Ensuring Epidemiological Consistency in Risk-Stratified Cancer Screening Models: A Novel Approach Based on Flemish Breast Cancer Screening.

OBJECTIVE: We present a novel dynamic risk-stratified breast cancer screening Markov model designed to maintain robust and epidemiologically consistent cancer prevalences, regardless of the number or composition of risk-stratification groups. This approach addresses a common limitation in existing models, where altering risk-group definitions or proportions can unintentionally distort overall incidence rates. By overcoming this constraint, the method lowers barriers to developing interactive, flexible and policy-relevant models that can be shared directly with decision makers. The paper serves

Epidemiological trends and comparative forecasting models of human brucellosis in inner mongolia autonomous region, mainland China, 2004-2024.

INTRODUCTION: Brucellosis remains a severe zoonotic threat in the Inner Mongolia Autonomous Region of China. METHODOLOGY: This study integrates a comprehensive epidemiological trend analysis with a novel methodological comparison of forecasting techniques to inform control strategies. RESULTS: Using reported human brucellosis surveillance data from Inner Mongolia for 2004-2024, joinpoint regression analysis revealed a persistently increasing yet fluctuating long-term trend (AAPC = 5.13%, P < 0.001), characterized by significant epidemic surges in 2004-2010 (APC = 22.43%, P < 0.001) and 2016-

Integrating epidemiological and economic models to estimate the cost of simulated foot-and-mouth disease outbreaks in Brazil.

The introduction of foot-and-mouth disease (FMD) leads to substantial economic impacts through animal loss, decreased livestock and meat production, increased government and private spending on control and eradication measures, and trade restrictions. This study evaluates the direct cost-effectiveness of four control and eradication scenarios of hypothetical FMD outbreaks in Rio Grande do Sul, Brazil, from August 2022 to August 2023. Our model simulation considered scenarios with depopulation of detected farms and emergency suppressive vaccination, and two enhanced scenarios featuring increase

Later life depression as risk factor for developing dementia: epidemiological evidence, predictive models, preventive strategies and future trends.

Current investigations in pre-symptomatic dementia have suggested that depressive mood, a treatable condition, may play an important role in the development of the disorder. However, whether depression in adulthood constitute a risk factor, or a prodrome of dementia remains unclear. A major implication in such dispute is the analytic framework used to identify putative risk factors. Indeed, if evaluated in the years immediately prior to dementia diagnosis the association between depression and dementia may reflect depressive symptoms as a prodrome of yet-undiagnosed dementia. Unfortunately, lo

Predictive testing of metalworking fluids: a comparison of 2 cumulative human irritation models and correlation with epidemiological data.

Metalworking fluids (MWF) have been reported as being an important cause of irritant contact dermatitis in metal workers. Our purpose was to determine whether the irritancy of different MWF assessed by 2 different types of predictive human in vivo tests could be compared with epidemiological data. 3 water-based MWF were tested in the same panel of subjects. Reactions were assessed by a visual score (VS), evaporimetry to evaluate the transepidermal water loss (TEWL) and chromametry to quantify erythema. Test 1: MWF were applied with Finn Chambers on the volunteers' mid-back, removed after 1 day

Stroke-prone SHR (SHRSP) as models for clinical and epidemiological studies on hypertension-related cardiac diseases in humans.

Spontaneously hypertensive rats (SHR) and stroke-prone SHR (SHRSP) were vectorcardiographically and pathologically studied for myocardial lesions with left ventricular hypertrophy, and findings were compared with those in normotensive Wistar-Kyoto rats (WK). Characteristic vectorcardiogram (VCG) in adult SHR was left superior (posterior) deviation of major QRS portion usually with ST-T changes, which corresponded to the pathological findings such as increased weight of the heart and the left ventricle. In old SHRSP, VCG showed deformity and deviation of QRS loop with prolonged P and QRS durati

A decision-theoretic framework for uncertainty quantification in epidemiological modelling.

Estimating, understanding, and communicating uncertainty is fundamental to statistical epidemiology, where model-based estimates regularly i … nds many of these ideas. Our article serves as a gateway for applying a wide range of approaches to epidemiological models. Altogether, our framework provides a foundation for more reliable, consistent, and policy-relevant uncertainty quantification in infectious disease epidemiology.

Understanding Huntington's Disease: Epidemiology, Mechanisms, and Modeling Approaches.

Huntington's disease (HD) is a monogenic, autosomal dominant neurodegenerative disorder. Huntington's disease is caused by a CAG trinucleotide repeat expansion in exon 1 of the huntingtin gene, which is located on the short arm of chromosome 4. Although HD has a well-defined genetic cause, the underlying molecular and cellular mechanisms remain complex and incompletely understood. This review examines the established functions of normal huntingtin protein and discusses the harmful consequences of CAG repeat expansions, which result in abnormally extended polyglutamine tracts. In this review, w

Modelling techniques in cholera epidemiology: A systematic and critical review.

Diverse modelling techniques in cholera epidemiology have been developed and used to (1) study its transmission dynamics, (2) predict and manage cholera outbreaks, and (3) assess the impact of various control and mitigation measures. In this study, we carry out a critical and systematic review of various approaches used for modelling the dynamics of cholera. Also, we discuss the strengths and weaknesses of each modelling approach. A systematic search of articles was conducted in Google Scholar, PubMed, Science Direct, and Taylor & Francis. Eligible studies were those concerned with the dynamic

Sensitivity Analysis in an Immuno-Epidemiological Vector-Host Model.

Sensitivity Analysis (SA) is a useful tool to measure the impact of changes in model parameters on the infection dynamics, particularly to q … ithin-host virus and antibody densities, governed by an ODE system. We then use SA for these immuno-epidemiological models to investigate the impact of immunological parameters on population-level disease dynamics such as basic reproduction number, final size of the epidemic or the infectiousness at different phases of an outbreak. As a case study, we consider Rift Vall …

A mechanistic hydro-epidemiological model of liver fluke risk.

The majority of existing models for predicting disease risk in response to climate change are empirical. These models exploit correlations between historical data, rather than explicitly describing relationships between cause and response variables. Therefore, they are unsuitable for capturing impacts beyond historically observed variability and have limited ability to guide interventions. In this study, we integrate environmental and epidemiological processes into a new mechanistic model, taking the widespread parasitic disease of fasciolosis as an example. The model simulates environmental s

Quantifying Zika: Advancing the Epidemiology of Zika With Quantitative Models.

When Zika virus (ZIKV) emerged in the Americas, little was known about its biology, pathogenesis, and transmission potential, and the scope of the epidemic was largely hidden, owing to generally mild infections and no established surveillance systems. Surges in congenital defects and Guillain-Barré syndrome alerted the world to the danger of ZIKV. In the context of limited data, quantitative models were critical in reducing uncertainties and guiding the global ZIKV response. Here, we review some of the models used to assess the risk of ZIKV-associated severe outcomes, the potential speed and s

Connecting the dots: network data and models in HIV epidemiology.

Effective HIV prevention requires knowledge of the structure and dynamics of the social networks across which infections are transmitted. These networks most commonly comprise chains of sexual relationships, but in some populations, sharing of contaminated needles is also an important, or even the main mechanism that connects people in the network. Whereas network data have long been collected during survey interviews, new data sources have become increasingly common in recent years, because of advances in molecular biology and the use of partner notification services in HIV prevention and tre

Invited commentary: structural equation models and epidemiologic analysis.

In this commentary, structural equation models (SEMs) are discussed as a tool for epidemiologic analysis. Such models are related to and compared with other analytic approaches often used in epidemiology, including regression analysis, causal diagrams, causal mediation analysis, and marginal structural models. Several of these other approaches in fact developed out of the SEM literature. However, SEMs themselves tend to make much stronger assumptions than these other techniques. SEMs estimate more types of effects than do these other techniques, but this comes at the price of additional assump

[Modelling the impact of vaccination on the epidemiology of varicella zoster virus].

BACKGROUND: The soon to come the availability of a combined MMR-varicella vaccine has re-stimulated the debate around universal infant vaccination against varicella. In France, the incidence of varicella is estimated at about 700,000 cases per year, with approximately 3500 hospitalisations and 15-25 deaths, the latter mainly occurring in those over 15years. Vaccination would certainly decrease the overall incidence of the disease but concerns about vaccination leading to a shift in the average age at infection followed by an increase in incidence of severe cases and congenital varicella, still

Epidemiological modelling for monitoring and evaluation of lymphatic filariasis control.

Monitoring and evaluation are critically important to the scientific management of any parasite control programme. If a management action is prescribed, monitoring plays a pivotal role in assessing the effectiveness of implemented actions, identifying necessary adaptations for management and determining when management objectives are achieved. Here, we focus on the control of the vector-borne parasitic disease, lymphatic filariasis, to show how mathematical models of parasite transmission can provide a scientific framework for supporting the optimal design of parasite control monitoring progra

Epidemiology of prenatal infections: an extension of the congenital rubella model.

The properties of prenatal infections are studied through the development of a theoretical model. The responses of such epidemic systems to natural changes in transmission rates, age at maternity, social stratification, geographical heterogeneity, epidemic instability and the introduction of vaccination, are examined. The effects of age-at-vaccination, efficacy of vaccination, wild virus interaction, and vaccine-immunity decay rates, are also examined in some detail. Several powerful putative influences upon the evolution of prenatally determined infective diseases are identified. They provide

An epidemiological model with arbitrary distributions for infection and relapse stages.

In low tuberculosis (TB) burden settings, recurrent tuberculosis is predominantly driven by relapse. Relapse, defined as the recurrence or re-emergence of a disease or condition after a period of remission or apparent recovery, poses a significant global public health challenge. The variability in the duration of infection and relapse stages among individuals calls for a rigorous mathematical framework to evaluate the impact of this heterogeneity on disease transmission dynamics. To address this, we develop a general integral equation model tailored to low TB burden settings, incorporating arb

Epidemiology and prediction model of patients with carcinosarcoma in the United States.

BACKGROUND: Carcinosarcoma is a rare biphasic tumor composed of both carcinoma and sarcoma elements, which occurs at various sites. Most studies are case reports or small population-based studies for a single disease site, so comprehensive evaluations of epidemiology and prognostic factors for carcinosarcoma are needed. METHODS: Surveillance, Epidemiology, and End Results (SEER)-8 (1975-2019) provided data for the epidemiological analysis. SEER-17 (2000-2019) provided data on the primary tumor sites, initial treatment, construction, and validation of the nomogram. RESULTS: The age-adjusted i
