Type II collagen gene variants and inherited osteonecrosis of the femoral head.

BACKGROUND: Avascular necrosis of the femoral head (ANFH) causes disability that often requires surgical intervention. Most cases of ANFH are sporadic, but we identified three families in which there was autosomal dominant inheritance of the disease and mapped the chromosomal position of the gene to 12q13. METHODS: We carried out haplotype analysis in the families, selected candidate genes from the critical interval for ANFH on 12q13, and sequenced the promoter and exonic regions of the type II collagen gene (COL2A1) from persons with inherited and sporadic forms of ANFH. RESULTS: We identif

Antibodies against the CB10 fragment of type II collagen in rheumatoid arthritis.

Antibodies against intact type II collagen (CII) are a feature of rheumatoid arthritis (RA) but have limited diagnostic value. Here we assess whether either of the two major cyanogen bromide fragments of CII, namely CB10 or CB11, are more sensitive substrates for the detection of antibodies in RA. Cleavage of bovine CII with cyanogen bromide yielded CB10 and CB11; these were purified by column chromatography for use in an enzyme-linked immunosorbent assay. Serum antibodies were measured in patients with RA, psoriatic arthritis (PsA), osteoarthritis (OA) and blood donors. Results were compared

Passive transfer by cells of type II collagen-induced arthritis in rats.

To investigate the role of immunologic hypersensitivity to collagen in the causation of type II collagen-induced arthritis in rats, passive transfer experiments were performed. Wistar/Lewis rats used in these experiments were demonstrated to be histocompatible by prolonged skin graft survival and mixed lymphocyte cultures. Popliteal lymph node weight assays excluded a potential for graft-vs.-host reactivity in this strain. 9 of 32 naive rats developed arthritis after intravenous receipt of pooled spleen and lymph node cells from donors that had been injected intradermally with type II collagen

[Points of undenatured type II collagen application in musculoskeletal pain syndromes treatment].

The dominant collagen of the cartilaginous matrix in adults is type II collagen. The amount of type II collagen in the intercellular matrix of cartilage is significantly reduced against the background of musculoskeletal system diseases. The basis of articular cartilage is hyaline cartilage tissue consisting of chondrocytes with tissue-specific antigens that induce the production of antibodies in patients with osteoarthritis (OA). Today, new approaches are being considered in the treatment of OA with the use of udenatured type II collagen (UC-II). Such molecular mechanisms of action of UC-II as

Association of HLA-DR and type II collagen autoimmunity with Meniere's disease.

To investigate HLA-associated genetic susceptibility to Meniere's disease in relation to type II collagen (CII) autoimmunity status, HLA-DRB1 genotyping and ELISA measurement of anti-CII antibody were performed in 41 Korean patients with Meniere's disease. In the anti-CII positive subgroup (20%) of patients, the frequency of HLA-DRB1*0405 was significantly increased (uncorrected) compared with both controls (63% vs 16%) and anti-CII negative patients (63% vs 12%). In the anti-CII negative subgroup, HLA-DRB1*1201 was significantly increased (uncorrected) (27% vs 10%) and DRB1*13 was decreased (

Urinary type II collagen neoepitope as an outcome measure for relapsing polychondritis.

Herein we describe the case of a man who was diagnosed as having relapsing polychondritis (RP) when he was 18 years of age and was treated over the course of 2 years with numerous immunosuppressive agents, including tumor necrosis factor alpha (TNFalpha) inhibitors. His respiratory symptoms were refractory to treatment. Serum and urine samples were obtained periodically for measurement of erythrocyte sedimentation rate (ESR), C-reactive protein (CRP) levels, anti-type II collagen (anti-CII) antibodies, and urinary type II collagen neoepitope (uTIINE) levels. The uTIINE assay is specific for co

[Human type II collagen induced autoimmune inner ear lesions in guinea pigs].

Guinea pigs were immunized with human type I, type II or type III collagen. Inner ear lesions were induced in some guinea pigs by immunizing them with human type II collagen. Histopathologic changes included endolymphatic hydrops, organ of Corti lesions and endolymphatic duct contained a large number of inflammatory cells. These histologic changes resembled those observed in patient with Meniere's disease. We didn't discover any inner ear lesions in guinea pigs by immunizing them with human type I and type III collagen, although their serum antibody titers were high and antibody presented in p

Characterisation of the major CNBr-Derived peptides of porcine type II collagen.

As part of a general study on the matrix macromolecules of pig cartilage, the CNBr-derived peptides of porcine type II collagen have been isolated from laryngeal cartilage and characterized. Type II was the only molecular type of collagen detected in laryngeal cartilage from 6-9 month old pigs. The major CNBr-peptides of this collagen were prepared by ion exchange and molecular sieve chromatography and characterized by SDS-polyacrylamide disc electrophoresis and amino acid analysis. Six peptides were recovered in high yield and were shown to be closely homologous to similar peptides previously

On the mechanics of networked type II collagen: Experiments, constitutive modeling, and validation.

In this study we investigate the mechanics of type II collagen fibrils, an essential structural component in many load-bearing tissues including cartilage. Although type II collagen plays a crucial role in maintaining tissue integrity, the stress-stretch and failure response of type II collagen fibrils in tension is not established in the current mechanics literature. To address this knowledge gap, we conducted tensile tests on isolated collagen networks from articular cartilage and established a validated constitutive model for type II collagen fibril. We identified two distinct failure mecha

Erythropoietin reduces the degree of arthritis caused by type II collagen in the mouse.

OBJECTIVE: Erythropoietin (EPO) is a potent stimulator of erythroid progenitor cells, and its expression is enhanced by hypoxia. The aim of this study was to investigate the effect of EPO on collagen-induced arthritis (CIA) in the mouse. METHODS: CIA was induced by intradermal injection of bovine type II collagen (CII) and Freund's complete adjuvant. Starting on day 25, some of the mice with CIA received daily subcutaneous injections of EPO (1,000 units/kg). Two other groups of mice received sham treatment alone or sham treatment followed by EPO treatment, respectively. Arthritis was assessed

Expression of collagen type I, II and III in loose body of osteoarthritis.

The expression of collagen type I, II, and III was investigated to evaluate phenotypic change in chondrocytes in loose bodies related to osteoarthritis. We assessed collagen type I, II, and III production in loose bodies from knee joints of ten osteoarthritic patients, using an immunohistochemical method with monoclonal antibodies. Collagen type III expression was identified in all ten loose bodies and was mainly located in cartilage, including chondrocytes and matrices, as well as in a layer of fibroid tissue on the surface. No positive signal for collagen type III was observed in necrotic os

Gene expression of type II collagens in chondro-osteophytes in experimental osteoarthritis.

The formation of chondro-osteophytes in osteoarthritic joints is a unique example of adult neochondrogenesis that bears some similarities to growth plate elongation and fracture callus formation. This study uses in situ hybridization histochemistry to define the molecular phenotype of cells in active chondro-osteophytes. Chondro-osteophytes are composed of fibrocytes and osteoblasts that express type I procollagen mRNA, mesenchymal prechondrochytes that express type IIA procollagen mRNA, and maturing chondrocytes that express type IIB procollagen mRNA. Based on the spatial pattern of gene expr

Independent deposition of collagen types II and IX at epithelial-mesenchymal interfaces.

Previous studies have demonstrated the presence of type II collagen (in mature chickens predominantly a 'cartilage-specific' collagen) in a variety of embryonic extracellular matrices that separate epithelia from mesenchyme. In an immunohistochemical study using collagen type-specific monoclonal antibodies, we asked whether type IX collagen, another 'cartilage-specific' collagen, is coexpressed along with type II at such interfaces. We confirmed that, in the matrix underlying a variety of cranial ectodermal derivatives and along the ventrolateral surfaces of neuroepithelia, type II collagen is

Nonexpression of cartilage type II collagen in a case of Langer-Saldino achondrogenesis.

A lethal short-limbed dwarfism was diagnosed at autopsy as the Langer-Saldino variant of achondrogenesis by radiological, histological, and gross pathological criteria. Cartilage was obtained for biochemical and ultrastructural analyses from the ends of long bones, from ribs and from a scapula of the newborn infant. At all sites, it had an abnormal gelatinous texture and translucent appearance. Biochemical analyses of the cartilages to identify pepsin-solubilized collagen alpha-chains and collagen-specific CNBr-peptides failed to detect type II collagen at any site where it would normally be t

Antibodies to native and denatured type II collagen in children with rheumatic diseases.

Sera of 153 children with rheumatic diseases were assayed by an enzyme-linked immunosorbent assay (ELISA) for the presence of antibodies to native (n) and to denatured (d) type II collagen (IIC). Among the study population, antibodies to nIIC were detected in 47% with juvenile rheumatoid arthritis (JRA), 21% with spondyloarthropathies, 37% with other rheumatic diseases, 70% with adult onset rheumatoid arthritis (RA), and in less than 1% of nonrheumatic disease control groups. Antibodies to dIIC were detected in 42% of children with JRA, 41% with spondyloarthropathies, 37% of children with othe

The Ultrastructural Localization of Type II, IV, and VI Collagens at the Vitreoretinal Interface.

BACKGROUND: The vitreoretinal interface is the border of the cortical vitreous and the inner surface of the retina. The adhesion of the cortical vitreous to the ILM, namely vitreoretinal adhesion, involves a series of complex molecular adhesion mechanisms and has been considered as an important pathogenic factor in many vitreoretinal diseases. The presence of type VI collagen at the vitreoretinal interface and its possible interaction with collagens and glycoproteins indicates that type VI collagen may contribute to the vitreoretinal adhesion. PURPOSE: To clarify the ultrastructural location

Demonstration of antibodies to denatured type I and type II collagen in juvenile rheumatoid arthritis, Still's syndrome and controls by [14C] collagen radioimmunoassay.

From 88 sera of patients with juvenile rheumatoid arthritis 30.6% showed antibodies to denatured type I collagen an 31.8% antibodies to type II collagen, a percentage which corresponds to radioimmunoassay results in adult RA. Rheumatoid factors were demonstrated with Waaler-Rose test in 14.7% and with latex test in 6.8% of investigated patients. Results of investigations with collagen types I and II correlated in 73.7%. Whilst type I collagen antibodies were found with equal frequency in active and non-active stages, type II antibodies were twice as frequent in active stages as in non-active s
