Effect of cyanide concentrations on the secondary structures of protein in the crude homogenates of the fish gill tissue.

The effect of cyanide concentrations on the secondary conformation of protein in the fish gill homogenate was determined using an attenuated total reflectance (ATR)/Fourier transform infrared (FT-IR) microspectroscopy. Gills from male Tilapia zillii were isolated and homogenized in pH 8.0 Tris buffer solution and subjected to FT-IR study. The results indicate that the amide I and III bands of protein in fish gill homogenate deformed markedly with the increase of cyanide concentration. The fish gill homogenate shows a maximum peak at 1650 cm(-1) in amide I band, suggesting the predominant propo

Cloning and characteristics of fish glial fibrillary acidic protein: implications for optic nerve regeneration.

Mammalian central nervous system neurons do not regenerate after axonal injury, unlike their counterparts in fish and amphibians. After axonal injury, glial cells in mammals do not support regrowth of axons, while in fish they support the regeneration process. Controversy exists as to whether or not the intact fish optic nerve expresses glial fibrillary acidic protein, a well-known marker for mature astrocytes, and thus whether its astrocytes differ in this respect from those of the brain and spinal cord, as well as from optic nerve astrocytes of other species. In an attempt to resolve this qu

Growth and development of skeletal anomalies in diploid and triploid Atlantic salmon (Salmo salar) fed phosphorus-rich diets with fish meal and hydrolyzed fish protein.

Diploid and triploid Atlantic salmon, Salmo salar were fed high-protein, phosphorus-rich diets (56-60% protein; ca 18g phosphorus kg-1 diet) whilst being reared at low temperature from start-feeding until parr-smolt transformation. Performances of salmon fed diets based on fish meal (STD) or a mix of fishmeal and hydrolysed fish proteins (HFM) as the major protein sources were compared in terms of mortality, diet digestibility, growth and skeletal deformities. Separate groups of diploids and triploids were reared in triplicate tanks (initially 3000 fish per tank; tank biomass ca. 620 g) from 0

Lysine-assisted ultrasonic treatment of fish myofibrillar proteins conformational rearrangement: A synergistic strategy for enhanced volatile compounds adsorption and emulsion stabilization.

This study elucidated the synergistic mechanism of lysine-assisted ultrasonic treatment in mediating sea bass myofibrillar proteins (MPs) conformational rearrangement and optimizing their flavor retention and emulsification dual functionalities. The lysine-assisted ultrasonic treatment reduced α-helices content to 24.1 %, exposing hydrophobic groups and enhancing volatile compound adsorption. Molecular docking confirmed hexanal, octanal, and heptanal bound to MPs via hydrophobic interactions, van der Waals forces, and hydrogen bonds. Additionally, collaborative treatment decreased the particle

Expanding the pH-Shift Technique for Sequential Extraction of Intact Proteins and Sulfated Polysaccharide From Fish Heads: A Novel Biorefinery Approach.

The pH-shift technology was successfully expanded for the sequential recovery of proteins and sulfated polysaccharides (SPs) from rainbow trout heads. Adjusting the pH of the remaining process water after protein recovery at their isoelectric point to 8 enabled precipitation and recovery of SPs with the aid of ethanol at both alkaline and acid process versions and named SP-11.5 and SP-2.5, respectively. The mass yield of SPs recovered using the alkaline process version was 3.25%, nearly double that of SP (1.75%) from the acid version. SP-11.5 contained higher levels of carbohydrates (61.22%),

Influence of Fish Skin (Labeo rohita) and Chia Seed Oil (Salvia hispanica) Blend on the Protein-Rich Breakfast Cereal Product Made Through the Use of Whole and Modified Chia Flour and Fish Skin Protein Isolates.

The term "Ready-to-Eat Breakfast Cereal" refers especially to the processed grain formulations suitable for human consumption without requiring further preparation to save time. It is easy to store, relatively shelf-stable, and lightweight. A significant proportion of them are made with corn, wheat, oats, and rice, with added flavors and fortifications. In this work, an attempt was made to utilize the rich antioxidative blend of fish skin oil (FSO) and chia seed oil (CO). Four formulations were prepared using four different chia seed flour, namely, raw chia seed flour breakfast cereal (RCB), d

Nuclear localisation of a fish-derived ice-binding protein and related transcriptional changes in Caenorhabditis elegans.

Ice-binding proteins (IBPs) are functional molecules that enable organisms to survive severe cold by binding specifically to ice crystals and suppressing their growth and recrystallisation. Although their primary role has traditionally been viewed as antifreeze activity, recent studies have suggested additional cell-protective functions, including possible effects on membrane stability. Heterologous expression of the fish type III IBP NfeIBP in Caenorhabditis elegans has previously been shown to increase tolerance to cold and freezing stress. However, the understanding of the intracellular loc

Cross-Linked Reticular Magnetic Beads Immobilizing Streptavidin for Fishing the Protein of Interest with High Sensitivity.

Magnetic beads immobilized with affinity ligands act as a potent tool to fish the protein of interest (POI) interacting with bioactive compounds, proteins, or nucleic acids. The high loading capacity of affinity ligands is crucial to assay sensitivity. Herein, we report a simply fabricated cross-linked carboxylic magnetic bead, Fe3O4@poly(acrylic acid-co-divinylbenzene) (PAA-co-DVB), with a reticular network, exhibiting excellent magnetic response, high carboxylic functionality, and a fast reaction rate during amidation. Streptavidin (SA) is immobilized onto Fe3O4@PAA-co-DVB through the encaps

Targeted screening of pancreatic lipase inhibitory peptides from quinoa protein hydrolysates by ligand fishing.

A ligand fishing strategy was developed to screen for pancreatic lipase (PL) inhibitory peptides from quinoa protein hydrolysate (QPH) using PL-immobilized Fe₃O₄@SiO₂ magnetic particles. An initial activity of 230 U/g, a PL loading capacity of 180 mg/g, an immobilization yield of 25 %, and retained 39.23 % of its activity after 10 reuse cycles. Three novel peptides (LNEILAHE, VETELEKR, TVSAPSPK) were identified by LC-MS/MS and exhibited moderate PL inhibitory activities with IC₅₀ values ranging from 0.48 to 0.92 mg/mL. They showed dose-dependent anti-adipogenesis effects in 3T3-L1 cells, with

Expression of SPARC and the osteopontin-like protein during skeletal development in the cichlid fish Oreochromis mossambicus.

BACKGROUND: Bones are mainly composed of calcium hydroxyapatite and a proteinous matrix. In this study, we focus on the bone matrix proteins, the fish osteopontin orthologous protein (osteopontin-like protein; OP-L) and SPARC, because the current knowledge regarding their expression is fragmentary or contradictory. RESULTS: We first provide a comprehensive and detailed description of skeletal development in the cichlid fish Oreochromis mossambicus. Following this, we analyzed the expression pattern of OP-L and SPARC in detail during development. OP-L expression was only found in tissues that

Spatial Expression of Otolith Matrix Protein-1 and Otolin-1 in Normally and Kinetotically Swimming Fish.

Kinetosis (motion sickness) has been repeatedly shown to affect some fish of a given clutch following the transition from 1g to microgravity or from hypergravity to 1g. This susceptibility to kinetosis may be correlated with irregular inner ear otolith growth. Otoliths are mainly composed of calcium carbonate and matrix proteins, which play an important role in the process of otolith mineralization. Here, we examine the morphology of otoliths and the expression pattern of the major otolith proteins OMP-1 and otolin-1 in a series of hypergravity experiments. In the utricle, OMP-1 is present in

Expression and function of cGMP-dependent protein kinase type I during medaka fish embryogenesis.

We isolated and characterized cDNA clones (PKG Ialpha and PKG Ibeta) for medaka fish cGMP-dependent protein kinase (PKG) Ialpha and Ibeta, and demonstrated that both are expressed in the embryos after late gastrula stage. Whole-mount in situ hybridization using each isoform-specific probe revealed that the transcripts of the PKG Ialpha gene were present in the spinal cord and gill arch, whereas those of the PKG Ibeta gene were only weakly expressed in these organs, but highly expressed in the otic vesicles. Injection of PKG Ialpha-specific morpholino antisense oligonucleotides (Ialpha-MO) into

Allicin-mediated protection of myofibrillar proteins in refrigerated mandarin fish (Siniperca chuatsi): Molecular interactions with endogenous proteases and inhibition of protein degradation.

Endogenous proteases accelerate myofibrillar protein (MP) degradation and quality loss in fish. This study investigated allicin's protective effects on refrigerated mandarin fish (Siniperca chuatsi). Allicin treatment significantly inhibited cathepsin B, cathepsin L, and calpain activities and reduced MP degradation, leading to lower MFI values and slower accumulation of TCA-soluble peptides. Molecular docking and enzyme assays demonstrated that allicin binds to key catalytic residues of these endogenous proteases, suggesting a multi-target inhibitory mechanism. SDS-PAGE and histological stain

Assessing seasonal changes in the efficiency of protein digestion in gastric and agastric fish using in vitro models.

The objective of this study was to evaluate whether the efficiency of protein digestion in selected freshwater fish species exhibits functional adaptations to seasonal fluctuations in water temperature, mediated by variations in the proteolytic capacity to hydrolyze substrates. To test this hypothesis, individuals of two species-the Prussian carp (Carassius gibelio) and the European perch (Perca fluviatilis)-were collected during two different seasons (spring and summer) in the Chany Lake basin (Western Siberia, Russia). Digestive enzyme extracts were employed in in vitro hydrolysis assays usi

Protein expression patterns and metal metabolites in a protogynous hermaphrodite fish, the ricefield eel (Monopterus albus).

BACKGROUND: The ricefield eel Monopterus albus undergoes a natural sex change from female to male during its life cycle, and previous studies have shown the potential mechanisms of this transition at the transcriptional and protein levels. However, the changes in protein levels have not been fully explored, especially in the intersexual stage. RESULTS: In the present study, the protein expression patterns in the gonadal tissues from five different periods, the ovary (OV), early intersexual stage gonad (IE), middle intersexual stage gonad (IM), late intersexual stage gonad (IL), and testis (TE

Interindividual variability in response to protein and fish oil supplementation in older adults: a randomized controlled trial.

BACKGROUND: Precision nutrition is highly topical. However, no studies have explored the interindividual variability in response to nutrition interventions for sarcopenia. The purpose of this study was to determine the magnitude of interindividual variability in response to two nutrition supplementation interventions for sarcopenia and metabolic health, after accounting for sources of variability not attributable to supplementation. METHODS: A 24 week, randomized, double-blind, placebo-controlled trial tested the impact of leucine-enriched protein (LEU-PRO), LEU-PRO plus long-chain n-3 PUFA (

Oral immunization with recombinant protein antigen expressed in tobacco against fish nervous necrosis virus.

Nervous necrosis virus (NNV), also known as betanodavirus, has been recently implicated in mass mortalities of cultured marine fish. An effective vaccine is urgently needed to protect fish against this virus. However, parenteral immunization methods are very stressful. Individual immunization for thousands of fish is very labor intensive and expensive. Therefore, we expressed NNV coat protein in tobacco chloroplasts and used it as an oral vaccine to induce immunities in fish followed by challenges with NNV. Our results revealed that mice (IgG and IgA) and fish (IgM) immunized with the oral vac
