Novel targeted bladder drug-delivery systems: a review.

The objective of pharmaceutics is the development of drugs with increased efficacy and reduced side effects. Prolonged exposure of the diseased tissue to the drug is of crucial importance. Drug-delivery systems (DDSs) have been introduced to control rate, time, and place of release. Drugs can easily reach the bladder through a catheter, while systemically administered agents may undergo extensive metabolism. Continuous urine filling and subsequent washout hinder intravesical drug delivery (IDD). Moreover, the low permeability of the urothelium, also described as the bladder permeability barrie

Targeted drug delivery strategies for oral cancer: the role of liposomes in drug delivery systems.

Oral cavity cancer is a common malignancy worldwide, and its aggressiveness and resistance to conventional treatments pose a major therapeutic challenge. Targeted drug delivery strategies have emerged as a promising approach to overcome the limitations of conventional therapies, improving drug efficacy while minimizing systemic side effects. Among the various drug delivery systems, liposomes have gained considerable attention owing to their biocompatibility, ability to encapsulate both hydrophilic and hydrophobic drugs, and surface modification capability to improve targeting accuracy. This re…

Delivery Strategies and Clinical Significance of Oral Colon-targeted Drug Delivery Systems.

Formulation strategies for colonic delivery offer a unique opportunity for the local delivery of drugs for symptomatic relief in inflammatory bowel disease (IBD), irritable bowel syndrome (IBS), and colorectal cancer, as well as for the systemic delivery of peptides and biologics. By releasing active pharmaceutical ingredients (APIs) directly at the site of action within the colon, these systems can achieve higher local exposure with reduced systemic effects. This is achieved by utilizing prodrugs, pH-sensitive polymers, time-dependent systems, pressure, microbiota, and multi-pronged systems.

Topical delivery of aceclofenac: challenges and promises of novel drug delivery systems.

Osteoarthritis (OA), a common musculoskeletal disorder, is projected to affect about 60 million people of total world population by 2020. The associated pain and disability impair the quality of life and also pose economic burden to the patient. Nonsteroidal anti-inflammatory drugs (NSAIDs) are widely prescribed in OA, while diclofenac is the most prescribed one. Oral NSAIDs are not very patient friendly, as they cause various gastrointestinal adverse effects like bleeding, ulceration, and perforation. To enhance the tolerability of diclofenac and decrease the common side effects, aceclofenac

Novel drug delivery systems: potential in improving topical delivery of antiacne agents.

Acne is the most common cutaneous disorder of multifactorial origin with a prevalence of 70-85% in adolescents. The majority of the acne sufferers exhibit mild to moderate acne initially, which progresses to the severe form in certain cases. Topical therapy is employed as first-line treatment in mild acne, whereas for moderate and severe acne, systemic therapy is required in addition to topical therapy. Currently, several topical agents are available that affect at least one of the main pathogenetic factors responsible for the development of acne. Although topical therapy has an important posi

Microneedle geometry modulates drug transport in transdermal delivery systems.

Microneedles have emerged as a promising strategy for transdermal drug delivery, owing to their ability to bypass the stratum corneum in a minimally invasive manner and enable controlled drug administration. In practice, microneedles have been developed with a wide range of geometries to meet diverse requirements in delivery efficiency, targeting, and release kinetics. However, the role of microneedle geometry in governing delivery performance remains incompletely understood. In this study, a modelling framework is developed to systematically investigate the effects of microneedle geometry on

Mechanistic insights into ultrasound-responsive implantable drug delivery systems.

Stimulus-responsive drug delivery systems have been widely explored to achieve controlled, site-specific drug release. Among various stimuli, ultrasound offers several advantages as an external trigger, including cost-effectiveness, deep tissue penetration, and precise spatiotemporal control. While hydrogel and microbubble associated systems have been extensively studied, reservoir-type implants remain underexplored despite their potential for long-term therapy. In this study, a biodegradable polymeric reservoir implant with porous rate-controlling membrane was developed, with systematically e…

Recent Advances in Endogenous Gas-Responsive Drug Delivery Systems.

Recent advances in Drug Delivery Systems (DDS) have focused on minimizing systemic side effects while enhancing therapeutic precision. Endogenous gas-responsive DDS represent a promising approach that utilizes biologically active gasotransmitters, including carbon monoxide (CO), nitric oxide (NO), and hydrogen sulfide (H2S), as internal triggers for targeted drug release. These gases play crucial roles in physiological processes such as tissue repair, vasodilation, and the regulation of inflammation, underscoring their significance in biological systems. However, their therapeutic application …

Exploring Hydrogel Nanoparticle Systems for Enhanced Ocular Drug Delivery.

Drug delivery to the ocular system is affected by anatomical factors like the corneal epithelium, blinking reflex, aqueous blood barrier, and retinal blood barrier, which lead to quick removal from the site and inefficient drug delivery. Developing a drug delivery mechanism that targets specific eye tissue is a major hurdle for researchers. Our study examines the challenges of drug absorption in these pathways. Hydrogels have been researched as a suitable delivery method to overcome some obstacles. These are developed alone or in conjunction with other technologies, such as nanoparticles. Many

Cancer Drug Delivery Systems Using Bacterial Toxin Translocation Mechanisms.

Recent advances in targeted cancer therapy hold great promise for both research and clinical applications and push the boundaries in finding new treatments for various currently incurable cancers. However, these therapies require specific cell-targeting mechanisms for the efficient delivery of drug cargo across the cell membrane to reach intracellular targets and avoid diffusion to unwanted tissues. Traditional drug delivery systems suffer from a limited ability to travel across the barriers posed by cell membranes and, therefore, there is a need for high doses, which are associated with adver…

Liposomal drug delivery systems for the treatment of leishmaniasis.

Human le ishmaniasis is a vector-borne, neglected infectious disease that is widely distributed in America, Africa, Europe, and Asia. Current therapy is based on old and toxic drugs, including antimonials, aminoglycosides, and amphotericin. As a neglected disease, investment in the development of new therapeutic molecules is scarce. Considering these aspects, the optimization of treatment through novel delivery systems for current therapeutic agents is an attractive alternative. The encapsulation into liposomes of drugs used in treating leishmaniasis increases the concentration of these molecu

Recent Advances in Polymer-Based Vaginal Drug Delivery Systems.

The vagina has been considered a potential drug administration route for centuries. Most of the currently marketed and investigated vaginal formulations are composed with the use of natural or synthetic polymers having different functions in the product. The vaginal route is usually investigated as an administration site for topically acting active ingredients; however, the anatomical and physiological features of the vagina make it suitable also for drug systemic absorption. In this review, the most important natural and synthetic polymers used in vaginal products are summarized and described

Evolution of Drug Delivery Systems for Recurrent Aphthous Stomatitis.

Recurrent aphthous stomatitis (RAS) is a disease marked by painful oral lesions on the buccal and labial mucosa or tongue. Drug delivery systems (DDS) for RAS include topical forms that manage wound healing, cover the ulcer, and relieve the associated pain. DDS targeting the oral mucosa face a major challenge, especially the short residence times in the mouth due to the effect of "saliva wash-out", which continually removes the drug. The objective of this review is to study the development of preparation forms and delivery systems of various types and preparations that have been used for RAS m…
