Mammalian embryonic development starts from a fertilized egg, which cleaves to form morula and blastocyst. At the same time, the early embryo is transported from the fallopian tube to the uterus for implantation. After implantation, the embryo undergoes gastrulation and forms a gastrula, further developing a new individual. The development of embryo in the uterus causes the difficulties in sampling and observation, hindering the understanding of mammalian embryonic development. Therefore, it is necessary to develop the technology to overcome the barrier of in vivo embryonic development. In Dec

Uterine (endometrial) receptivity is the ability of the endometrium to successfully attach to the embryo, to promote its implantation, to nourish it and keep it alive [...].

High-content time-lapse embryo imaging assessed by machine learning is revolutionizing the field of in vitro fertilization (IVF). However, the vast majority of IVF embryos are not transferred to the uterus, and these masses of embryos with unknown implantation outcomes are ignored in current efforts that aim to predict implantation. Here, whether, and to what extent the information encoded within "sibling" embryos from the same IVF cohort contributes to the performance of machine learning-based implantation prediction is explored. First, it is shown that the implantation outcome is correlated

Communication between the maternal endometrium and developing embryo/conceptus is critical to support successful pregnancy to term. Studying the peri-implantation period of pregnancy is critical as this is when most pregnancy loss occurs in cattle. Our current understanding of these interactions is limited, due to the lack of appropriate in vitro models to assess these interactions. The endometrium is a complex and heterogeneous tissue that is regulated in a transcriptional and translational manner throughout the oestrous cycle. While there are in vitro models to study endometrial function, th

The vertebrate intestine has a continuous dorsal mesentery between pharynx and anus that facilitates intestinal mobility. Based on width and fate the dorsal mesentery can be subdivided into that of the caudal foregut, midgut, and hindgut. The dorsal mesentery of stomach and duodenum is wide and topographically complex due to strong and asymmetric growth of the stomach. The associated formation of the lesser sac partitions the dorsal mesentery into the right-sided "caval fold" that serves as conduit for the inferior caval vein and the left-sided mesogastrium. The thin dorsal mesentery of the mi

We present a reconstruction of the aortic arches of a human embryo at stage 13. Only a few original observations have been reported during the last 110 years and only two with three-dimensional reconstructions. The reconstruction technique originated in 1885. Modern computerized reconstruction techniques have no evident advantages and are more expensive. Aortic arch malformations are responsible for disorders which cannot be understood without a knowledge of their embryology. The embryonic aortic system is a typical example of Haeckel's law of recapitulation. A human embryo at stage 13 was pre

We studied the histopathologic features of the temporal bones in a human embryo with unilateral aural atresia. The developmental stage of the embryo was at stage 22 in the Carnegie system, and the estimated ovulation age was 8 weeks. There were severe hypoplastic changes in Meckel's and Reichert's arch cartilages without differentiation of the auditory ossicles, hypoplasia of the tubotympanic recess, and resultant abnormal passing of the facial nerve in the affected ear. Abnormal lateral extension of the cartilaginous otic capsule replaced a posterior half of the middle ear region and seemed t

Hand plates with preaxial polydactyly from 13 human embryos of the Carnegie stages 17-23 were examined macro- and microscopically. Morphological features in early pathogenesis of preaxial polydactyly are (1) an abnormal extension and a delayed involution of the apical ectodermal ridge on the preaxial border of the hand plate in stages 17 and 18, (2) a precocious development of an interdigital notch between the duplicated thumbs in stages 17 and 18, and (3) bifurcation of the distal part of the first digital ray in stage 19. A disorder of the interaction between limb ectoderm and mesoderm is co

Human embryo implantation has remained largely inaccessible to mechanistic study. Two recent Cell papers by Molè et al.1 and Li et al.2 now introduce complementary in vitro platforms that reconstruct key features of the human implantation niche, enabling interrogation of embryo-endometrium interactions and opening translational avenues to model implantation failure and test therapeutic strategies.

Both uterine endometrium and embryo contribute to implantation success. However, their relative role in the implantation success is still a matter for debate, as are the roles of endometrial receptivity analysis (ERA), endometrial scratch (ES), endometrial microbiome, and intrauterine or intravenous measures that are currently advocated to improve the implantation success. There is insufficient evidence to suggest that the endometrium is more important than the embryo in determining the implantation success and the utility of these measures, especially when euploid embryos are transferred is l

Mammalian embryos produced in vitro have poor embryo quality and low developmental ability compared with in vivo embryos. The main manifestations are the low number of blastocysts, the low ratio of the number of inner cell mass cells to the number of trophoblastic cells, and the high apoptosis rate of blastocysts, resulting in low embryo implantation rate. Therefore, optimizing in vitro culture conditions has become a key technology to im-prove the quality of preimplantation embryos. Oviduct Epithelial cells exosomes (OEVs) can be absorbed and internalized by embryos to improve the blastocyst

The biology of fertility, early development, and pregnancy is variable across mammalian species. In addition, while the physiology and pathophysiology of human diseases can be investigated in other animal models (principally, rodents), differences between human and lower mammals often present limitations in the applicability of physiological processes from rodent models to human biology. Since 1984, when the first live birth from rhesus monkey in vitro fertilization and embryo transfer was reported (Bavister et al., Proc Natl Acad Sci 81:2218-2222, 1984), there has been progress in the impleme
