Non-steroid hormones and tissue polypeptide antigen (TPA) in emetic and non-emetic pregnancy.

In order to investigate further the endocrine and metabolic features of the common condition emesis gravidarum, serum concentrations of some non-steroid hormones and tissue polypeptide antigen (TPA) were determined in 102 healthy pregnant women. 62 complained of nausea and vomiting in early pregnancy. Significantly higher and lower levels of human chorionic gonadotropin were noted in early and late pregnancy, respectively, in women with emesis gravidarum. A significant rise in serum prolactin and TPA was found throughout pregnancy in all subjects, no differences between emetic and non-emetic p

Effects of anti-emetics on water excretion in humans.

1. The anti-emetic drug metoclopramide has been shown to stimulate secretion of the antidiuretic hormone arginine vasopressin. Since metoclopramide is used to treat nausea, which is another potent stimulus to vasopressin secretion, the aim of this study was to determine whether metoclopramide might limit free water excretion and so cause hyponatraemia. 2. Metoclopramide 20 mg (0.2-0.3 mg/kg), prochlorperazine 12.5 mg (0.1-0.2 mg/kg) and placebo were administered intravenously in a double blind randomized crossover way at 2 week intervals and the effects on urine flow rate, plasma osmolality, t

Evaluation of anti-emetics in association with intrathecal diamorphine.

Intrathecal diamorphine is associated with a high incidence of emetic symptoms. Six anti-emetic drugs representing various chemical groups were given in random order to patients undergoing total hip replacement and who had received intrathecal diamorphine 0.5-1.0 mg. The phenothiazines, perphenazine and prochlorperazine, were more effective than the others. It is suggested that this might be a useful model for the evaluation of new anti-emetics.

An outline of anti-emetic treatment.

With the advent of new chemotherapeutic agents, and their well-known side effect of emesis, the need for a greater facility with anti-emetics has emerged. As well as the common problem of chemotherapy-induced emesis, other problems have become apparent such as delayed emesis and anticipatory emesis. The control of such emetic problems may be affected by certain patient characteristics, such as a history of chronic high alcohol intake, and age. Blockade of the different types of neuroreceptors can lead to effective emetic control. Dopamine receptor blockers such as the phenothiazines, butyrophe…

Advances in anti-emetic therapy.

Evaluation of the activity of anti-emetic drugs in randomized controlled trials has, in most cases, demonstrated the superiority of treatmen … fficacy of metoclopramide when used in combination. For the future, the use of combinations of anti-emetics with differing sites of action and non-overlapping toxicities, may lead to further improvement in efficacy. Combinations of centrally-acting drugs such as the cannabinoids plus dopamine antagonists such as metoclopramide or domperidone, are worth ev …

Assessment of emetic and antiemetic activity.

Compounds having acute emetic or antiemetic activities have been quantitatively evaluated and compared with the reference standards, apomorphine and chlorpromazine, respectively. The bioassay for an emetic required the establishment of mean threshold emetic doses in groups of dogs for both the reference standard apomorphine and the test compound. The bioassay for antiemetics was based upon the degree to which the test drug elevated the threshold emetic dose for apomorphine. That effect was contrasted with the change in emetic threshold produced by equimolar concentrations of chlorpromaine.
