The circadian clock in the mouse habenula is set by catecholamines.

Circadian rhythms are those variations in behavioral and molecular processes of organisms that follow roughly 24 h cycles in the absence of any external cue. The hypothalamic suprachiasmatic nucleus (SCN) harbors the principal brain pacemaker driving circadian rhythms. The epithalamic habenula (Hb) contains a self-sustained circadian clock functionally coupled to the SCN. Anatomically, the Hb projects to the midbrain dopamine (DA) and serotonin (5-HT) systems, and it receives inputs from the forebrain, midbrain, and brainstem. The SCN is set by internal signals such as 5-HT or melatonin from t

Circadian clock mechanism driving mammalian photoperiodism.

The annual photoperiod cycle provides the critical environmental cue synchronizing rhythms of life in seasonal habitats. In 1936, Bünning proposed a circadian-based coincidence timer for photoperiodic synchronization in plants. Formal studies support the universality of this so-called coincidence timer, but we lack understanding of the mechanisms involved. Here we show in mammals that long photoperiods induce the circadian transcription factor BMAL2, in the pars tuberalis of the pituitary, and triggers summer biology through the eyes absent/thyrotrophin (EYA3/TSH) pathway. Conversely, long-dur

Circadian clock cryptochrome proteins regulate autoimmunity.

The circadian system regulates numerous physiological processes including immune responses. Here, we show that mice deficient of the circadian clock genes Cry1 and Cry2 [Cry double knockout (DKO)] develop an autoimmune phenotype including high serum IgG concentrations, serum antinuclear antibodies, and precipitation of IgG, IgM, and complement 3 in glomeruli and massive infiltration of leukocytes into the lungs and kidneys. Flow cytometry of lymphoid organs revealed decreased pre-B cell numbers and a higher percentage of mature recirculating B cells in the bone marrow, as well as increased num

Circadian Clock Regulates Bone Resorption in Mice.

The circadian clock controls many behavioral and physiological processes beyond daily rhythms. Circadian dysfunction increases the risk of cancer, obesity, and cardiovascular and metabolic diseases. Although clinical studies have shown that bone resorption is controlled by circadian rhythm, as indicated by diurnal variations in bone resorption, the molecular mechanism of circadian clock-dependent bone resorption remains unknown. To clarify the role of circadian rhythm in bone resorption, aryl hydrocarbon receptor nuclear translocator-like (Bmal1), a prototype circadian gene, was knocked out sp

Measuring circadian clock function in human cells.

Circadian clocks are present in most cells and are essential for maintenance of daily rhythms in physiology, mood, and cognition. Thus, not only neurons of the central circadian pacemaker but also many other peripheral tissues possess the same functional and self-sustained circadian clocks. Surprisingly, however, their properties vary widely within the human population. In recent years, this clock variance has been studied extensively both in health and in disease using robust lentivirus-based reporter technologies to probe circadian function in human peripheral cells as proxies for those in n…

Hearing damage and deafness: a role for the circadian clock.

Severe noise can cause permanent hearing damage. A recent study now shows that the capacity to recover from noise damage varies with time of day, driven by circadian clock control of a nerve growth factor (BDNF) in the inner ear.

Local Circadian Clocks Are Desynchronized From the Central Circadian Pacemaker in a Mouse Model of Inflammatory Arthritis.

OBJECTIVE: Rheumatoid arthritis (RA) is a chronic autoimmune disease affecting joints marked by daily and intermittent surges in inflammatory cytokine levels that result in "flares" of disease activity. Because circadian proteins regulate inflammatory responses in arthritis, continuous behavioral- and tissue-level monitoring may clarify therapeutic dynamics. Using the K/BxN serum transfer arthritis (STA) mouse model, we tested whether locomotor activity and circadian clock gene expression track RA progression from onset through symptom resolution. METHODS: C57BL/6NJ and PER2::luciferase mice

The Circadian Clock controls Hepatic Inflammation in MASLD.

BACKGROUND & AIMS: The circadian clock dictates time-of-day dependent gene, protein expression, and metabolite levels to ensure liver homeostasis. Circadian misalignment is implicated in the development of various metabolic disorders, but its contribution to the progression of Metabolic dysfunction-Associated Steatotic Liver Disease (MASLD) toward Metabolic dysfunction-Associated Steatohepatitis (MASH) remains poorly understood. Here, we aimed to dissect the role of the circadian clock in MASH development. METHODS: Using an environmental model of chronic circadian misalignment (n=6-12/group),

Autonomous Redox Rhythms and Their Coupling to the Circadian Clock.

Circadian rhythms have traditionally been considered to be outputs of genetic transcription-translation feedback loop (TTFL) oscillators. However, the discovery of circadian peroxiredoxin oxidation rhythms in anucleate red blood cells suggests that cells also possess an autonomous circadian redox rhythm. The conservation of this rhythm across all lineages of life, as well as in TTFL-defective backgrounds in multiple organisms, further indicates its importance. Rather than functioning as an isolated system, the redox rhythm is bidirectionally coupled to the genetic circadian clock. Perturbation

Circadian Clock: A Regulator of Immunity in Autoimmune Diseases.

AIM: Autoimmune diseases, characterized by the immune system mistakenly attacking the body's own tissues, are a growing global concern, with increasing prevalence. The circadian clock is a fundamental regulator of physiological processes, critically modulating immune functions. This review explores the intricate connections between circadian rhythms and immune responses in autoimmune pathogenesis and how disruptions exacerbate disease. METHODS: This synthesis examines recent research on circadian regulation of immune functions (leukocyte trafficking, cytokine secreion, phagocytosis) and autoi

Circadian clock in thyroid cancer: Effects and mechanisms.

The circadian clock, an endogenous timekeeping system governed by core genes such as BMAL1, CLOCK, PER, and CRY, regulates physiological processes critical to thyroid function. Disruption of circadian rhythms (resulting, for example, from shift work and/or exposure to artificial light at night) has emerged as a significant risk factor for thyroid carcinogenesis. This review elucidates the dual role of circadian rhythms in maintaining thyroid homeostasis and their mechanistic contributions to cancer initiation, progression, and therapy resistance. We highlight molecular pathways linking clock d

Circadian Clock Regulation of Developmental Time in the Kidney.

We report the emergence of an endogenous circadian clock that regulates organogenesis in mouse fetal kidney. We detect circadian rhythms both in vivo with transcriptional profiling and ex vivo by bioluminescence. High-resolution structural analysis of embryonic explants reveals that global or local clock disruption results in defects that resemble human congenital abnormalities of the kidney. The onset of fetal rhythms strongly correlates with the timing of a distinct transition in branching and growth rates during a gestational window of high fetal growth demands. Defects in clock mutants typ

Features of the circadian clock in the avian pineal gland.

The effects of different environmental factors on the circadian rhythm of melatonin release by chicken pineal glands were analyzed in 5-day experiments in vitro. Periodic, brief 4-h illumination at around midnight resulted in phase inversion in 2 days. Exposure to repeated 41 degrees C temperature elevation at around midnight caused no phase shift in the basic melatonin rhythm. Repeated inversion of the polarity of the magnetic field also affected rhythmic melatonin release. Additionally, expressions of Per1, Per2, Per3, Bmal1, Bmal2, Clock, Cry1, and Cry2 clock genes were simultaneously detec

Is self-reported morbidity related to the circadian clock?

Morningness and eveningness preference, an endogenous component of the circadian clock, is characterized by an interindividual difference in circadian phase and requires of humans a specific timing of behavior. The biological rhythms of morning and evening types are consequently phase shifted with fixed socioeconomic constraints. The impact of this phase shift on health is widely debated. The purpose of the authors' study was to determine the influence of morningness/eveningness preference on self-reported morbidity and health in an active population. A total of 1165 nonshift workers of the Fr

Molecular analysis of avian circadian clock genes.

Unlike mammals, avian circadian rhythms are regulated by a multiple oscillatory system consisting of the retina, the pineal and the suprachiasmatic nucleus in the hypothalamus. To understand avian circadian system, we have cloned Clock and Period homologs (qClock, qPer2 and qPer3) and characterized these genes in Japanese quail. Overall, qCLOCK, qPER2 and qPER3 showed approximately 79%, approximately 46% and approximately 33% amino acid identity to mCLOCK, mPER2, mPER3, respectively. Clock was mapped to quail chromosome 4 and chicken chromosome 4q1.6-q2.1. Per2 and Per3 genes were both localiz

Circadian Rhythms and Pain: A Narrative Review on Clock Genes and Circadian-Based Interventions.

Pain is a common symptom of many diseases and seriously affects the quality of life. Circadian rhythm is the regular, cyclical physiological, biochemical, and behavioral changes that occur within a 24-hour period in biological organisms, primarily regulated by clock genes. Pain sensitivity may have circadian rhythms, with clock genes likely influencing this pain-related rhythmicity. Therefore, restoring normal circadian rhythms and regulating the expression of clock genes are regarded as viable strategies to combat the development of pain. First, this review elucidates the core operational mec

Disruption of local circadian clocks in aristolochic acid-induced nephropathy in mice.

BACKGROUND: Aristolochic acid I (AAI), an emerging biogenic contaminant widely present in Aristolochic plants, has been implicated in the pr … rter mice and Per2-dLuc/Bmal1-dLuc reporter cell lines. To further elucidate the regulatory role of circadian clocks in AAI-induced nephropathy, mice with global or kidney-specific knockout of Bmal1, as well as mice subjected to experimental jetlag, were utilized. RESULTS: Our findings revealed a sex-dependent nephrotoxicity of AAI, with males exhibiting greater …
