Genetic differentiation of invasive Aedes albopictus by RAPD-PCR: implications for effective vector control.

Aedes albopictus is an invasive mosquito species of great concern to public health as it is responsible for the biological transmission of several pathogens causing dengue, chikungunya, yellow fever, etc. In 2009, this mosquito was detected for the first time in Agra City. This study represents the first genetic analysis of A. albopictus from India. Random amplified polymorphic DNA (RAPD) was used to study the genetic structure of A. albopictus in four populations from different larval habitats. Seven RAPD primers produced 141 loci. The results displayed rich genetic variation among larval pop

Genetic structure and phylogeography of Aedes aegypti, the dengue and yellow-fever mosquito vector in Bolivia.

Between the 16th and 18th centuries, Aedes aegypti (Diptera: Culicidae), a mosquito native to Africa, invaded the Americas, where it was successively responsible for the emergence of yellow fever (YF) and dengue (DEN). The species was eradicated from numerous American countries in the mid-20th century, but re-invaded them in the 1970s and 1980s. Little is known about the precise identities of Ae. aegypti populations which successively thrived in South America, or their relation with the epidemiological changes in patterns of YF and DEN. We examined these questions in Bolivia, where Ae. aegypti

A longitudinal molecular surveillance of genetic heterogeneity of Orientia tsutsugamushi in humans, reservoir animals, and vectors in Puducherry, India.

BACKGROUND: Scrub typhus (ST), is a vector borne zoonotic disease, transmitted by the larva of the trombiculid mites. The enzootic cycle of the pathogen involves rodents/shrews as the animal reservoirs and humans are the accidental dead-end host. A transposon-like activity in its major antigen 56 kDa, has led to the evolution of several serotypes/strains, and more than 40 serotypes are reported globally. Puducherry, India, is endemic to scrub, but limited data exist on local serotype distribution across hosts and vectors. METHODOLOGY: A longitudinal molecular surveillance was conducted in Pud

Immune shielding of viral vectors via capsid engineering with genetically encoded zwitterionic peptides.

Pre-existing neutralizing antibodies (NAbs) and blood proteins can rapidly inactivate therapeutic viral vectors and trigger immune toxicities in patients. To overcome these challenges, we engineered the surfaces of adenovirus (Ad) by incorporating genetically encoded "protective shields" to reduce the recognition by host factors. AlphaFold3 modeling showed that inserting a structured biotin acceptor peptide (BAP) into hypervariable region 5 (HVR5) of the viral capsid protein hexon produced a rigid surface protrusion, whereas inserting a disordered, zwitterionic glutamic acid-lysine (EK) peptid

Pyrethroid resistance in Aedes aegypti: genetic mechanisms worldwide, and recommendations for effective vector control.

BACKGROUND: The Aedes aegypti mosquito, a primary vector of arboviruses such as dengue, Zika, yellow fever, and chikungunya, poses a significant public health threat worldwide. Its adaptability and genetic diversity complicate control efforts, enabling rapid resistance evolution. Pyrethroid insecticides, a cornerstone of vector control, target voltage-gated sodium channels, yet resistance driven by knockdown resistance (kdr) mutations and detoxification mechanisms have undermined their efficacy. METHODS: This review summarizes findings from a targeted literature search, exploring the genetic

Comparative analysis of diagnostic methods and genetic analysis for canine vector-borne diseases in Thailand.

Canine vector-borne diseases (CVBDs), transmitted by arthropods, are widespread globally and pose significant health risks to dogs. Diagnosing CVBDs, particularly co-infections, is challenging for laboratory technicians. This study in Thailand evaluated 270 canine blood samples for vector-borne pathogens using microscopy, conventional PCR (cPCR), and the Vcheck M Canine Vector 8 Panel (Vcheck). Microscopy was used for initial screening, while cPCR and Vcheck tested for exposure to eight CVBDs-causing genera (Ehrlichia, Hepatozoon, Mycoplasma, Anaplasma, Rickettsia, Babesia, Leishmania, and Bar

Analysis of Asperger Syndrome Using Genetic-Evolutionary Random Support Vector Machine Cluster.

Asperger syndrome (AS) is subtype of autism spectrum disorder (ASD). Diagnosis and pathological analysis of AS through resting-state fMRI data is one of the hot topics in brain science. We employed a new model called the genetic-evolutionary random Support Vector Machine cluster (GE-RSVMC) to classify AS and normal people, and search for lesions. The model innovatively integrates the methods of the cluster and genetic evolution to improve the performance of the model. We randomly selected samples and sample features to construct GE-RSVMC, and then used the cluster to classify and extract lesio

[Construction of an adenoassociated, viral derived, expression vector to correct the genetic defect in Morquio A disease].

INTRODUCTION: Mucopolysaccharidosis IVA (Morquio A) is caused by a deficiency of N-acetylgalactosamine-6-sulphate-sulphatase, a lysosomal enzyme required for the stepwise degradation of keratan-sulfate and chondroitin-6-sulfate. A deficiency in this enzyme results in an accumulation of glycosaminoglycans in several tissues. Currently, no effective therapies exist and only supportive measures are used to treat some manifestations of the disease. An ideal therapy is one that can be administrated early in life, has low mortality, and leads to long-term expression of the enzyme. Gene therapy emerg

Genetic differentiation in the African malaria vector, Anopheles gambiae s.s., and the problem of taxonomic status.

Of the seven recognized species of the Anopheles gambiae complex, A. gambiae s.s. is the most widespread and most important vector of malaria. It is becoming clear that, in parts of West Africa, this nominal species is not a single panmictic unit. We found that the internal transcribed spacer (ITS) of the X-linked rDNA has two distinct sequences with three fixed nucleotide differences; we detected no heterozygotes at these three sites, even in areas of sympatry of the two ITS types. The intergenic spacer (IGS) of this region also displays two distinct sequences that are in almost complete link

The genetic architecture of target-site resistance to pyrethroid insecticides in the African malaria vectors Anopheles gambiae and Anopheles coluzzii.

Resistance to pyrethroid insecticides is a major concern for malaria vector control. Pyrethroids target the voltage-gated sodium channel (VGSC), an essential component of the mosquito nervous system. Substitutions in the amino acid sequence can induce a resistance phenotype. We use whole-genome sequence data from phase 2 of the Anopheles gambiae 1000 Genomes Project (Ag1000G) to provide a comprehensive account of genetic variation in the Vgsc gene across 13 African countries. In addition to known resistance alleles, we describe 20 other non-synonymous nucleotide substitutions at appreciable po

Genetic Diversity, Haplotype Relationships, and kdr Mutation of Malaria Anopheles Vectors in the Most Plasmodium knowlesi-Endemic Area of Thailand.

Plasmodium knowlesi, a malaria parasite that occurs naturally in long-tailed macaques, pig-tailed macaques, and banded leaf monkeys, is currently regarded as the fifth of the human malaria parasites. We aimed to investigate genetic diversity based on the cytochrome c oxidase subunit I (COI) gene, detect Plasmodium parasites, and screen for the voltage-gated sodium channel (VGSC)-mutation-mediated knockdown resistance (kdr) of Anopheles mosquitoes in Ranong province, which is the most P. knowlesi-endemic area in Thailand. One hundred and fourteen Anopheles females belonging to eight species, in

Investigating genetic differentiation between brackish and fresh water collections of the arboviral vector Aedes aegypti.

BACKGROUND: Aedes aegypti is typically regarded as a freshwater mosquito; however, recent studies have documented its development in brackish water habitats in coastal regions, including Sri Lanka's Jaffna Peninsula. Compared with freshwater populations, brackish water samples in Jaffna display enhanced salt tolerance throughout larval-to-adult development, along with differences in gene expression, cuticle morphology and composition, and insecticide susceptibility. METHODS: To explore the genetic basis of these differences, we performed a comparative genomic analysis using 5135 genome-wide s

Establishment of a reverse genetics system for feline panleukopenia virus and feasibility study of a live vector vaccine.

A reverse genetics system for the feline panleukopenia virus (FPV) strain JL2280 was successfully established in this study. By optimizing the cloning strategy for the terminal hairpin structures, a full-length infectious clone, pFPV, harboring a KpnI genetic marker was constructed. The recombinant virus (rFPV) was successfully rescued in CRFK cells and exhibited replication kinetics, pathogenicity, and morphological characteristics comparable to those of the wild-type virus. To evaluate the potential of FPV as a live vector, a recombinant virus expressing enhanced green fluorescent protein (EG

Population genetics of dengue vector Aedes aegypti (Diptera: Culicidae) from northeastern region Brazil.

The Aedes aegypti (Linnaeus, 1762) is the principal vector of dengue, chikungunya, Zika, and yellow fever viruses, worldwide. The rate of infected people notifications, have risen in recent years in of Brazil, especially dengue, reinforces the need for the genetic monitoring of these populations, to provide insights into the population dynamics of this vector. So, a population study has been conducted using sequences of 2 mitochondrial genes in Ae. aegypti 22 populations from northeastern Brazil. The analyses identified 33 and 28 haplotypes of ND4 and COI, respectively, with high levels of gen

Synthesizing environmental, epidemiological and vector and parasite genetic data to assist decision making for disease elimination.

We present a framework for identifying when conditions are favourable for transmission of vector-borne diseases between communities by incorporating predicted disease prevalence mapping with landscape analysis of sociological, environmental and host/parasite genetic data. We explored the relationship between environmental features and gene flow of a filarial parasite of humans, Onchocerca volvulus, and its vector, blackflies in the genus Simulium. We generated a baseline microfilarial prevalence map from point estimates from 47 locations in the ecological transition separating the savannah and

Genetic diversity and Wolbachia infection in the Japanese encephalitis virus vector Culex tritaeniorhynchus in the Republic of Korea.

BACKGROUND: Culex tritaeniorhynchus, a major vector of Japanese encephalitis virus (JEV), is found across a broad geographical range, including Africa, Asia, Australia and Europe. Understanding the population structure and genetic diversity of pathogen vectors is increasingly seen as important for effective disease control. In China and Japan, two countries in close proximity to the Republic of Korea (ROK), Cx. tritaeniorhynchus has been categorized into two clades based on the DNA barcoding region of mitochondrial cytochrome c oxidase subunit I (COI), suggesting the presence of cryptic specie

The influence of roads on the fine-scale population genetic structure of the dengue vector Aedes aegypti (Linnaeus).

Dengue is endemic in tropical and subtropical countries and is transmitted mainly by Aedes aegypti. Mosquito movement can be affected by human-made structures such as roads that can act as a barrier. Roads can influence the population genetic structure of Ae. aegypti. We investigated the genetic structure and gene flow of Ae. aegypti as influenced by a primary road, España Boulevard (EB) with 2000-meter-long stretch and 24-meters-wide in a very fine spatial scale. We hypothesized that Ae. aegypti populations separated by EB will be different due to the limited gene flow as caused by the barrie

Reverse genetics approaches of Borna disease virus: applications in development of viral vectors and preventive vaccines.

The plasmid-based reverse genetics system, which involves generation of recombinant viruses from cloned cDNA, has accelerated the understanding of clinical and virological aspects of different viruses. Borna disease virus (BoDV) is a nonsegmented, negative-strand RNA virus that causes persistent intranuclear infection in various vertebrate species. Since its first report, reverse genetics approaches with modified strategies have greatly improved rescue efficiency of recombinant BoDV and enhanced the understanding of function of each viral protein and mechanism of intranuclear persistency.
