Kinship practices underpin all traditional societies, forming the basis for socially sanctioned reproductive unions, residence patterns and the inheritance of rights and property 1,2 . Although the relationship between biological relatedness and kinship is not always straightforward, ancient DNA studies are increasingly used to examine the extent to which biological relatedness underpinned social constructs of kinship in prehistoric societies 3-6 . Here, we report the analysis of genome-wide data for 534 individuals from the Arras Culture of Middle Iron Age northeast England (including 390 fro

Millions of scientific specimens are housed in museum collections, a large part of which are fluid preserved. The use of formaldehyde as fixative and subsequent storage in ethanol is especially common in ichthyology and herpetology. This type of preservation damages DNA and reduces the chance of successful retrieval of genetic data. We applied ancient DNA extraction and single stranded library construction protocols to a variety of vertebrate samples obtained from wet collections and of different ages. Our results show that almost all samples tested yielded endogenous DNA. Archival DNA extract

BACKGROUND: The development of complex urban societies in Mesopotamia fundamentally shaped human history, yet the genetic dynamics underlying this process remain poorly understood. Here, we sequence DNA from 17 individuals spanning the Bronze and Iron Ages at Bakr Awa, one of northeastern Iraq's largest ancient settlements located at the border between Mesopotamia and Iran.
RESULTS: Genome-wide analyses reveal substantial genetic heterogeneity during the Bronze Age, characterized by influences of Anatolian, Levantine, and Caucasus/Yamnaya-related ancestries on the local background - complemen

Specific lineages of serogroup O1 Vibrio cholerae are notorious for causing cholera pandemics, of which there have been seven since the 1800s. Much is known about the sixth pandemic (1899–1923) and the ongoing seventh pandemic (1961–present), but we know very little about the bacteriology of pandemics 1 to 5. Moreover, although we are learning about the contribution of non-O1 non-pandemic V. cholerae to cholera dynamics during the current pandemic, we know almost nothing about their role in the past. A recent ancient DNA study has presented what may be the first molecular evidence of a V. chol

Two recent studies that re-examined through novel approaches previous shotgun sequencing data from prehistoric/historic Europeans uncovered several autosomal and sex chromosome aneuploidies (Anastasiadou et al., 2024; Rohrlach et al., 2024). These disorders, which are common in contemporary humans, were trisomies 18 and 21, Klinefelter syndrome (47,XXY), 47,XYY syndrome, and mosaic Turner syndrome X/XX. These discoveries about prehistoric/historic occurrence of constitutional chromosomal syndromes with high clinical significance in modern medical genetics are an important breakthrough. They co

The expansion of anatomically modern humans (AMHs) from Africa around 65,000 to 45,000 y ago (ca. 65 to 45 ka) led to the establishment of present-day non-African populations. Some paleoanthropologists have argued that fossil discoveries from Huanglong, Zhiren, Luna, and Fuyan caves in southern China indicate one or more prior dispersals, perhaps as early as ca. 120 ka. We investigated the age of the human remains from three of these localities and two additional early AMH sites (Yangjiapo and Sanyou caves, Hubei) by combining ancient DNA (aDNA) analysis with a multimethod geological dating st

INTRODUCTION: The Italian Peninsula and Western Balkans preserve rich archaeological records that extend from pre-Roman times through the Greek colonial, Roman, and Byzantine periods. Genetic investigation of these collections often requires access to specialized high-throughput technologies which are not commonly available or cost-effective method to handle the problem of broken skeletal remains.
METHODS: We developed and validated an ancient DNA protocol for Italian and Albanian archeological skeletal samples spanning approximately 2500 years. The protocol included artificial fragmentation

DNA library construction methods can affect the recovery of ancient DNA, thus influencing downstream analyses. While single-stranded library preparation (ssLib) has been shown to outperform double-stranded (dsLib) for highly degraded vertebrate host DNA, especially for samples older than 40,000 years, few studies have examined how library protocols shape ancient microbial community reconstruction. Here, we compare the sequencing output of paired ssLib and dsLib dental calculus libraries generated from 12 Neanderthals and two Chalcolithic humans, prepared using implementations of the Gansauge e

BACKGROUND: Until now, in the absence of direct microbiological evidence, the cause of the Plague of Athens has remained a matter of debate among scientists who have relied exclusively on Thucydides' narrations to introduce several possible diagnoses. A mass burial pit, unearthed in the Kerameikos ancient cemetery of Athens and dated back to the time of the plague outbreak (around 430 BC), has provided the required skeletal material for the investigation of ancient microbial DNA.
OBJECTIVE: To determine the probable cause of the Plague of Athens.
METHOD: Dental pulp was our material of choic

Crawford Lake has an exceptional stratigraphic record that began recording biannual (varved) sedimentation in the lake basin ~750 years ago, preserving evidence of shifting cultural zones and agricultural practices, from Late Woodland Period Indigenous agriculturalists to the impacts of industrialization during the late 19th century. It was selected as the candidate site for the proposed 'Anthropocene' epoch in 2023-a proposal ultimately rejected in 2024-but the lake's significance extends beyond a formal stratigraphic boundary. Its sediments preserve a long record of human-ecosystem entanglem

Outbreaks of infectious diseases repeatedly affected medieval Europe, leaving behind a large number of dead often inhumed in mass graves. Human remains interred in two burial pits from 14th century CE Germany exhibited molecular evidence of Salmonella enterica Paratyphi C (S. Paratyphi C) infection. The pathogen is responsible for paratyphoid fever, which was likely the cause of death for the buried individuals. This finding presented the unique opportunity to conduct a paratyphoid fever association study in a European population. We focused on HLA-DRB1*03:01 that is a known risk allele for en

Tuberculosis is known to have afflicted humans throughout history and re-emerged towards the end of the 20th century, to an extent that it was declared a global emergency in 1993. The aim of this study was to apply a rigorous analytical regime to the detection of Mycobacterium tuberculosis complex (MTBC) DNA in 77 bone and tooth samples from 70 individuals from Britain and continental Europe, spanning the 1st-19th centuries AD. We performed the work in dedicated ancient DNA facilities designed to prevent all types of modern contamination, we checked the authenticity of all products obtained by

The Nile crocodile (Crocodylus niloticus) is an ancient icon of both cultural and scientific interest. The species is emblematic of the great civilizations of the Nile River valley and serves as a model for international wildlife conservation. Despite its familiarity, a centuries-long dispute over the taxonomic status of the Nile crocodile remains unresolved. This dispute not only confounds our understanding of the origins and biogeography of the 'true crocodiles' of the crown genus Crocodylus, but also complicates conservation and management of this commercially valuable species. We have take

The specific mechanisms through which human biology and disease susceptibility evolved with major shifts in West Eurasian environments and societies over the last 10,000 years(1)-particularly rising infectious burden(2)-remain poorly characterized, despite ancient DNA studies(3-6) identifying hundreds of candidate loci under positive selection(6). Here, we identify specific immune diseases/traits, genes/variants, pathways, and tissues/cell types impacted by natural selection by systematically integrating variant-level selection statistics with genome-wide association study (GWAS), quantitative

Long-term changes in phytoplankton communities are key to understanding and predicting ecosystem responses to climate-driven circulation change. By analyzing sedimentary ancient DNA from two sediment cores in the Yellow Sea, we reconstructed mid-late Holocene phytoplankton dynamics during the intensification of the Yellow Sea Warm Current (YSWC). We found that phytoplankton communities differed significantly between sites, with stronger spatial differentiation in eukaryotes than in cyanobacteria. Dinoflagellates and chlorophytes dominated overall, while Dictyochophyceae (silicoflagellates) wer

Aneuploidies, and in particular, trisomies represent the most common genetic aberrations observed in human genetics today. To explore the presence of trisomies in historic and prehistoric populations we screen nearly 10,000 ancient human individuals for the presence of three copies of any of the target autosomes. We find clear genetic evidence for six cases of trisomy 21 (Down syndrome) and one case of trisomy 18 (Edwards syndrome), and all cases are present in infant or perinatal burials. We perform comparative osteological examinations of the skeletal remains and find overlapping skeletal ma

Historical narratives of early colonization in Quebec frequently portray French-Indigenous relations as cooperative and characterized by admixture; which are often mobilized in modern debates on false-indigeneity. These narratives have been studied through genealogical, genetic, and bioarchaeological analyses, yet individuals of African descent remain mostly absent from them, while Indigenous peoples are framed largely in relation to colonists. Notre-Dame Cemetery (1683-1796) in Tiohtià:ke-Montréal offers a unique window onto one of the earliest urban colonial populations in Nouvelle France, b

Metagonimiasis is foodborne intestinal parasitism occurring by the definitive hosts' ingestion of raw or undercooked fish, mostly commonly sweetfish. Most Metagonimus infection is caused by Metagonimus yokogawai but also rarely by Metagonimus takahashii as well as Metagonimus miyatai. Despite recent molecular work on Metagonimus spp., there are still insufficient data to reveal the genetic characteristics of ancient M. yokogawai in a wide geo-historical scope. In this study, we were successful in the analysis of M. yokogawai ancient DNA (aDNA) using coprolite samples retrieved from 16th- to 17

Structure and function of lake microorganisms are sensitive to environmental disturbances; yet their differential responses to climatic and anthropogenic forcings over centennial scales remain poorly understood. Here, we analyzed a sediment core from Lake Daihai in northern China, combining metagenomics and stable isotope analyses. Our results revealed that microbial community assembly was dominated by deterministic processes, whereas stochasticity increased notably after 1850, coinciding with the termination of the Little Ice Age and intensified human impacts. Notably, microbial community str

Sex identification of ancient animal biological remains can benefit our understanding of historical population structure, demography and social behavior. Traditional methods for sex identification (e.g., osteological and morphometric comparisons) may be ineffective when animal remains are not well preserved, when sex distinguishing characteristics have not yet developed, or where organisms do not exhibit sex-associated phenotypic dimorphisms. Here we adapt a method developed for human sex determination so that it can be used to identify the sex of ancient and modern animal taxa. The method ide
