Further mechanistic study on intestinal absorption enhanced by unsaturated fatty acids: reversible effect by sulfhydryl modification.

In order to study the relationship between the sulfhydryl (SH) modification of membrane-associated proteins and the oleic acid-induced permeability enhancement of the colonic mucosa, in vitro and in situ absorption studies were performed using rat colon and carboxyfluorescein as an impermeable dye. The pretreatment of the mucosa with diamide, a bifunctional sulfhydryl modifier, in in vitro experiments with the everted colonic loops reduced the absorption enhancing effect of oleic acid in a concentration-dependent manner, less inhibitory effect, though just a little, was observed as compared to

Possible involvement of direct stimulation of protein kinase C by unsaturated fatty acids in platelet activation.

Arachidonate and other unsaturated fatty acids stimulated platelet protein kinase C in a dose-dependent manner (5-50 micrograms/ml), when the activity was assayed with either isolated substrates or the platelet cytosol. When human platelets were stimulated by arachidonate, two types of platelet activation were observed. Platelet activation induced by a low level of arachidonate (0.1-5 micrograms/ml) was inhibited by aspirin, but activation induced by a high level of arachidonate (10-50 micrograms/ml) was not. These activations were associated with the phosphorylation of 40K and 20K proteins. O

Inhibitory effect of unsaturated fatty acids on lymphocyte-antigen interaction with special reference to multiple sclerosis.

Using the macrophage electrophoretic mobility (MEM) test, the in vitro effects of oleic, linoleic and arachidonic acid, as well as Naudicell oil and methyl gamma-linolenate, on lymphocyte response to various antigens (PPD, thyroid antigen and EF) were studied in patients with mutiple sclerosis, patients with other neurological diseases, and in normal subjects. In all three groups, linoleic and arachidonic acid showed a significant inhibition; the latter was greater than the former, when tested at physiological concentrations, suggesting that unsaturated fatty acids may exert an immunoregulator

θ-Metallacarboranes as Sustainable Photoredox Oxidation Catalysts of Unsaturated Fatty Acids From Oil and Pork Lard.

The sustainable management and valorization of unsaturated fatty acid-rich lipid streams is a key challenge within circular economy strategies. Herein, we present a proof of concept for the photoredox oxidation of unsaturated fatty acids using θ-metallacarborane complexes Na[Co(1,2-C2B9H11)2] ([Na]1) and Na[Fe(1,2-C2B9H11)2] ([Na]2) as water-soluble, earth-abundant photoredox catalysts. Commercial sunflower oil and pork lard served as convenient model systems mimicking waste lipid streams, while targeting future application to postconsumer used cooking oils. Under UVA irradiation in aqueous me

Diversity of Susceptibility to Unsaturated Fatty Acids in Enterococcus Strains and Their Synergistic Effects With Antibiotics.

Enterococci are commensal bacteria in humans that are found in the intestine, pharynx and oral cavity and sometimes cause opportunistic infections such as intra-abdominal infections and bacteraemia. Several enterococcal species, especially E. faecium and E. faecalis, have become clinical problems as drug-resistant bacteria, including vancomycin-resistant Enterococci (VRE). Therefore, new antibacterial agents against enterococcal infections are needed. Because several fatty acids (FAs) have been reported to possess antibacterial activity, in this study, the effects of 3 unsaturated fatty acids

Long-chain unsaturated free fatty acids reduce the host cell invasion of Listeria monocytogenes outbreak strains.

The Gram-positive bacterium Listeria monocytogenes is a highly adaptable pathogen capable of causing severe foodborne infections, particularly in vulnerable populations. During infection, L. monocytogenes uses a variety of virulence factors to invade and multiply within host cells. The transcriptional regulator PrfA controls the expression of these virulence factors and is essential for the intracellular lifestyle of L. monocytogenes. Long-chain unsaturated free fatty acids (FFAs) have long been recognized for their antimicrobial activity and were recently shown to inhibit PrfA-dependent virul

The Staphylococcus aureus response to unsaturated long chain free fatty acids: survival mechanisms and virulence implications.

Staphylococcus aureus is an important human commensal and opportunistic pathogen responsible for a wide range of infections. Long chain unsaturated free fatty acids represent a barrier to colonisation and infection by S. aureus and act as an antimicrobial component of the innate immune system where they are found on epithelial surfaces and in abscesses. Despite many contradictory reports, the precise anti-staphylococcal mode of action of free fatty acids remains undetermined. In this study, transcriptional (microarrays and qRT-PCR) and translational (proteomics) analyses were applied to ascert

Effects of unsaturated fatty acids on calcium-activated potassium current in gastric myocytes of guinea pigs.

AIM: To investigate the effects of exogenous unsaturated fatty acids on calcium-activated potassium current (I(K(Ca))) in gastric antral circular myocytes of guinea pigs. METHODS: Gastric myocytes were isolated by collagenase from the antral circular layer of guinea pig stomach. The whole-cell patch clamp technique was used to record I(K(Ca)) in the isolated single smooth muscle cells with or without different concentrations of arachidonic acid (AA), linoleic acid (LA), and oleic acid (OA). RESULTS: AA at concentrations of 2,5 and 10 micromol/L markedly increased I(K(ca)) in a dose-dependent

Acidic phospholipids with unsaturated fatty acids inhibit the binding of origin recognition complex to origin DNA.

Origin Recognition Complex (ORC) is a candidate initiator of chromosomal DNA replication in eukaryotes. We recently reported that cardiolipin inhibits the interaction of Origin Recognition Complex ORC with origin DNA, as is the case of DnaA, the initiator of chromosomal DNA replication in prokaryotes. We report here that another acidic phospholipid, phosphatidylglycerol (PG), also inhibits the interaction. Synthetic PG with only unsaturated fatty acids inhibits ORC-binding to origin DNA more strongly than PG with only saturated fatty acids. On the other hand, phosphatidylcholine (neutral phosp

Free radical generation in human leukocytes by CIS-unsaturated fatty acids is a calmodulin dependent process.

The mechanism(s) involved in the generation of free radicals in human leukocytes by cis-unsaturated fatty acids, gamma-linolenic acid (GLA), and arachidonic acid (AA), was investigated. Calmodulin antagonists, chlorpromazine and trifluoperazine, inhibited free radical generation by human leukocytes in vitro induced by GLA, AA PMA (Phorbol myristate acetate), formyl-methionyl-leucyl-phenylalanine (FMLP) and lipopolysaccharide (LPS). On the other hand, chloroquine, a phospholipase A2 inhibitor, and colchicine, a microtubule disrupting agent, were without any effect. When sub-optimal concentratio

Serum bile acid and unsaturated fatty acid profiles of non-alcoholic fatty liver disease in type 2 diabetic patients.

BACKGROUND: The understanding of bile acid (BA) and unsaturated fatty acid (UFA) profiles, as well as their dysregulation, remains elusive in individuals with type 2 diabetes mellitus (T2DM) coexisting with non-alcoholic fatty liver disease (NAFLD). Investigating these metabolites could offer valuable insights into the pathophy-siology of NAFLD in T2DM. AIM: To identify potential metabolite biomarkers capable of distinguishing between NAFLD and T2DM. METHODS: A training model was developed involving 399 participants, comprising 113 healthy controls (HCs), 134 individuals with T2DM without NA

Influence of unsaturated fatty acids, membrane fluidity and oxygenation on the survival of an E. coli fatty acid auxotroph following gamma-irradiation.

Escherichia coli K1060, a fatty acid auxotroph unable to either synthesize or degrade unsaturated fatty acids (uFAs), was used to study the effect of membrane fluidity on survival after exposure to ionizing radiation. Using this strain of E. coli, significant alterations in the fatty acid composition of the membrane have been produced and verified by gas chromatography. Linolenic, oleic, elaidic and palmitelaidic acids were the uFAs used. Survival above the transition temperature (Tt) (liquid crystal in equilibrium gel) was comparable for these fatty-acid-supplemented membranes after exposure

Lipid Membrane Remodeling by the Micellar Aggregation of Long-Chain Unsaturated Fatty Acids for Sustainable Antimicrobial Strategies.

Antimicrobial fatty acids derived from natural sources and renewable feedstocks are promising surface-active substances with a wide range of applications. Their ability to target bacterial membrane in multiple mechanisms offers a promising antimicrobial approach for combating bacterial infections and preventing the development of drug-resistant strains, and it provides a sustainable strategy that aligns with growing environmental awareness compared to their synthetic counterparts. However, the interaction and destabilization of bacterial cell membranes by these amphiphilic compounds are not ye

Unsaturated Fatty Acids Affect Quorum Sensing Communication System and Inhibit Motility and Biofilm Formation of Acinetobacter baumannii.

The increasing threat of Acinetobacter baumannii as a nosocomial pathogen is mainly due to the occurrence of multidrug-resistant strains that are associated with the real problem of its eradication from hospital wards. The particular ability of this pathogen to form biofilms contributes to its persistence, increases antibiotic resistance, and promotes persistent/device-related infections. We previously demonstrated that virstatin, which is a small organic compound known to decrease virulence of Vibrio cholera via an inhibition of T4-pili expression, displayed very promising activity to prevent

Unsaturated long-chain fatty acids induce the respiratory burst of human neutrophils and monocytes in whole blood.

BACKGROUND: It is increasingly recognized that infectious complications in patients treated with total parenteral nutrition (TPN) may be caused by altered immune responses. Neutrophils and monocytes are the first line of defence against bacterial and fungal infection through superoxide anion production during the respiratory burst. To characterize the impact of three different types of lipid solutions that are applied as part of TPN formulations, we investigated the unstimulated respiratory burst activation of neutrophils and monocytes in whole blood. METHODS: Whole blood samples were incubat

The Unsaturated/Saturated Fatty Acid Ratio: A Metabolic Hub and Therapeutic Vulnerability in Glioblastoma.

Glioblastoma (GBM) exhibits profound metabolic reprogramming, among which the balance between unsaturated and saturated fatty acids (UFA/SFA) emerges as a critical determinant of tumor behavior and treatment response. Recent studies have shown that fatty acid metabolic reprogramming is a key mechanism driving GBM progression. Abnormalities in fatty acid uptake, synthesis, desaturation, and oxidation collectively reshape the lipid composition of tumor cells, particularly by altering the unsaturated/saturated fatty acid ratio. Monounsaturated fatty acids mainly promote tumor cell proliferation a

PhoQ is an unsaturated fatty acid receptor that fine-tunes Salmonella pathogenic traits.

The Salmonella enterica PhoP/PhoQ two-component signaling system coordinates the spatiotemporal expression of key virulence factors that confer pathogenic traits. Through biochemical and structural analyses, we found that the sensor histidine kinase PhoQ acted as a receptor for long-chain unsaturated fatty acids (LCUFAs), which induced a conformational change in the periplasmic domain of the PhoQ protein. This resulted in the repression of PhoQ autokinase activity, leading to inhibition of the expression of PhoP/PhoQ-dependent genes. Recognition of the LCUFA linoleic acid (LA) by PhoQ was not

Retention behaviour of unsaturated fatty acid methyl esters on porous graphitic carbon.

The eluotropic strength of binary mobile phases was calculated for three homologous series of cis, trans, and cis-cis unsaturated fatty acid methyl esters (FAMEs). Binary mobile phases with chloroform, dichloromethane, or tetrahydrofuran as strong solvent and methanol or acetonitrile as weak solvent were tested. The volume fraction of strong solvent in the binary phases was between 0.3 and 0.8. Curves of eluotropic strength versus volume fraction of strong solvents showed similar trends to previously published results for saturated homologues. Correlation coefficients of the plots of eluotropi
