We recently described a murine model for mucopolysaccharidosis VII in mice that have an inherited deficiency of beta-glucuronidase (beta-D-glucuronoside glucuronosohydrolase, EC 3.2.1.31). Affected mice, of genotype gusmps/gusmps, present clinical manifestations similar to those of humans with mucopolysaccharidosis VII (Sly syndrome) and are shown here to have secondary elevations of other lysosomal enzymes. The authors report that osmic acid, in vitro, increases the activity of beta-glucuronidase in serum and synovial, ascitic and pleural fluids, whereas five other metals of similar molecular weight (platinum, gold, mercury, thallium and lead) are either inactive or inhibitory. By contrast the osmium diminishes the activity of LDH and acid phosphatases in serum and synovial fluid. This effect on beta-glucuronidase is thought to be due to inhibition of a natural inhibitor present in biological fluids, a protein found by several authors.

The value of the cytochemical beta-glucuronidase (beta-gluc.) reaction in the differential diagnosis of acute leukaemia was assessed in a series of 100 adult patients. A purely granular type of reaction was observed in 7 out of 8 cases of lymphoblastic leukaemia and in 2 of 11 cases of acute leukaemia of uncertain type. Breast milk or formula milk and serum samples from 34 breast-fed babies, 15 formula-fed babies, and their mothers were examined at 3 and 21 days of neonatal age. Infant faecal samples were obtained on day 21 only. At both ages serum total bilirubin concentrations were significantly higher in breast-fed than in formula-fed infants. The intestinal beta-glucoronidase was studied in germfree, monocontaminated and conventional rats. The greater part of the beta-glucuronidase of the caecum and the large intestine of the contaminated animals was of bacterial origin. No bacterial beta-glucuronidase was found in the small intestine. Monocontamination with Escherichia coli gave activities corresponding to those of the conventional rats, whereas content from the caecum and the large intestine of the rats monocontaminated with Streptococcus pyogenes showed an activity approximately 10 per cent of that of the conventional rats.

Breast milk and formula milk and the corresponding serum samples from 20 breast-fed babies, 20 formula-fed babies, and their mothers were examined at 3 days of age for beta-glucuronidase enzyme. Serum indirect bilirubin levels were also examined for all the infants. OBJECTIVES: Polymorphonuclear neutrophil (PMN) dysfunction is associated with diabetes. We examined the gingival crevicular fluid (GCF) beta-glucuronidase (BG) and interleukin-8 (IL-8) levels of periodontitis patients with and without type 2 diabetes mellitus (DM).
MATERIAL AND METHODS: Forty five adults with type 2 DM and 32 adults without DM, both with chronic periodontitis were enrolled. GCF was collected from eight posterior sites in each quadrant, and periodontal parameters were recorded. GCF was assayed for IL-8 by ELISA and BG by a fluorometric assay.

A phenotypically normal mother of a mucopolysaccharidosis VII child, is reported with an unusually low beta-glucuronidase activity.
