BACKGROUND: Ulnar mammary syndrome (UMS) is an autosomal dominant disorder caused by heterozygous pathogenic variants in the T-box transcription factor 3 (TBX3) gene. The phenotype is classically characterized by upper limb defects and apocrine/mammary gland hypoplasia. Endocrine abnormalities include hypogonadotropic hypogonadism (HH), partial growth hormone deficiency and dysmorphic features, while ectopic pituitary gland and various congenital anomalies have also been described. Here, we report a family with a unique clinical presentation.
METHODS: Exome sequencing was performed for twin s

This paper describes approaches to care and associated service use by families with a child with autism. A combined telephone and self-administered survey was completed by 301 families with a child, 8 years old or younger, in North Carolina, during the winter of 2003-2004. Findings indicate that 66% of families used one or more approach to care and there was a significant (p < 0.05) association between approach and the pattern of service use. There appears to be a distinctive set of services associated with each approach to care, but with overlap between them. Speech/language therapy at school

Family history of alcohol abuse and personality characteristics measured by the MacAndrew Alcoholism Scale (MAC), two factors that have been associated with the development and expression of alcoholism in clinical populations, were examined in a nonclinical, putatively normal sample. Analyses revealed that although family history positive subjects (FHP) drank as much and as often as family history negative subjects (FHN), the FHP subjects experienced significantly more alcohol-related problems. Consistent with previous findings with clinical samples, both family history of alcohol abuse and th

We describe a 19-year-old girl with Kindler syndrome. She has suffered from bullae on pressure areas of the skin since birth. These healed with atrophic scars and caused marked atrophy of the skin of the palms and soles and wrinkled and parchment-like skin of the dorsum of the hands and feet. Since infancy the patient has also suffered from severe photosensitivity on exposed areas and developed poikiloderma on the skin of her face, neck, chest, and upper back. Since age 17 years the patient has been free of bullae but moderate photosensitivity exists. Oral examination showed limitation of mout

The importance of family relations for the therapy of psychiatric disorders in children and adolescents is discussed reporting results from an empirical study carried out in an inpatient sample of a child and adolescent psychiatric unit with regard to the quality of mother-child resp, father-child relation and its influence on coping processes. Whether adolescents suffering from a psychiatric disorder are able to cope with their problems during the course of a therapy, depends on their individual and social resources. Therapy outcome is to a great extent influenced by the emotional quality of …

Background: Keratoconus (KC) is a complex corneal disorder with a strong genetic component. The present study aimed to identify candidate genes related to KC in Chinese families. Methods: Family-based exome sequencing was performed in ten patients suffering from KC who belong to five families with two affected members in each. The candidate rare variants were identified with multi-step bioinformatics analysis. The STRING website was used to perform the protein interaction of the identified genes. Results: Our analyses identified 32 candidate rare variants in 13 genes by family-based exome sequ

To explore the association between infertility-related stress, family adaptability and family cohesion in infertile couples and the determinants of infertility-related stress in infertile couples. Fertility Problem Inventory (FPI) and Family Adaptability and Cohesion Evaluation Scales (FACESII-CV) were used to measure the infertility-related stress and family adaptability and cohesion of infertile couples. T-test, ANOVA and multiple comparisons (LSD) were conducted to compare the FPI scores of different demographic characteristics subgroups. Stepwise multivariate linear regression was used to

The current systematic review summarizes the evidence from studies examining the risk and protective factors associated with family related violence in refugee families. Data included 15 peer-reviewed qualitative and quantitative studies. In order to gain an overview of the identified risk and protective factors an ecological model was used to structure the findings. At the individual level, parental trauma experiences/mental illness, substance abuse and history of child abuse were found to be risk factors. Family level risk factors included parent-child interaction, family structure and famil

In this study, we examined whether family adversity and parental marital quality were associated with family-of-origin relational ethics, a hallmark feature of contextual therapy theory. Data were collected from 211 adult clients at a university-based therapy center between 2014 and 2018. A three-step hierarchical regression model was tested, controlling for age and gender. Parental marital happiness and parental infidelity, as well as indicators of family adversity such as physical abuse experiences and living with a parent or caregiver with drug dependence in childhood, significantly correla

Introduction: Jeffry Hyman (1951-2001), an introvert boy from Queens, New York City, grew up to become one of the most iconic figures in New York's punk rock scene in the mid-1970s: Joey Ramone. In this study, we explored how Joey and his family coped with his health issues and peculiar behaviors, which were eventually identified as symptoms of obsessive-compulsive disorder (OCD). Methods: A narrative review of biographical sources was conducted to explore and infer the impact of Joey's OCD on his close relationships. Results: The onset of OCD and concomitant hoarding behaviors during his teen

Amyloidosis has been well known since Rudolph Virchow named the condition in the 19th century. Most physicians were aware of the association between amyloidosis to chronic suppurative conditions and multiple myeloma. However, familial Mediterranean fever, although probably as ancient as the Galenic era, was appropriately identified only in the 21st century. The nomenclature was variable throughout history, but the name "FMF" has been universally adopted since the report from Heller and colleagues in 1958. In 1967, Sokmen and Ozdemir of Ankara University published a report on 194 patients, of w

We report a case of a male fetus born to an unrelated couple with a fetal phenotype of an omphalocele and inferior vermian hypoplasia. Prenatal trio exome sequencing identified a maternally inherited pathogenic CDKN1C variant consistent with Beckwith-Wiedemann syndrome (BWS). This finding prompted targeted testing of the proband's sibling, who was confirmed to carry the same variant. Posterior fossa abnormalities have been reported in cases with BWS, and specifically in children with the CDKN1C loss-of-function variant.

Age-related macular degeneration (AMD) is the leading cause of blindness in aging adults, yet no FDA-approved therapies exist for early and intermediate disease. Multiple interconnected pathways, including mitochondrial dysfunction, oxidative stress, and inflammation, contribute to retinal pigment epithelium (RPE) degeneration in AMD, highlighting the need for developing new cytoprotective strategies. The MTRNR2L gene family, comprising nuclear paralogs of mitochondrial-derived peptide humanin, has emerged as a potential regulator of these pathways. Humanin has demonstrated anti-apoptotic, ant

BACKGROUND: Women with a family history of breast and/or ovarian cancer have an increased ovarian cancer risk. Yet it remains uncertain if common ovarian cancer risk factors-especially those that are modifiable-affect this high-risk population similarly to the general population.
METHODS: Using the Danish and Swedish nationwide registers, we established 2 nested case-control study populations in women with a family history of breast and/or ovarian cancer (2138 ovarian cancers, 85 240 controls) and women without (10 730 ovarian cancers, 429 200 controls). The overall and histology-specific ass

The aim of our study was to determine regions of loss of heterozygosity, copy number variation analysis, and single nucleotide polymorphisms (SNPs) in Brazilian patients with cystinuria. A linkage study was performed using DNA samples from six patients with cystinuria and six healthy individuals. Genotyping was done with the Genome-Wide Human SNP 6.0 arrays (Affymetrix, Inc., Santa Clara, CA, USA). For validation, SNPs were genotyped using a TaqMan® SNP Genotyping Assay Kit. The homozygote polymorphic genotype of SNP rs17383719 in the gene PBX1 was more frequent (P = 0.015) in cystinuric patie

We describe a family with an autosomal dominant familial dyskinesia resembling myoclonus-dystonia associated with a novel missense mutation in ADCY5, found through whole-exome sequencing. A tiered analytical approach was used to analyse whole-exome sequencing data from an affected grandmother-granddaughter pair. Whole-exome sequencing identified 18,000 shared variants, of which 46 were non-synonymous changes not present in a local cohort of control exomes (n = 422). Further filtering based on predicted splicing effect, minor allele frequency in the 1000 Genomes Project and on phylogenetic cons
