Effect of semi-permeable membrane coverage on carbon dioxide emissions during composting: Insights into carbon metabolism pathways, dominated by the tricarboxylic acid cycle.

The tricarboxylic acid (TCA) cycle is the main carbon metabolism pathway leading to carbon loss in the form of carbon dioxide (CO2). Research on the CO2 emission reduction mechanism based on the TCA cycle under different composting strategies is relatively lacking. This study investigated the effects of semi-permeable membrane coverage on CO2 emissions and pathways of carbon metabolism during three batches of large-scale dairy manure aerobic composting by using gas chromatography and high-throughput sequencing technologies. The results showed that the CO2 emission rate peaked at the initial st

On the Role of Water as a Catalyst in Prebiotic Chemistry.

In this manuscript we provide computational support to the catalytic role of water in all kinds of pseudopericyclic reactions operating in the reductive acid cycle, as well as in other metabolic processes. Water catalysis is not limited to those reactions where simple translocation of hydrogen atoms occurs, such as those represented by NuH+E→Nu-EH general equation. Indeed, water catalysis is more general and extremely important in tautomerization reactions of the type HX-Y=Z→X=Y-ZH, which operate in the reductive citric acid cycle and metabolic processes. Moreover, the comprehensive theoretica…

Plasma metabolomics reveals distinct metabolic alterations and biomarkers of disease activity in HLA-B27-positive acute anterior uveitis.

HLA-B27 positivity is strongly associated with acute anterior uveitis (AAU). This study investigated HLA-B27-dependent metabolic alterations in AAU and identified potential biomarkers of disease activity using plasma metabolomics. Untargeted and targeted metabolomics were employed to compare plasma metabolic profiles of HLA-B27-positive and negative patients. Distinct metabolic signatures, characterised by significant alterations in pyruvic acid and L-kynurenine, were identified in HLA-B27-positive individuals compared to HLA-B27-negative individuals. Furthermore, citric acid levels in these H

Metabolic bottleneck at TCA cycle entry point and competition with trehalose production hinder growth of methylotrophic yeasts during methanol assimilation.

Methylotrophic yeasts are promising hosts for bioproduction from methanol, but their growth is significantly reduced in the presence of methanol. In this study, we investigated metabolic bottlenecks in Komagataella phaffii, Ogataea polymorpha, and Candida boidinii during methanol assimilation through comparative metabolome analysis. We found a common decrease in α-ketoglutarate-derived amino acids and tricarboxylic acid (TCA) cycle intermediates in all three species during methanol assimilation, suggesting a metabolic bottleneck around the TCA cycle entry. Additionally, K. phaffii and O. polym

Mitochondrial defects of brain and muscle.

The brain and muscle are particularly vulnerable to mitochondrial defects as are the heart, kidney and liver. This hierarchy of organ involvement is reflected in the clinical signs and symptoms associated with these diverse encephalomyopathic syndromes. The biochemical correlates involve pyruvate metabolism, the citric acid cycle and the respiratory chain. Lactic acidosis is the metabolic signature in these diseases although not present in all syndromes, witness Leber hereditary optic neuropathy. Ragged-red fibers are equally distinctive in biopsied skeletal muscle but are present only with bi…

Agenesis of corpus callosum and optic nerve hypoplasia due to mutations in SLC25A1 encoding the mitochondrial citrate transporter.

BACKGROUND: Agenesis of corpus callosum has been associated with several defects of the mitochondrial respiratory chain and the citric acid cycle. We now report the results of the biochemical and molecular studies of a patient with severe neurodevelopmental disease manifesting by agenesis of corpus callosum and optic nerve hypoplasia. METHODS AND RESULTS: A mitochondrial disease was suspected in this patient based on the prominent excretion of 2-hydroxyglutaric acid and Krebs cycle intermediates in urine and the finding of increased reactive oxygen species content and decreased mitochondrial …

Succinate as a signaling molecule in the mediation of liver diseases.

Succinate, one of the intermediates of the tricarboxylic acid (TCA) cycle, plays an essential role in the metabolism of mitochondria and the production of energy, and is considered as a signaling molecule in metabolism as well as in initiation and progression of hepatic diseases. Of note, succinate activates a downstream signaling pathway through GPR91, and elicits a variety of intracellular responses, such as succinylation, production of reactive oxygen species (ROS), stabilization of hypoxia-inducible factor-1α (HIF-1α), and significant impact in cellular metabolism because of the pivotal ro

Dissolved organic carbon diminishes manganese oxide-driven oxidation of chromium.

Oxidation of chromium (Cr)-bearing minerals by manganese (Mn) oxides is viewed as the dominant mechanism controlling geogenic production of Cr(VI) and its contamination of groundwater. This process may be modulated by other chemical constituents found in the natural environment, but such confounding factors have not been quantified. Here, we evaluated the mechanism of Cr(III) oxidation by mixed-valence Mn oxide in the presence of citric and gallic acids, two natural organic matter (NOM) constituents commonly found in the soil environment. Incubation experiments showed that each organic acid en

Urinary organic acid profiles of Reye's syndrome patients.

The status of mitochondrial functioning in Reye's syndrome was assessed by comparing organic acid profiles from nine pathological urines with those from normal urines. It was found that Reye's syndrome urines have a normal content of succinic, oxaloacetic, aconitic, and citric acids suggesting that the enzymes of the acid cycle are functional. Elevated pyruvate and depressed alpha-ketoglutarate levels were observed. Abnormal urinary constituents detected were salicylic and adipic acids. Presence of the latter indicates that the enzymes of fatty acid beta-oxidation are functional.

A Novel Anode Material Li2 FeGeO4 with Lithium Storage Mechanism Controlled by Both Iron and Germanium Elements.

To obtain anode materials with high capacity/energy density for lithium-ion batteries, a polyanionic compound Li2 FeGeO4 is prepared, which combines the conversion-type Fe-based oxide and the alloy-type Ge-based oxide at the atomic scale. The influence of citric acid in the sol-gel process on the structure and performance of the calcined products (LFG0, LFG1, and LFG2) is investigated. The results demonstrate that citric acid does not affect the phase of Li2 FeGeO4 . However, with the increase of citric acid, the crystallinity and grain size of the final product are reduced and its dispersion

Changes in Palatability Processing across the Estrous Cycle Are Modulated by Hypothalamic Estradiol Signaling.

Consumption varies across the stages (metestrus, diestrus, proestrus, estrus) of a rat's estrous cycle, changing in ways that might be expected to reflect, in part, a direct impact of hormones on taste palatability. Evidence regarding this hypothesis has been mixed, however, and critical within-subject experiments comparing consumption of multiple tastes with distinct valences across all estrous phases have been few. Here, we assayed female rats' licking of palatable (saccharin, sucrose, NaCl) and aversive (quinine-HCl, citric acid) tastes in brief-access trials, while tracking their estrous c

[Integrative transcriptomics-metabolomics approach to identify metabolic pathways regulated by glutamine synthetase activity].

Glutamine synthetase (GS), the only enzyme responsible for de novo glutamine synthesis, plays a significant role in cancer progression. As a … metabolism under glutamine-stripping conditions. The affected processes include DNA synthesis, the citric acid cycle, and reactive oxygen species (ROS) detoxification. This suppression originates from the inherent inability of cells to autonomously synthesize glutamine under glutamine-depleted conditions.
