Placenta-derived decidua stromal cells (DSCs) are being investigated as an alternative to other sources of mesenchymal stromal cells (MSCs) for cellular therapy. DSCs are more effective in treating acute inflammatory diseases in human and this is our preclinical safety study of human DSCs in Sprague-Dawley rats and Balb/c mice. In the present study, using immunohistochemistry, we studied the expression of CD30 and CD30-L in 35 deciduas obtained from women following elective abortion during normal physiological gestation and in 60 deciduas obtained from women after spontaneous abortion with or without signs of inflammation. Hysterosocpic intrauterine findings and levels of human decidua-associated protein 200 (hDP 200) in the uterine fluid were recorded in 116 women investigated for infertility or recurrent abortions. We focus on the roles of decidual and cumulus oophorus T cells. It was suggested that Th1-type cytokines (IFN-gamma and TNF-beta), which promote allograft rejection, may compromise pregnancy, whereas the Th2-type cytokines (IL-4, IL-5, IL-10) inhibiting the Th1 responses promote allograft tolerance and therefore may improve fetal survival. Cytotoxic and suppressor activity of decidual cells lymphoid population were analysed in 14 women with normal pregnancy and labour, in 8 women with light pre-eclampsia and 11 women with early and premature rupture of amniotic sac. In late stages of pregnancy decidua is characterised by high suppressor activity of the lymphoid cell population. Complete placentas and membranes were obtained from women after uncomplicated singleton pregnancies. Six were collected after labour at term, six after preterm labour and six after elective Caesarean section at term. Leukotriene C4 (LTC4), leukotriene D4 (LTD4) and leukotriene E4 (LTE4) release was examined using a short-term incubation technique. Release by amnion was significantly higher than that by the other tissues for the three modes of delivery. Comparison of LTC4, LTD4 and LTE4 release by individual tissues with regard to the type of delivery showed no significant difference.
