubercular Agents: Characterization by Quantitative Structure-Activity Relationship Studies.

Tuberculosis, caused by Mycobacterium tuberculosis (Mtb), remains one of the top ten causes of death worldwide and the main cause of mortality from a single infectious agent. The upsurge of multi- and extensively-drug resistant tuberculosis cases calls for an urgent need to develop new and more effective antitubercular drugs. As the cinnamoyl scaffold is a privileged and important pharmacophore in medicinal chemistry, some studies were conducted to find novel cinnamic acid derivatives (CAD) potentially active against tuberculosis. In this context, we have engaged in the setting up of a quantit

Preparation of goreisan suppository and pharmacokinetics of trans-cinnamic acid after administration to rabbits.

Goreisan suppository is prepared as a hospital preparation, and successfully used for the treatment of diarrhea and vomiting in young children with common cold. While clinical efficacy of the suppository has been reported, few studies have been carried out to clarify the preparation procedure and pharmacokinetics of the suppository. In this study, trans-cinnamic acid (CA) was used as a representative substance of goreisan constituents, and assayed by HPLC-UV. We investigated the properties of goreisan suppositories prepared using various sizes of pulverized goreisan extract granules, in vitro

Disclosure of cinnamic acid/4,9-diaminoacridine conjugates as multi-stage antiplasmodial hits.

4,9-diaminoacridines with reported antiplasmodial activity were coupled to different trans-cinnamic acids, delivering a new series of conjugates inspired by the covalent bitherapy concept. The new compounds were more potent than primaquine against hepatic stages of Plasmodium berghei, although this was accompanied by cytotoxic effects on Huh-7 hepatocytes. Relevantly, the conjugates displayed nanomolar activities against blood stage P. falciparum parasites, with no evidence of hemolytic effects below 100 µM. Moreover, the new compounds were at least 25-fold more potent than primaquine against

Can the epoxides of cinnamyl alcohol and cinnamal show new cases of contact allergy?

BACKGROUND: Cinnamyl alcohol is considered to be a prohapten and prehapten with cinnamal as the main metabolite. However, many individuals who are allergic to cinnamyl alcohol do not react to cinnamal. Sensitizing epoxides of cinnamyl alcohol and cinnamal have been identified as metabolites and autoxidation products of cinnamyl alcohol. OBJECTIVE: To investigate the clinical relevance of contact allergy to epoxycinnamyl alcohol and epoxycinnamal. METHODS: Irritative effects of the epoxides were investigated in 12 dermatitis patients. Epoxycinnamyl alcohol and epoxycinnamal were patch tested

Cinnamon spice and everything not nice: many features of intraoral allergy to cinnamic aldehyde.

Intraoral allergic contact dermatitis (ACD) is an uncommonly reported entity. The most commonly implicated allergens are metals that are incorporated into dental appliances. Intraoral ACD to nonmetal allergens is even less frequently described. Cinnamic aldehyde is widely used as a flavoring agent in foods and dentifrices. However, intraoral ACD to cinnamon flavoring agents has only been sporadically reported. In these cases, a variety of sources have been implicated, including candy, chewing gum, mouthwash, lip sunscreen, cinnamon toast, volatile oils, and toothpaste. The clinical presentatio

The suitability of hexyl cinnamic aldehyde as a calibrant for the murine local lymph node assay.

The murine local lymph node assay (LLNA) for the prospective identification of contact allergens assesses skin sensitization potential as a function of proliferative activity induced in lymph nodes draining the site of topical exposure to test chemical. This method has been endorsed recently as a stand alone test for the identification of contact allergens. We have now examined the suitability of hexyl cinnamic aldehyde (HCA), a recommended positive control for skin sensitization testing, as a calibrant for comparing the consistency of LLNA responses with time, and between laboratories, and th
