We report a 9-year-old boy who presented with mitral valve perforation secondary to Ustilago species fungal infective endocarditis. Subsequent evaluation revealed previously undiagnosed HIV infection. Fungal endocarditis due to Ustilago species is exceedingly rare, and presentation as the initial manifestation of HIV has not been previously described in the pediatric population. The atypical presentation posed significant diagnostic challenges, emphasizing the need to consider underlying immunodeficiency, including HIV infection, in children presenting with unusual or opportunistic infections.

Continuous cropping obstacle from Chinese yam (Dioscorea spp.) is widespread in China, and it seriously reduced the yield and quality. Rhizosphere soil microbiome is rich and associated with continuous cropping obstacle. However, the effect of long-term consecutive monoculture (LTCM) of Chinese yam on rhizosphere soil fungal community is still limited. In this study, fields that were consecutively cropped with Chinese yam for 1, 10 and 20 years were subjected to rhizosphere soil fungal analysis. High-throughput sequencing was used to characterize rhizosphere soil fungal community structure and

Fungal plasma membrane proteins represent key therapeutic targets for antifungal agents, yet their structure and spatial distribution in the native context remain poorly characterized. Herein, we employ an integrative multimodal approach to elucidate the structural and functional organization of plasma membrane protein complexes in Candida glabrata , focusing on prominent and essential membrane proteins, the polysaccharide synthase β-(1,3)-glucan synthase (GS) and the proton pump Pma1. Cryo-electron tomography (cryo-ET) and live cell imaging reveal that GS and Pma1 are heterogeneously distribu

The health and social consequences of untreated mental health disorders and heavy alcohol use are substantial, particularly in settings with high HIV-prevalence. Transdiagnostic approaches may offer a more efficient public health response, particularly where behavioral health services are limited. The present study explored the implementation of Common Elements Treatment Approach (CETA), a transdiagnostic intervention, in Western Uganda, from the perspectives of multilevel stakeholders. One-on-one in-depth interviews with multilevel stakeholders involved in the rollout of CETA in Uganda were c

Amyotrophic lateral sclerosis (ALS) is a fatal human motor neuron disease leading to muscle atrophy and paralysis. Mutations in superoxide dismutase 1 (SOD1) are associated with familial ALS (fALS). The SOD1 mutants in ALS have a toxic-gain of function by destabilizing the functional SOD1 homodimer, consequently inducing fibril-like aggregation with a cytotoxic non-native trimer intermediate. Therefore, reducing SOD1 oligomerization via chemical modulators is an optimal therapy in ALS. Here, we report the discovery of Phialomustin-B, an unsaturated secondary metabolite from the endophytic fung

The galactomannan (GM) that is produced by the human fungal pathogen Aspergillus fumigatus is an emblematic biomarker in medical mycology. The GM is composed of two monosaccharides: mannose and galactofuranose. The furanic configuration of galactose residues, absent in mammals, is responsible for the antigenicity of the GM and has favoured the development of ELISA tests to diagnose aspergillosis in immunocompromised patients. The GM that is produced by A. fumigatus is a unique fungal polysaccharide containing a tetramannoside repeat unit and having three different forms: (i) membrane bound thr

We have previously rationally designed, synthesized and tested a number of 3-deazapurine analogues, which inhibit the ubiquitous fungal nucleobase transporter FcyB, through binding in its major substrate binding site, by specifically interacting with Asn163. Here, in an effort to further understand the molecular details of structure-activity relationships in all three major nucleobase transporters of fungi, we extend this study by designing, based on our previous experience, synthesizing and testing further 3-deazapurine analogues. We thus identify seven new compounds with relatively high affi

Ovarian cancer is one of the most lethal gynecological malignancies with poor prognosis due to high drug resistance and low tumor susceptibility. This study investigated the cytotoxic effects of four sterols of fungal origin, ergosterol (ES), cholesterol (CS), β-sitosterol (βS), and lanosterol (LS) on human ovarian cancer SKOV3 cells, revealing that they exert their effects by correlating with autophagy and ferroptosis‑related signaling, and analyzed the structure-activity relationship. SKOV3 tumor cells were treated with four types of sterols for 24 h. Cell viability, cell cycle, apoptosis, m

Materials in low Earth orbit (LEO) face radiation, atomic oxygen erosion, and extreme temperature fluctuations, which can severely compromise their structural and functional integrity. Developing lightweight, multifunctional materials capable of withstanding these harsh conditions is critical for long-term space exploration and sustainable extraterrestrial settlements. This study evaluates the structural stability and radiation shielding efficacy of polylactic acid (PLA) and biocomposites, including PLA infused with fungal melanin, synthetic melanin, or animal melanin, and a compressed myceliu

The rising prevalence of life-threatening fungal infections, particularly in immunocompromised individuals, necessitates a deeper understanding of all facets of the host immune response. While much focus has been placed on cellular immunity, the contribution of Immunoglobulin M (IgM), the first antibody produced during an immune response, remains a relatively underexplored area in the context of systemic mycoses. This comprehensive survey explores the role of IgM in antifungal immunity, with a focus on life-threatening fungal infections. As the earliest antibody isotype, IgM achieves remarkabl

Fungal cell walls and their biosynthetic enzymes are potential targets for novel antifungal agents. Recently, two mannosyltransferases, namely core-mannan synthases A (CmsA/Ktr4) and B (CmsB/Ktr7), were found to play roles in the core-mannan biosynthesis of fungal-type galactomannan. CmsA/Ktr4 is an α-(1→2)-mannosyltransferase responsible for α-(1→2)-mannan biosynthesis in fungal-type galactomannan, which covers the cell surface of Aspergillus fumigatus Strains with disrupted cmsA/ktr4 have been shown to exhibit strongly suppressed hyphal elongation and conidiation alongside reduced virulence

UNLABELLED: The fungal pathogen Aspergillus fumigatus has been implicated in a drastic increase in life-threatening infections over the past decade. However, compared to other microbial pathogens, little is known about the essential molecular processes of this organism. One such fundamental process is DNA replication. The protein responsible for ensuring processive DNA replication is PCNA (proliferating cell nuclear antigen, also known as the sliding clamp), which clamps the replicative polymerase to DNA. Here we present the first crystal structure of a sliding clamp from a pathogenic fungus (

Bacterial and fungal communities associated with plant roots are central to the host health, survival and growth. However, a robust understanding of the root-microbiome and the factors that drive host associated microbial community structure have remained elusive, especially in mature perennial plants from natural settings. Here, we investigated relationships of bacterial and fungal communities in the rhizosphere and root endosphere of the riparian tree species Populus deltoides, and the influence of soil parameters, environmental properties (host phenotype and aboveground environmental settin

White grub larvae are important soil-dwelling pests in many regions of Mexico as they attack many important crops such as maize. The use of synthetic chemicals is currently the main control strategy, but they are not always effective; thus, other alternatives are needed. Microbial control using entomopathogenic fungi represents an important alternative strategy, and species within the genera Beauveria and Metarhizium are considered amongst the most promising candidates. Seventeen Beauveria spp. and two Metarhizium spp. isolates were obtained in surveys of white grub larvae from different regio

BACKGROUND: To report a case series of microbial keratitis showing atypical yeast-like structures in direct microscopy which were culture-negative but subsequently identified as yeast cells by PCR and DNA sequencing.
MAIN TEXT: This is a retrospective, non-comparative case series of eight patients with infectious keratitis, where smear examination (potassium hydroxide + calcofluor white) showed spore like structures resembling yeast. There was no growth in any solid culture media. Routine PCR assay was performed using pan fungal primers followed by Sanger sequencing and nucleotide sequences w

INTRODUCTION: Bacterial wilt caused by Ralstonia solanacearum is a major constraint to tomato production worldwide. Rhizosphere microbial community is closely linked to plant health. However, how R. solanacearum invasion alters the assembly processes and co-occurrence network in tomato rhizosphere fungal communities remains largely unclear.
METHODS: Here, we collected rhizosphere soil samples from three major tomato-producing regions in China to investigate how R. solanacearum invasion affects fungal community composition, network topology, and assembly processes in tomato rhizosphere.

Arbuscular mycorrhizal fungi (AMF) are important root symbionts that enhance nutrient acquisition, water uptake, stress tolerance, and soil ecosystem functioning in terrestrial plants. In perennial plantation systems like Hevea brasiliensis, understanding the dynamics of root-associated AMF communities under varying seasonal and nutrient conditions is essential for developing sustainable nutrient management strategies, particularly in emerging rubber growing regions. In the present study, we investigated the, composition, diversity, and seasonal dynamics of AMF communities directly associated

Honey bee gut microbiome studies have primarily emphasized bacteria, leaving fungal communities comparatively overlooked despite their ecological and functional importance. Whole-genome shotgun metagenomics of Apis cerana and Apis mellifera revealed fungal assemblages dominated by Ascomycota, with Basidiomycota and Microsporidia in minor proportions, alongside gut bacterial communities composed mainly of Pseudomonadota, Bacillota, and Actinomycetota. The bacterial diversity was markedly higher in A. mellifera (Shannon = 5.90; Simpson = 0.98) than in A. cerana (Shannon = 4.01; Simpson = 0.94; p

Plant and fungal cell walls are essential for growth, adaptation, and survival, with their intricate architectures dictating both resistance to stress and susceptibility to antifungal or biomass-degrading strategies. Understanding how these walls are built, remodeled, and function at the molecular level is therefore central to both clinical and biotechnological applications. Solid-state nuclear magnetic resonance (ssNMR) has emerged as a uniquely powerful tool for this purpose, as it reveals the structure, dynamics, and interactions of intact biopolymers without disrupting their native organiz
