Antiulcerogenic effect and cytotoxic activity of semi-synthetic crotonin obtained from Croton cajucara Benth.

Trans-dehydrocrotonin, the major diterpene isolated from the bark of Croton cajucara, has good antiulcerogenic activity which, however, is accompanied by toxic effects. On the basis of these results, a semi-synthetic crotonin, named 4SRC, was prepared to determine whether this substance has similar antiulcerogenic activity with lower or no toxicity. The natural crotonin was also isolated from the bark of C. cajucara but was not used due to the small amount obtained. The cytotoxic effect of semi-synthetic crotonin, expressed as cell viability, was assessed in (a) lung fibroblast cell line (V79)…

Discovery of Three Macrocyclic Diterpenoid Skeletons from Croton laevigatus and Their NF-κB Inhibitory Activity.

Phytochemical investigation of Croton laevigatus yielded six macrocyclic diterpenoids, craeviganoids A-F (1-6), featuring three novel carbon skeletons: two distinct types of 13-membered diterpenoid lactones (1-3) bearing a rare bridgehead alkene, and a 14-membered furanoditerpenoid skeleton (4-6). Their structures were elucidated by spectroscopic analysis, X-ray crystallography, and quantum chemical calculations, revealing distinct solid-state and solution conformations for 1. Moreover, compound 4 potently inhibited NF-κB and exhibited anti-inflammatory activity.

Cardiovascular Effects of the Essential Oil of Croton argyrophylloides in Normotensive Rats: Role of the Autonomic Nervous System.

Cardiovascular effects of the essential oil of Croton argyrophylloides Muell. Arg. (EOCA) were investigated in normotensive rats. In saline-pretreated anesthetized or conscious rats, intravenous (i.v.) injection of the EOCA induced dose-dependent hypotension. Dose-dependent tachycardia was observed only in conscious rats. In anesthetized rats, cervical bivagotomy failed to enhance EOCA-induced hypotension but unmasked significant bradycardia. In conscious rats, i.v. pretreatment with methylatropine, but not with atenolol or L-NAME, reduced both hypotensive and tachycardiac responses to EOCA. H…
