Functional Roles of Two β-Tubulin Isotypes in Regulation of Sensitivity of Colletotrichum fructicola to Carbendazim.

Colletotrichum fructicola is the major pathogen of anthracnose in tea-oil trees in China. Control of anthracnose in tea-oil trees mainly depends on the application of chemical fungicides such as carbendazim. However, the current sensitivity of C. fructicola isolates in tea-oil trees to carbendazim has not been reported. Here, we tested the sensitivity of 121 C. fructicola isolates collected from Guangdong, Guangxi, Guizhou, Hainan, Hunan, Jiangsu, and Jiangxi provinces in China to carbendazim. One hundred and ten isolates were sensitive to carbendazim, and 11 isolates were highly resistant to …

Biocontrol of Colletotrichum fructicola in the Postharvest Banana Fruit Using the Siderophore-Producing Strain BX1.

Banana anthracnose, caused by Colletotrichum fructicola, significantly reduced the postharvest fruit quality. Employing biocontrol strategies offers a sustainable approach to enhance agricultural practices. The Burkholderia sp. strain BX1 hinders the growth and appressorium formation of C. fructicola, and its sterile filtrate lowers the anthracnose incidence while preserving the fruit quality. Scanning electron microscopy and genomic analyses confirmed BX1 as Burkholderia pyrrocinia. AntiSMASH analysis identified three siderophores with high similarity, and improved MALDI-TOF IMS confirmed the…

The Laccase Family Gene CsLAC37 Participates in Resistance to Colletotrichum gloeosporioides Infection in Tea Plants.

Fungal attacks have become a major obstacle in tea plantations. Colletotrichum gloeosporioides is one of the most devastating fungal pathogens in tea plantations that can severely affect tea yield and quality. However, the molecular mechanism of resistance genes involved in anthracnose is still largely unknown in tea plants. Here, we found that the laccase gene CsLAC37 was involved in the response to fungal infection based on a transcriptome analysis. The full-length CDS of CsLAC37 was cloned, and its protein sequence had the closest relationship with the Arabidopsis AtLAC15 protein compared t…

Molecular, morphophysiological, and pathogenic characterization of Colletotrichum isolates from strawberry plants in Santa Catarina, Brazil.

Strawberry (Fragaria × ananassa) holds significant importance within the small fruits group of temperate climates. However, its high susceptibility to diseases, particularly anthracnose caused by Colletotrichum isolates, can severely limit production. This study aimed to identify and characterize Colletotrichum species causing strawberry anthracnose in Santa Catarina state, Brazil. Isolates were collected from strawberry plants exhibiting typical anthracnose symptoms on different plant parts, in both organic and conventional fields. A phylogenetic analysis was conducted using six loci, includi…

Quorum sensing-mediated inter-specific conidial anastomosis tube fusion between Colletotrichum gloeosporioides and C. siamense.

Many plant pathogenic filamentous fungi undergo fusion of conidia through conidial anastomosis tubes (CATs), which is believed to facilitate horizontal gene transfer between species. We discovered a remarkable inter-specific CAT fusion between two important plant fungal pathogens Colletotrichum gloeosporioides and C. siamense. In an invitro assay, under no selection pressure, the inter-specific CAT fusion was preferred with higher frequency (25% ± 5%) than intra-specific CAT fusion (11% ± 3.6%). Different stages of CAT fusion viz. CAT induction, homing, and fusion were observed during this int…

Transcriptional reprogramming of major defense-signaling pathways during defense priming and sugarcane-Colletotrichum falcatum interaction.

Red rot caused by Colletotrichum falcatum poses a serious threat to sugarcane cultivation in many tropical and sub-tropical countries. Deciphering the molecular network of major defense-signaling pathways in sugarcane cultivars with varying red rot resistance is essential to elucidate the phenomenon of defense priming exerted by resistance inducers. Therefore, in this study, expression pattern of transcripts coding for major defense-signaling pathway regulatory genes was profiled during compatible and incompatible interactions and in response to defense priming using qRT-PCR. Candidate genes t…

Diversity, Virulence, and Fungicide Sensitivity of Colletotrichum Species Associated with Apple Bitter Rot in Nova Scotia.

To date, 21 distinct Colletotrichum species have been identified as causal agents of bitter rot on apple fruit. However, whereas bitter rot occurs in nearly all apple-growing regions, the Colletotrichum species responsible for this disease are not globally prevalent and have distinct regional distributions. Because Colletotrichum species can differ in pathogenicity, virulence, and fungicide sensitivities, characterizing the species present in a growing region is essential for effective disease management. In this study, we obtained 130 Colletotrichum isolates from symptomatic fruit collected i…

Insights into tandem dioxygenases-copper radical oxidases in fungal Phytopathogens of the Colletotrichum acutatum species complex.

Fungal phytopathogens represent a major threat to global agriculture, causing extensive yield losses. Among them, Colletotrichum spp. are infamous for their capacity of infecting a wide variety of monocot and dicot hosts. Motivated by the recent discovery in Colletotrichum orbiculare of a redox relay between Copper Radical Alcohol Oxidases from the AA5_2 subfamily and tandem peroxidases involved in plant infection, we investigated species from the Colletotrichum species complex (C. acutatum and C. tamarilloi) where the tandem peroxidase gene is absent. Instead, putative ring-cleavage dioxygena…

HSP17.4 mediates salicylic acid and jasmonic acid pathways in the regulation of resistance to Colletotrichum gloeosporioides in strawberry.

In this study, we used virus-mediated gene silencing technology and found that the HSP17.4 gene-silenced cultivar Sweet Charlie plants were more susceptible to Colletotrichum gloeosporioides than the wild-type Sweet Charlie, and the level of infection was even higher than that of the susceptible cultivar Benihopp. The results of differential quantitative proteomics showed that after infection with the pathogen, the expression of the downstream response genes NPR1, TGA, and PR-1 of the salicylic acid (SA) signalling pathway was fully up-regulated in the wild-type Sweet Charlie, and the expressi…

Comparative transcriptomic analysis indicates genes associated with local and systemic resistance to Colletotrichum graminicola in maize.

The hemibiotrophic fungus Colletotrichum graminicola may cause severe damage to maize, affecting normal development of the plant and decreasing grain yield. In this context, understanding plant defense pathways at the inoculation site and systemically in uninoculated tissues can help in the development of genetic engineering of resistance against this pathogen. Previous work has discussed the molecular basis of maize - C. graminicola interaction. However, many genes involved in defense have not yet been exploited for lack of annotation in public databases. Here, changes in global gene expressi…

First Report of Colletotrichum fioriniae Causing Postharvest Decay on 'Nittany' Apple Fruit in the United States.

Bitter rot of apple is caused by Colletotrichum acutatum and C. gleosporioides and is an economically important disease in the mid-Atlantic and southern regions of the United States (1). However, other Colletotrichum spp. have been found to infect apple and pear fruit in Croatia that include C. fioriniae and C. clavatum (3). The disease is favorable under wet, humid conditions and can occur in the field or during storage causing postharvest decay (2). In February 2013, 'Nittany' apples with round, brown, dry, firm lesions having acervuli in concentric rings were observed at a commercial cold s…

Multi-Omics Profiling Unveils Antifungal Secondary Metabolites Mediating Epicoccum layuense LQ's Biocontrol Efficacy Against Colletotrichum fructicola.

Colletotrichum fructicola is a significant phytopathogen in both pre- and postharvest stages of fruit development and storage. The development of environmentally friendly biological control agents has attracted increasing research interest. In this study, we characterized a fungal strain (Epicoccum layuense LQ) that strongly inhibits C. fructicola. A potato dextrose broth culture filtrate of strain LQ inhibited the vegetative growth of C. fructicola by approximately 80% at a 1:10 (vol/vol) dilution. Cytological observations revealed that the filtrate disrupted mitosis and cellular polarity dur…

A Fungal Glycoside Hydrolase From Colletotrichum asianum Mini-Chromosomes Targets Host Chitinase to Suppress Plant Immunity.

Colletotrichum asianum, the cause of mango anthracnose, is a major threat to mango production worldwide. However, the mechanisms underlying plant invasion by C. asianum are poorly understood. Mini-chromosomes are increasingly recognised as indispensable components in the virulence of plant-pathogenic fungi. Here, we identify CaMutA, a secreted glycoside hydrolase 71 effector encoded on a mini-chromosome, as a critical pathogenicity factor that suppresses plant immune responses and is essential for infection by C. asianum. CaMutA directly interacts with the mango chitinase MiChi1. When heterolo…

Natamycin, a New Biofungicide for Managing Crown Rot of Strawberry Caused by QoI-Resistant Colletotrichum acutatum.

Anthracnose crown rot of strawberry, caused by Colletotrichum acutatum, is an important disease affecting California nursery and fruit production. Preplant dip treatments of transplants with fludioxonil-cyprodinil or azoxystrobin are industry standards for managing the disease and have been used extensively. Following reports of reduced efficacy of azoxystrobin in the field, high levels of quinone outside inhibitor (QoI) resistance were detected in California isolates of the pathogen. Resistance was associated with the G143A mutation in the cytochrome b gene, similar to a previous report from …
